Operating manual

Thank you for choosing a DKL Velp fully automatic digestion unit. DKL carries out the fully-automatic preliminary digestion of a variety of samples (food, animal feed, fodder, soil, water…) for nitrogen/protein determination using the Kjeldahl method. The analytical procedure is then completed by treating the digested samples in a Velp UDK steam distillation unit. Safety warnings:

Before using the unit, please read the operating manual supplied with the apparatus carefully. Warning! Hot surface! Protective conductor terminal. Do not dispose of this equipment as urban waste.

This unit must be used for laboratory applications only.

The manufacturer declines all responsibility for any use of the unit that does not comply with the instructions. Improper use or not specified could compromise the safety of the product. SAFETY PRECAUTIONS In order to prevent the risk of electric shock, fire and personal injury when the unit is in use, basic safety measures must always be taken including:

Ensure that liquids do not come into contact with the electric power cable or with the electrical parts of the instrument.

Check that the power supply corresponds to the rating plate on the rear of the unit.

When replacing the power cable, make sure that the new cable has the same characteristics as the original and that it is earthed.

Do not use the unit if it is not working correctly. In case of malfunctioning, contact your nearest service centre.

The personal protective equipment, containers and products used must be compatible with the temperature setting on the instrument.

The personal protective equipment used must be compatible with the risks arising from the material being processed and the glass containers.

The heating block can reach temperatures of up to 450°C. High temperatures are reached and/or maintained during the heating stage, during the work cycle and during cooling at the end of the work cycle.

For applications where the digestion process produces fumes, acid gases or corrosive substances (e.g. the Kjeldahl method), the instrument should be combined with specific fumes suction and abatement systems. See Velp’s models JP fume suction pump code F30620198 and SMS scrubber code F307C0199.

1. Introduction

VELP digestion/mineralization units are designed for various applications from the determination of nitrogen and protein according to the Kjeldahl method (TKN) in food analysis, to environmental analyses (Chemical Oxygen Demand) and chemical-pharmaceutical applications. Velp Scientifica heating blocks are designed for the wet digestion of liquid or solid samples of up to 15 ml or 5 g in size. The DKL Series includes a wide range of models for the simultaneous digestion of 8, 12, 20 or 42 samples using test tubes of various dimensions: DKL 8 (8 x 250 ml), DKL 12 (12 x 250 / 400 ml), DKL 20 (20 x 250 ml), DKL 42/26 (42 x 100 ml). The digestion time depends on the type of sample and the method used. The temperature range varies from 10 to 450°C, ideal for all types of standard digestion as well as for the Kjeldahl method. The compact design of the instrument saves valuable laboratory bench space. With Velp Scientifica digesters, working conditions are totally reproducible and safe as well as reducing the consumption of reactives and saving time thanks to faster heating. The low power consumption reduces the cost of each analysis and limits emissions of CO2 into the environment.

Check to make sure the digester system is complete after unpacking. Identify the parts included in the supply of the model ordered and check to make sure that all the components listed in the relative column are present. The various components are packed individually.

The DKL uses innovative techniques and state-of-the-art technology:

The structure of the digester is manufactured from stainless steel with a protective coating of special resins and paints studied and tested to ensure a high resistance to chemical and mechanical agents.

The heating block is manufactured from an aluminium alloy which ensures excellent temperature homogeneity at all temperature settings resulting in complete and homogeneous digestion and a high level of reliability.

The heating block temperature is controlled by a microprocessor with PID logic. The temperature probe is a Pt100 platinum probe which carries out self-calibration each time the unit is turned-on. This ensures the high precision and reproducibility of tests.

The user interface has an innovative graphic display and multi-functional keys, is quick and easy to use and guides the user during programming as well as monitoring the various stages of the digestion process.

The innovative electronics with rewritable microprocessor makes it possible to update the software with new versions or new languages simply by downloading the upgrade from a PC via USB connection.

The automatic handling of samples in and out of the heating block including lifting for cooling, offers increased user-safety thanks to the absence of manual activities.

The instrument is protected against overheating by a thermostat with manual reset. A probe located on the heating block cuts off the power supply when a temperature of more than 500°C is detected. The manual reset button is located on the bottom of the instrument (identified by a label marked “SAFETY THERMOSTAT”).

