In still other species, T2RNAses have beendescribed to provide hostprotectionagainst pathogens or to trigger cellular senescence (7,8). Al- though involved in apparently different biological pro- cesses, the mostbasic and relevant featuresofRNAseT2 proteinsaretheir localization in the extracellularmilieu (where RNAs are conventionally thought to be unavail- able)and their ability to controlcell growth. Therefore, afunctional link between these two features has been envisioned, suggesting that RNAses belonging to this family, besides intracellular RNA digestion, are en- dorsed with novel biological functions that areper- formed outside the cell. Among these biological functions, the well-described control of cellular growth mediatedby T2 RNAses is particularlyattractingfrom theoncological point of view. Indeed, RNASET2 was found byour group to be hypoexpressed or absent at thetranscript level in several primary ovarian tumors and ovarian cell lines (9).More- over, we reported aclear inhibition of tumor growth fol- lowing inoculation in nude mice of RNASET2-overex- pressing cellsfrom the Hey2Met3 ovarian cancer cell line (10). Despitesuch a strong effect in vivo, the RNASET2-transfected cells did notshow any change whencancer-relatedparameters were assayed in vitro.
