The ProteOn XPR36 protein interaction array system is an SPR optical biosensor that provides real-time data on the affinity, specificity, and interaction kinetics of protein interactions. Using proprietary XPR™ technology, a unique approach to multiplexing, this system generates a 6 x 6 interaction array for the simultaneous analysis of up to six ligands with up to six analytes. The ProteOn XPR36 instrument incorporates the microfluidics, optical detection, and temperature control systems that are required for interaction analysis. It integrates a high-efficiency crisscross microfluidics system with a highly sensitive optical system to generate interaction array data on up to six ligands with panels of up to six analytes. Flexible Sample ConfigurationThe autosampler accommodates the ProteOn sample rack and ProteOn microplates. The sample rack holds 72 ProteOn sample vials with pierceable caps. The microplate layout accommodates two 96-well ProteOn standard microplates or ProteOn deep-well microplates. The illuminated autosampler platform is also temperature controlled.

Instrument Status LEDsA series of LEDs indicate instrument communication with ProteOn Manager™ software, temperature of the autosampler and sensor chip chambers, experiment status, and autosampler status.

Bar Code RecognitionBar codes provide automatic recognition of sensor chip type, expiration date, and lot number. Bar codes also provide a usage record and direct linkage between the chip identification and its associated protocol and experiment.

Advanced Optics and Interspot Referencing The ProteOn XPR36 protein interaction array system uses imaging technology to detect the SPR response for each spot on a sensor chip. The ProteOn XPR36 system measures a total of 78 spots: 36 spots represent the interaction data from the 6 x 6 array, and 42 interspot references can be used for reference subtraction.

Innovative Fluidics and Parallel ProcessingThe 6 x 6 interaction array and microfluidics system provide efficient parallel processing of multiple samples. Two sets of six 0.5 ml syringe pumps (sample syringes and buffer syringes) operate in unison for parallel aspiration and injection of six samples or reagents.

The buffer compartment holds two 2 L bottles for running buffer, and valve control between the two buffer bottles is hardware and software controlled. An in-line degasser removes bubbles from the buffer as it moves through the fluidics system.

They have been thoroughly tested for a wide range of protein-protein, protein-peptide, protein–small molecule, and protein-DNA interactions. Each sensor chip includes a unique bar code for identification and a usage record. ProteOn Multiple Protein Interaction KitThe interaction between five mutant TEM1 b-lactamase proteins and wild-type b-lactamase inhibitor protein (BLIP) demonstrates the power of the ProteOn XPR36 system to produce a detailed kinetic analysis of multiple simultaneous interactions and to map protein interfaces.

The ProteOn multiple protein interaction kit includes a ProteOn TEM1/BLIP protein set, with sufficient materials to immobilize five TEM1 mutant proteins in five channels and to perform a detailed kinetic analysis of the interaction of each of the five mutant proteins with BLIP.

ProteOn Protein–Small Molecule Kit The interaction between the carbonic anhydrase II protein and the small molecule 4-carboxy benzene sulfonamide (4-CBS) demonstrates the capability of the ProteOn XPR36 system to detect low molecular weight analytes. The kit also provides a benchmark for optimal system performance.

ProteOn Running Buffers ProteOn running buffers (PBS, PBS/Tween, and PBS/Tween/EDTA) are prefiltered and supplied in 2 L bottles sufficient for approximately 2 weeks of daily operation.

ProteOn Chip Normalization Solution ProteOn chip normalization solution is a 50% glycerol solution used for normalization of ProteOn sensor chip surfaces. It is provided in a 100 ml bottle sufficient for 6 months of daily use.

ProteOn Maintenance ChipsA maintenance (MNT) chip and a cleaning (CLN) chip are available individually or as part of the ProteOn maintenance kit.

ProteOn Amine Coupling KitThe ProteOn amine coupling kit provides the reagents required for coupling ligands that have amine groups (for example, proteins and nucleic acids) to surface carboxyl groups of ProteOn GLC, GLM, and GLH sensor chips.

The innovative surface chemistry of ProteOn sensor chips, combined with the ProteOn amine coupling reagents, minimizes electrostatic and nonspecific binding effects and optimizes ligand activities and coupling capacities. An ethanolamine hydrochloride solution is included (also available separately) for deactivation of the residual active sites on the sensor chip following activation.

Sufficient reagents are provided for 60 moderate activations or up to 600 light activations.

ProteOn Immobilization Buffer Kit ProteOn immobilization buffers (four acetate buffers ranging from pH 4.0 to 5.5) are designed to dilute protein samples for coupling to ProteOn sensor chips. The entire range of buffers may be used to determine the optimal pH for immobilization. Each buffer is sufficient for 100 ligand immobilizations. These buffers are also sold individually.

ProteOn Regeneration Kit and BuffersProteOn regeneration buffers are ideal for a wide variety of regeneration conditions and are available as a complete kit of nine solutions or individually. They include glycine buffers, SDS, sodium hydroxide, hydrochloric acid, phosphoric acid, and sodium chloride.

ProteOn Maintenance KitThe ProteOn maintenance kit contains solutions and chips for post-experiment and weekly maintenance and cleaning of the ProteOn XPR36 protein interaction array system. Each kit includes enough supplies for 6 months of routine maintenance when following the ProteOn Manager™ software maintenance and cleaning protocol wizards.