The Program menu contains up to 54 work programs including 30 presets. For each work program it is possible to select up to four ramps (4 temperatures with the relative time of permanence). Temperature settings ranging from 10°C to 450°C can be selected and the time of permanence at the selected temperature can be set within a range of 1 to 999 minutes. The Print menu can be used to enter data relative to the test or tests in progress which can then be transferred to a computer and stored in accordance with GLP (Good Laboratory Practice). It is therefore possible to save and/or print sample data and test parameters for future reference. The Language menu can be used to choose the language in which information is shown on the display. It is possible to select one of the following five languages: Italian, English, French, Spanish, Russian and Chinese. The Settings menu can be used to choose the unit of measure in which temperature is shown on the display (°Celsius o °Fahrenheit o Kelvin). It is also possible to modify the date and time and to activate or disactivate warning messages. DKL digesters guarantee unmatched savings whist ensuring an extremely high level of reliability. 1.3 Keys and functions

MAINS SWITCH

The mains switch is used to turn the instrument on and off. In the position marked I the green light indicates that the instrument has been turned on.

CONTROL PANEL

The control panel has a graphic display and 4 multi-functional keys offering easy access to the instrument’s various functions and offering guided operator support during programming and standard use. Each window that appears on the display gives a description of the function carried out by each key. In this way it’s possible access to the 6 main menus.

The START and STOP keys can also be found on the control panel.

Use the START key to start the program selected. Use the STOP key to:

stops the work-cycle

return to the main menu.

2. Assemby and installation

2.1 Assembly

Place the instrument on a clean, level and non-flammable surface.

Turn the digester so that the back of the instrument is facing you and begin

installing the lift.

Remove the 4 screws on the rear casing (1).

Position the lift against the instrument aligning the 4 holes with the 4 points of attachment (1).

Use the previously removed screws to fix the lift to the instrument (1).

Connect the lift cable to the relative socket labelled LIFT at the back of the instrument (2). This socket is isolated from the live parts by double insulation.

Turn the digester so that the control panel faces you.

Place the sample rack on the lift using the appropriate hooks (3).

Remove the drip tray from the suction cup.

Place the suction cup on the lift using the appropriate hooks (4) and be sure that the glass parts of suction cup are correctly aligned with test tube/sample rack.

Position the instrument at a distance of at least 15cm from any walls. NOTA: Always remove the lift before moving the DKL.

2.2 Hydraulic connections

Connect the viton tube 10x14 (supplied with the SMS Scrubber), to the pipe union on the left-hand side of the suction cup.

NOTE For applications where the digestion process produces fumes, acid gases or corrosive substances (e.g. the Kjeldahl method), specific accessories are required as well as fumes suction and abatement systems. Velp Scientifica offers the following systems: JP recirculating water pump for fumes aspiration (code F30620198) SMS Scrubber (cod. F307C0199) Failure to use the abovementioned instruments and accessories in these applications will result in the warranty becoming void. Diagram showing how to connect the various instruments:

When using the DKL 42/26 digester, Velp recommends the use of 2 JP pumps in order to guarantee efficient fumes removal (see the connection scheme below).

2.3 Electrical connection

Before connecting the unit to the power supply make sure that the mains switch is in the OFF position “0” and check that the values on the rating plate of the unit correspond to those of the power supply. Make sure that the mains circuit is earthed. Connect the unit to the power supply using the earthed cable supplied, the socket should be easily accessible. NOTE: When the mains switch is turned off the unit is completely disconnected from the mains circuit avoiding any waste when the unit is not in use.

3. Operating controls

3.1 Quick guide

This page shows the software of the heating block. The purpose is to help the user identify the different possibilities offered by the instrument and how to reach them easily.

3.2 Main menu

When the instrument is turned on, the display shows the name of the manufacturer and the software version installed for a few seconds.

The standard window is then displayed:

The first line shows the model of DKL (top left) and the date/time (top right). The second line shows the name of the last program used. The center of the display shows the temperature of the heating block and the ramp settings of the program.

To start the work-cycle press key .

The ▼ and ▲ keys (bottom left) move the Lift up and down.

When the Lift is active, the display shows “Attention Lift Movement”. Be careful with moving parts.

NOTE: If the Lift cable is not connected to the Digester, the keys ▼ and ▲ are not visible.

The Menu key gives access to other functions (main menu).

The "Main Menu" offers the following sub-menus that can be selected using the ▼ and ▲ keys:

3.3 Program menu

The "program menu" offers a variety of programs using standard methods as well as a number of programs that can be used to create personalized methods for specific requirements.

3.3.1 Program selection

Press OK to access the Program menu:

Press OK to access “Standard methods” offering 30 standard programs that cannot be edited, or “Personal methods” offering 24 customizable programs. The checkmark indicates the program selected.

Press OK to confirm:

The display shows the parameters of the 4 ramps. Press Confirm to confirm and access the standard window to start the work-cycle. If a customizable program has been selected, press Edit to modify the ramp parameters.

