96-Well Microplate Assay Kits PrinciplesMany of our most popular assay kits utilize custom-engineered enzymes as a detection system. Enzymatic systems offer superior sensitivity, specificity and accuracy when compared to other detection methods. Our assays do not involve laborious, time consuming sample preparation protocols. In many cases, direct analyte measurement can be performed with no required sample preparation steps. Our products are designed to have long shelf-life stability and do not require expensive instruments, trained personnel, extensive washes or long incubation times. A temper-ature-controlled microplate reader, capable of measuring absorbance and fluorescence over time is the only instru-ment required to run and perform our user-friend assay kits.

Sample application is perhaps the most important factor when selecting the best product for your research needs. This will determine the necessary assay sensitivity and therefore the preferred detection method. A) Cell Migration/Invasion plates: The cells are added to the Top Chamber and the Control Migration/Invasion Inducer or chemoattractant are added to the Bottom Chamber. B) Cell Migration (3 µm-pore): Erythroleukemia cells were starved over-night and treated with Control Migration Inducer for 48 h or left untreated (control cells). Treatment with Control Migration Inducer demonstrated a significant increase in migration as compared to untreated cells. C) Cell Invasion (8 µm-pore, Collagen I coating) using 75837-076: NIH-3T3 and HT-1080 cells were starved overnight and treated with Control (Cnt) Invasion Inducer or remain untreated (No Treatment). Diabetes has been recognized as a disorder of glucose intolerance for many decades. Insulin stimulates the glucose uptake into muscle & adipose tissues to maintain the whole-body glucose homeostasis. Among many different methods available for measuring glucose uptake in cells & tissues, 2-deoxyglucose (2-DG) has been widely used because of its structural similarity to glucose. As with glucose, 2-DG can be taken up by glucose transporters, and metabolized to 2-DG-6-phosphate (2-DG6P). 2-DG6P, however, cannot be further metabolized, and thus accumulates in the cells. The accumulated 2-DG6P is, therefore, directly proportional to 2-DG (or glucose) uptake by cells. Glucose Uptake Kits (Cell-Based) Hydroxyproline & Collagen Assay KitsHydroxyproline (Hyp) is a non-standard amino acid, found in collagen (COL), which contains 12-14% Hyp by mass. Hyp measurement is a direct measure of the amount of COL. COL is the most abundant protein in mammals and several conditions (i.e. tumor invasion/metastasis, rheumatoid arthritis, cardiopulmonary fibrosis) affect its turnover. Hyp and Collagen estimation protocols are based upon Hyp oxidation and Eh-rlich’s reagent under acidic conditions (Perchloric Acid- PCA). However, PCA requires special handling and waste-disposal protocols. BioVision’s Hydroxyproline Assay Kits (K555 and K206) and Total Collagen Assay Kits (K218 and K406) use proprietary developer solutions to accurately measure Hyp and COL in tissue and protein/peptidase hydrolysates. They are quick, can detect a minimum of 0.05 µg hydroxyproline/0.5 µg col-lagen per well in a 96-well format and generate colorimetric results that are virtually identical.
