The Thermo Scientific™ Dionex™ UltiMate™ DAD 3000 detector is a high-resolution, 1024-element diode array detector (DAD) available in Rapid Separation (200 Hz) and Standard (100 Hz) versions. It operates with the Thermo Scientific™ Dionex™ Chromeleon™ Chromatography Data System software to provide a variety of spectra views, including 3-D plotting and automated chromatogram handling. The high resolution and low-noise performance of the DAD-3000 family makes it ideal for the most sensitive and accurate library searches and peak purity analyses.

3Design OverviewTheDAD-3000opticalbenchdesignincludestwolightsources–deuteriumandtungstenlamp–togetherwith1024diodesandpowerfulelectronics(Figure1).Polychromaticlightpassesthroughtheflowcell.Thedispersedwavelengthsaresimultaneouslydetectedbythediodearray.Thisensuresthesamesensitivityforsingle-andmulti-wavelengthoperation,providingconvenient,optimized,andsensitivequantificationofmaincompoundsandrelatedimpurities.

Reliable QuantificationNoiselevelswhenusingthewideslitare±8 µAUwithatypicalbaselinedriftof<0.5 mAU/hat254nm(flowingmobilephase).Thelownoiseandbaselinedriftallowfordetectionofweaksignals(goodsignal/noiseratio)andconsistent,reproducibleintegration.Atypicaldetectorlinearityupto2.0AUshowscorrelationcoefficientsof>0.999andaresidualstandarddeviationbelow5%withcaffeine.Thewidelinearityrangeallowsreliablequantificationofprimaryandsideproductsinthesamerun.

Sensitivity and Spectral ResolutionWhenacquiringspectraldataforpeakidentificationand/orpeakpurityanalysis,highresolutionspectraareessential.Atthesametime,itisimportanttoobtaingoodsignal-to-noiseratiosfordetectionofweaksignals.TheDAD-3000’sfamilyelectronicsaredesignedtoprovidehighspectralresolutionincombinationwithlownoise.

Figure2showsanexampleofanexplosiveresidueanalysis.Onesamplewasautomaticallyinjectedontwocolumnsofvaryingselectivity.Targetanalyteswereconfirmedbyspectralmatch,evenatverylowconcentrations.Chromeleon’scustomizablereportingfunctionsprovideinstantaneousaccesstoresults,listconfirmedorunconfirmedtargetanalytes,quantifythesubstances,anddefinetheirpeakpurity.

UV Spectra AcquisitionTheDAD-3000RSprovidesuptoeightsingledatachannelsandfullspectralscansat200Hzforeventhemostchallengingapplications.Figure4showstheseparationof10analytes,uracilandninealkylphenones,in10seconds.Thefullspectralscancaneasilykeepupwiththisspeed.PeakidentificationandpeakpurityanalysisareasconvenientandpowerfulasforconventionalLCseparations.Combiningthesefeatureswiththelownoisespecification,theDAD-3000RSisidealforultrafastanalysesandreadytomeetfuturechallenges.

Optimize for Best Sensitivity or Spectral ResolutionBothrapidseparationversions,theDAD-3000RSandtheMWD-3000RS,provideavariableslitattheentranceofthespectrograph.Thisisaneffectivetooltofurtheradaptthedetectortotheapplicationrequirements.Anarrowslitprovidesthehighestspectralresolution,resolvingevenveryfineabsorbancespectra,suchasforpyrene(Figure5).However,UVabsorbancebandsaretypically30nmandwider.Inthesecases,abroadslitresultsinthehighestdetectionsensitivity.Morelightpassesthroughthespectrograph,reducingbothshort-termnoiseandthelimitofdetection.

5Rapid Multiple Channel DetectionTheRapidSeparationversionoftheMultipleWavelengthDetectoroffershighspeedandsensitivityforbothsingleandmultiplewavelengthdataacquisition.Itsdiodearraytechnologyachievesmulti-channeldetectionofuptoeightchannelswithoutmovingparts.

Regardlesshowmanywavelengthsarerecorded,thedetectorcanmeasureallwavelengthsatthesamehighsensitivityandatamaximumdatacollectionrateof200Hz.Up-to-datevariablewavelengthdetectorsachievemultiplewavelengthdetectionbyrapidlyswitchingbetweenalternatewavelengths,butthisadverselyaffectsshort-termnoiseandreducesthemaximumdatacollectionratesignificantly.TheMWDdoesnothavethislimitation.

Upgrade MWD to Full DAD FunctionalityMWDdetectorsarebasedonphotodiodearraytechnology.TheonlydifferencefromDADdetectorsisthattheMWDdoesnotsupporttheacquisitionof3-Dspectra.

These3-Dspectracapabilitiesarefield-upgradable,turningyourMWDintoafull-blownDAD.Youmaythereforestaggeryourinvestmentindiodearraytechnology,purchasetheinstrumentyouneedtoday,andadaptittoyourfutureapplicationneedslater.

Ergonomic Design and Advanced System MonitoringAllfluidicpartsoftheMWDandDADdetectorseriescanbeaccesseddirectlyfromthefront.Thedoorfoldsdown,servingasaminiaturelabbenchtoplacetoolsandpartsduringmaintenance(Figure7).

Thedeuteriumandtungstenlampsofthedetectorcanalsobeaccessedfromthefrontbyopeningthefingertightnutsofthelampcover.Changingtheselampsisequallyfastandeasy.ModificationsofflowcellsorlampsareautomaticallytrackedbyintegratedIDchips.AllrelevantinformationisstoredinthelampandmoveswiththedetectorifitistransferredtoanotherLCsystem.Chromeleonconstantlymonitorstheseandvariousothercounterstoprovideyouwithearlywarningswhenapartisdueforreplacement.Inaddition,ChromeleonsoftwaremonitorsandverifiestheinstrumentqualificationstatustomeetyourGLPcompliancegoals(Figure8).