3.3.2 Parameter settings

To create a customized method, press Edit to access the "Edit Method" screen:

Select "Program Name" and press the Edit button. Use the alphanumeric keyboard to assign a name to the program in question.

Use the ◄ and ► keys to move the cursor and OK to confirm the letter selected. You can enter up to 14 digits for each data field. Press Save/Exit to save the changes and return to the previous menu.

Confirm the program name by pressing Yes. Select "1st ramp" from the "Edit method" screen using the ▼ and ▲ keys. Press Edit to confirm.

Use the + and – keys to increase or decrease the temperature value (keep pressed for fast run). Press OK to confirm, the cursor will automatically move to the Time value which can be modified in the same way.

Press Save/Exit followed by Yes from the “Confirm action” window, to save the modification and to return to the previous menu.

3.4 Print menu

The instrument has a USB connection on the rear panel that can be used for transferring and storing data according to Good Laboratory Practice (G.L.P.). The Print menu allows the user to choose whether to transfer data automatically at the end of each work-cycle or only when requested. This menu also allows the user to enter and store data concerning the sample examined and the parameters used for each test. Press Exit to return to the previous menu.

3.4.1 Print data

It is possible to transfer the data referring to the method selected to a HyperTerminal using the PC UBS interface settings. Select "Print Data" using the ▼ and ▲ keys and press OK to confirm. Press Exit to return to the previous menu.

3.4.2 Data settings

Use this option to enter the test parameters that will be saved on file. Select “Set up data” and press OK to confirm.

Select the parameter to be modified and press OK. Use the alphanumeric keyboard to modify the parameter selected.

Use the ◄ and ► keys to move the cursor and OK to confirm the letter selected. You can enter up to 14 digits for each data field. Press Save/Exit to save the changes and return to the previous menu.

3.4.3 Print options

Use this option to modify the settings for data transmission. Select “Print options” and press OK to confirm.

Use ◄ and ► to select automatic data transmission of data at the end of each digestion. For manual data transmission select "Print data". Press OK to confirm or Exit to return to the previous menu without saving the modification.

3.4.4 PC USB interface settings

Test data can be transmitted and stored on a PC by connecting it to the USB socket labeled USB at the back of the digester (this socket is isolated from the live parts by double insulation). The Hyper Terminal program installed in Microsoft Windows 95 – 98 -2000 – XP - Vista and Windows 7 software can be used with the digester connected to a PC. Use the following PC serial connection settings:

Baud rate 19200 Data length 8 Parity None Bit of Stop 1 Handshaking None

PC Connection: Use a standard USB cable type A/B. In case of problems of connection between instrument and PC, we recommend to download the updated driver FTDI from the manufacturer website: http://www.ftdichip.com/Drivers/VCP.htm

3.4.5 Data transmission file

3.6.7 Blackout alert

Select “Blackout alert” and press OK to confirm. This function can be used to record the duration of the power failure in order to assess the possibility of continuing the analysis (see section 5). If this option is turned off, the work-cycle will automatically be continued after a power failure.

3.6.8 Automatic Lift

Select “Automatic Lift” and press OK to confirm. If this function is disabled the Lift must be moved up and down manually by the operator using ▲ and ▼. With the function enabled lift movement is controlled automatically by the digester at the beginning and end of each analysis.

3.6.9 Drip tray alert

Select “Drip tray alert” and press OK to confirm.

3.7 Instrument information

Select “Instrument information” and press OK to confirm. In this menu is possible view the software version installed on the digester and get the information about the number of tests performed and the movement of Lift.

3.8 Service menu

Select “Service Menu” and press OK to confirm. From this menu it is possible modify the instrument settings and update the software.

3.8.1 Update software

Select “Update software” and press OK to confirm. The software can be updated via a USB port. Press OK on the confirmation screen and launch the dedicated program from the PC .

Software version, dedicated program to be installed in the PC and software installation guide have to be requested by e-mail to [email protected].

3.8.2 Keyboard Check

Select “Keyboard check” and press OK to confirm . Use to test the function keys on the digester.

Press each function key in sequence. If the key is working correctly a checkmark √ will appear. Check each single key before pressing Exit to return to the previous menu.

3.8.3 Lift Check

Select “Lift Check” and press OK to confirm.

Press OK to perform an automatic self-test on the lift. At the end of the test, the result is displayed on the screen.

Press Exit key to return to the previous menu.

4. Start heating

To start the work cycle select the desired program and press the START button from the main menu. The instrument will automatically display the last program used.

