SUPPLEMENTAL INFORMATION Preparation of artificial diet for rearing Beet Armyworm (Spodoptera exigua) NOTE: Details in this procedure refer to preparation of 10L of diet. Adjust amounts accordingly to prepare alternate amount (Table 1). The best way to prepare the diet, and control temperatures is by using a steam jacketed mixing kettle, such as this one: http://www.insectrearing.com/products/dpmixk.html 1. Weigh and gather the proper amounts of all ingredients before starting the kettle (see table below). a. All ingredients can be set out uncovered at room temperature except for the Aureomycin, which must be covered. Aureomycin becomes dried out and hard when exposed to air. b. For Vitamin Mix, cholesterol, methyl paraben, sodium benzoate, sorbic acid, and aureomycin i. Place household sieve over large cup ii. Use the sieve to break up clumps and to facilitate mixing. iii. Add the appropriate amounts of vitamin mix, cholesterol, methyl paraben, sodium benzoate, sorbic acid, and aureomycin to deli cup 2. Pour 3500 mL of Milli-Q water into the Kettle. 3. Turn the dial on the kettle to “10” and place the thermometer in the water. 4. While the water is heating up, place 180 g of Agar into a 2 L beaker 5. Add 250 mL Milli-Q water and stir with spatula 6. Add another 250 mL Milli-Q water 7. Continue adding water until 1000 mL of Milli-Q water is in the beaker and keep stirring until all of the powder is wet and mixed. a. Note: The water added to the agar keeps the agar suspended in liquid and prevents clumping. Actual amount of water needed to do so will vary and is not important. However it is important to not exceed the overall amount of water for the diet amount prepared (i.e. 8500 mL for 10 L). b. Note: DO NOT add the agar directly to the kettle and attempt to bring to a boil. Doing so will create a hard crust of burnt agar along the bottom and the kettle will shut off without bringing the solution to a boil. 8. Once the kettle temperature reaches about 85C, turn on the mixer a. Note: Throughout the rest of the procedure, have the mixer set to the highest speed possible that does not splash 9. Add the agar solution to the kettle. Take a small portion of water from the remaining Milli-Q water (approximately 500 mL) and rinse the contents of the 2 L beaker into the kettle. 10. Continue to heat the solution for about 1 minute after it begins to foam and boil.If the solution is not allowed to boil long enough, the agar will not dissolve properly and the diet with not solidify when cooled. 11. Turn the kettle dial to off. Add the remaining Milli-Q water (~ 3500 mL) to the kettle slowly. a. Allow the temperature to equalize before proceeding. There is often a large initial drop in temperature, but it quickly rises again. 12. Once the temperature of the solution cools to 75C begin to slowly add the ingredients listed in table 1, one by one. 13. Pause periodically to allow the powder to mix into the solution 14. Once the temperature of the solution has cooled to 70C begin to slowly add the remaining ingredients one at a time in the order listed. The sieved mixture should be added last. a. Again, pause between each one to allow them to thoroughly mix into the solution. 15. Turn kettle back on and place dial on #5. Temperature of diet should be around 60°C. 16. Place the diet into desired containers, allow to solidify and store at 4°C until ready to use.

Preparation of artificial diet for rearing Beet Armyworm (Spodoptera exigua)

The best way to prepare the diet, and control temperatures is by using a steam jacketed mixing kettle, such as this one:
