Mucoci l iary t r a n s p o r t is considered t o be an important defense mechanism a g a i n s t i nha led tox ican t s and microorganisms. Never the less , l i t t l e is known about pul­monary c i l i a , t h e i r s t r u c t u r e , how they func t ion , and how t h e i r a c t i v i t y i s con t ro l l ed .

Most knowledge of t h e movement and biochemistry of c i l i a has been der ived by e x t r a p o l a t i o n from work wi th u n i c e l l u l a r organisms o r i n v e r t e b r a t e s . Although the biochemistry and t h e b a s i c mechanisms of m o t i l i t y may be the same i n mam­malian ce i l ia as i n t h e more primitive ones, t h e p a t t e r n of movement and mechanism of c o n t r o l are s p e c i a l i z e d f e a t u r e s t h a t vary from organism t o organism and from t i s s u e t o t i s s u e . No f u n c t i o n a l s t u d i e s of mammalian lung c i l i a can r e l i a b l y be regarded as r e f l e c t i n g t h e normal s i t u a t i o n because the a c t i v i t y of the c i l i a , and the amount and na tu re of s e c r e t o r y a c t i v i t y , may be per turbed by p r e p a r a t i v e procedures .

Recent ly , primary abnorma l i t i e s i n t h e f i n e s t r u c t u r e of human b ronch io la r c i l i a have been observed i n persons wi th s i t u s inve r sus , chronic s i n u s i t i s and b r o n c h i e c t a s i s (Kartagener 's t r i a d ) which i s a n i n h e r i t e d syndrome a s s o c i a t e d wi th repea ted pulmonary i n f e c t i o n s . But whether o t h e r g e n e t i c d e f e c t s i n c i l i a r y s t r u c t u r e may be a s s o c i a t e d wi th pulmonary d i s o r d e r s i s e n t i r e l y unknown. Nor is i t known how exogenous ( t o x i c a n t s , pharmacological agen t s ) o r endogenous (hormones, mediator) compounds a f f e c t c i l i a and t h , e i r func t ion .

The purpose of t h i s announcement is t o encourage submission of r e sea rch g ran t a p p l i c a t i o n s which focus on i n vivo o r i n v i t r o s t u d i e s of t h e s t r u c t u r e and func t ion of normal mammalian c i l i a from t h e upper lower r e s p i r a t o r y t ract . S imi l a r s t u d i e s , however, have not been carr ied o u t i n the lung. A v a r i e t y of ch ron ic and progress ive pulmonary d i s e a s e s r e s u l t from t h e i n h a l a t i o n of o rgan ic o r i no rgan ic material. Agents which are known t o produce c e l l u l a r i n f i l t r a t i o n and/or f i b r o s i s i n the lung inc lude such d i v e r s e subs tances as pigeon droppins , fung i , act inomycetes , n i t r o f u r a n compounds, s i l i c a , and bery l l ium salts. Although s t u d i e s have i n v e s t i g a t e d t h e in f luence of t h e s e agents on lung d i s e a s e , l i t t l e is known regard ing the c e l l u l a r responses of t h e lung t o t h e s e and o t h e r inha led agents o r of t he e x t e n t t o which these responses are p r o t e c t i v e o r d e s t r u c t i v e . Inha led subs tances may produce changes i n immunologicaily-active cells r e s u l t i n g i n de t r imen ta l e f f e c t s which may o r may not be of an immunologic na tu re . It is a l s o p o s s i b l e t h a t a u t o r e a c t i v e responses , such as t h e product ion of a n t i n u c l e a r ant ibody, might arise as a r e s u l t of pre l iminary c e l l u l a r damage caused by a n inha led agen t , and t h a t such auto-immune phenomena might p lay a r o l e i n ch ron ic lung d i s e a s e i n i t i a t e d by t h e i n c i t i n g agent .