Press to start the program displayed. If the lift is in the up position the following warning message will be displayed (only if the drip tray alert is enabled):

Press a second time and the LIFT will automatically move to the operating position. Digestion starts and the display highlights the first ramp.

NOTE: The symbol indicates that the instrument is heating. When the temperature of the heating block reaches the set temperature, an acoustic signal indicates the start of the set time. The display shows:

Once all the ramps have been performed heating stops and the lift moves to the middle position.

If during the set time is pressed, the instrument will run continuously at the set temperature and the display shows:

To interrupt the work-cycle press . NOTE: When starting a new program, if the temperature of the heating block is higher than the set temperature the work cycle will not start. In order to start the work cycle wait until the block cools to a temperature lower than the set temperature. NOTE: If you did not use all the positions of the digester, use the accessory A00000243 " Glass cap" for empty positions, placing it in the sample rack.

5. Arrests

5.1 Blackout

In the event of a blackout or power failure, when the power returns the work cycle automatically continues from the point where it left off if the blackout function is disabled. However, if the blackout function is active, a warning message indicates the duration of the power failure and the operator can decide whether to continue the analysis or not.

5.2 Faulty probe

If the probe measuring the temperature of the heating element is faulty, the following warning message is displayed:

NOTE: In this state it is only possible to move the Lift.

5.3 Overheating

If the temperature of the heating is too high, the following warning message is displayed and the work-cycle is automatically interrupted.

NOTE: Before starting a new analysis, allow the heating block to cool down. If the instrument no longer heats-up, see paragraph 7.2.

6. End-of-work operations

At the end of each test it is good practise to place the drip tray in its’ seat to prevent any liquid dripping onto the heating block. Before cleaning the instrument, disconnect it from the power supply by removing the plug and make sure that the heating block is cool. Clean the unit with soft cloth and a non-flammable, mild detergent. NOTA: To keep the instrument in optimum condition, any drops of acid that drip onto any of the surfaces should be removed immediately.

7. Maintenance

Routine maintenance and extraordinary maintenance are not necessary with the exception of periodically cleaning the instrument as described in the previous paragraph. The instrument must be transported in its original packaging and any indications present on the original packaging must be followed (e.g. palletised).

7.1 Replacing the fuses

Replace the fuses only when necessary, i.e. if the unit doesn’t turn on and the mains switch does not light up. The fuses must only be replaced by qualified personnel and only with fuses supplied by Velp. To replace the fuses is needed: - unplug the unit; - locate the fuse holders at the back of the instrument; - unscrew the fuse caps.

7.2 Rearming the safety thermostat

If the display is on but the heating block does not heat-up, check to see if the safety thermostat has tripped cutting-off the power supply to the resistance. The safety thermostat trips if a temperature of higher than 500°C is detected. The manual reset button is located on the bottom of the instrument (identified by the label SAFETY THERMOSTAT).

7.3 Lubrication of the guide

If the sample rack or the suction cap move hard, set the lift in the top position and lubrificate the guide with a dry spray lubricant (es. SVITOL TECHNIK - DRY) to limit the friction. During this operation mind the sensors on the topside of the guide.

8. Disposing of the unit

The instrument is classified as electrical/electronic apparatus and must be disposed of accordingly. The unit is subject to waste separation and cannot be disposed of as urban waste under EEC directive 2002/96/CE. For more information please contact the relative division of your local town council.

9.2 Accessories for testing the heating block temperature

The Digester has a built-in electronic temperature control system resulting in excellent temperature control without over-heating and without temperature oscillations near the set point. The PT 100 temperature probe on the heating block ensures excellent precision. The probe is calibrated automatically each time the unit is switched on thanks to the microprocessor software, therefore temperature alignment is not necessary. Should it be necessary to check the temperature readings of the instrument, this can be done by placing an original Velp Scientifica thermometer in the hole on the top of the heating block. Before proceeding with the control sure you have removed the sample rack and the suction cup.

13. Warranty

The unit is guaranteed against production defects for 25 months from our invoice date. Warranty claims can be made only if the system has been installed and used as specified on the manual and carried out by qualified service personnel as appointed by VELP Scientifica. In accordance with this guarantee VELP SCIENTIFICA undertakes to repair any units resulting as faulty due to the quality of the materials used or poor workmanship. Units rendered faulty due to inexpert handling/use or carelessness will not be replaced or repaired under warranty. For more details please contact your Distributor. Exclusions: The guarantee will be considered null and void for faults resulting from: - inexperience and carelessness of the operator, - repairs, maintenance or replacement of parts carried out by personnel or
