IndicationsPrevention of nausea and vomiting associated with highly emetogenic cancer chemotherapy, Prevention of nausea and vomiting associated with initial and repeat courses of moderatelyemetogenic cancer chemotherapy.Prevention of nausea and vomiting associated with radiotherapy in patients receiving eithertotal body irradiation, single high-dose fraction to the abdomen, or daily fractions to theabdomen.Prevention of postoperative nausea and/or vomiting. As with other antiemetics, routineprophylaxis is not recommended for patients in whom there is little expectation that nauseaand/or vomiting will occur postoperatively. In patients where nausea and/or vomiting must beavoided postoperatively, ondansetron tablets and syrup is recommended even when theincidence of postoperative nausea and/or vomiting is low.

DOSAGE AND ADMINISTRATION1. Prevention of Nausea and Vomiting Associated With Highly EmetogenicCancer Chemotherapyo The recommended adult oral dosage of Onlit is 24 mg given as three 8-mgtablets administered 30 minutes before the start of single-day highlyemetogenic chemotherapy, including cisplatin ≥ 50 mg/m 2. Multiday, singledose administration of a 24 mg dosage has not been studied.o Intravenous ondansetron has been shown to be equally effective in the followingdose schedules over the first 24 hours of chemotherapy:A single dose of 8mg( Onlit- Rapitab 8) by slow intravenous or intramuscular injection immediately before

A single dose of 32mg diluted in 50-100ml of saline or other compatible infusion fluid andinfused over not less than 15 minutes immediately before chemotherapy.The selection of dose regimen should be determined by the severity of the emetogenicchallenge.The efficacy of ondansetron in highly emetogenic chemotherapy may be enhanced by theaddition of a single intravenous dose of dexamethasone sodium phosphate, 20mgadministered prior to chemotherapy.2. Prevention of Nausea and Vomiting Associated With Moderately EmetogenicCancer Chemotherapy• The recommended adult oral dosage is one 8-mg Onlit Tablet given twice a day. The firstdose should be administered 30 minutes before the start of emetogenic chemotherapy, witha subsequent dose 8 hours after the first dose. One 8-mg Onlit Tablet should beadministered twice a day (every 12 hours) for 1 to 2 days after completion of chemotherapy.Pediatric Use• For pediatric patients 12 years of age and older, the dosage is the same as for adults. Forpediatric patients 4 through 11 years of age, the dosage is one 4-mg Onlit Tablet or 4 mg(2 teaspoonful equivalent to 10ml) of Onlit Syrup given 3 times a day. The first dose shouldbe administered 30 minutes before the start of emetogenic chemotherapy, with subsequentdoses 4 and 8 hours after the first dose. One 4-mg Onlit Tablet or 4mg (2 teaspoonfulequivalent to 2 mg of 10ml) of Onlit Syrup should be administered 3 times a day (every8 hours) for 1 to 2 days after completion of chemotherapy.

3. Prevention of Nausea and Vomiting Associated With Radiotherapy, Either TotalBody Irradiation, or Single High-Dose Fraction or Daily Fractions to the Abdomen• The recommended oral dosage is one 8-mg Onlit Tablet given 3 times a day.• For total body irradiation , one 8-mg Onlit Tablet should be administered 1 to 2 hoursbefore each fraction of radiotherapy administered each day.For single high-dose fraction radiotherapy to the abdomen , one 8-mg Onlit Tablet should beadministered 1 to 2 hours before radiotherapy, with subsequent doses every 8 hours afterthe first dose for 1 to 2 days after completion of radiotherapy.For daily fractionated radiotherapy to the abdomen , one 8-mg Onlit Tablet should beadministered 1 to 2 hours before radiotherapy, with subsequent doses every 8 hours afterthe first dose for each day radiotherapy is given.4.Postoperative Nausea and Vomiting PONVThe recommended dosage is 16 mg given as two 8-mg OnlitTablets 1 hour before inductionof anesthesia.For the prevention of PONV Ondansetron can be administered orally or by intravenous orintramuscular injection.Ondansetron may be administered as a single dose of 4mg given by intramuscular or slowintravenous injection at induction of anesthesia.For treatment of established PONV a single dose of 4mg given by intramuscular or slowintravenous injection is recommended.

Do not exceed therecommended dose.Contra-IndicationsOpidol-P is contra-indicated in patients with a known hypersensitivity to tramadol,paracetamol or other opioids such as codeine.It is also contraindicated in cases of severe liver function impairment and in acuteintoxication with alcohol It should not be administered to patients who are receiving monoamine oxidaseinhibitors or within two weeks of their withdrawal.Opidol-P must not be used for narcotic withdrawal treatment

Opidol-P should not be given to patients with respiratory depression especially in thepresence of cyanosis and excessive bronchial secretions.Opidol-P should not be given to patients with increased intracranial pressure or centralnervous system depression due to head injury or cerebral disease.Safety during pregnancy and lactation has not been established. Tramadol has beenshown to cross the placenta.
