1 Introduction
Thank you for purchasing the Meros High Speed Digital Microscope System
from Dolomite Microfluidics.
These instructions have been carefully prepared to guide the installer and
end-user through the installation and use of the High Speed Digital
Microscope System.
Before attempting to use this product, please read this manual thoroughly
and follow the instructions carefully. In doing so you will ensure the safety of
yourself and that of others around you, and you can look forward to your
purchase giving you long and satisfactory service.
Please keep the manual easily accessible and in immediate vicinity of the
module. Only suitably qualified and trained personnel should operate this
unit.
The High Speed Digital Microscope System is a high quality and flexible
solution for general microscopy and high speed image capture in microfluidic
applications. It features an easy-to-use microscope with wide zoom range
and long working distance. The microscope stage is designed to
accommodate all types of microfluidic chips and enables users to quickly
locate and observe the area of interest.

2 Safety
Warning and safety information
Do not look directly at the light coming from the lens of the
Microscope or from the reflection from the light control stage. It is
extremely bright and may cause eye damage.
The camera housing may be hot, avoid direct contact especially
after long periods of use.
The Microscope has an uneven weight distribution, take care
during assembly and if moving.
Do not cover the instrument during use.
This equipment should only be used by competent, suitably trained
personnel after they have read and understood this manual, and
considered any hazard involved.
Place the instrument on a solid horizontal surface. Ensure the area
around the unit is clear.
Adequate protection including appropriate PPE and ventilation
must be provided if hazardous chemicals are to be used in
conjunction with this unit. In the case of accidental spillage,
carefully wipe with a dry cloth, taking into account the nature of the
spilled liquid and the necessary safety precautions.
Comply with all safety and accident prevention regulations
applicable to laboratory work.
Always connect the instruments to an earthed AC power outlet.
The operating voltage is indicated on the specification sticker. Non
observance of this provision may result in damage to the
Microscope or in personal injury or damage to property.
This product does not comply with the ATEX directive and must
not be used in explosive atmospheres.

Disassembling any part of the Microscope may invalidate the
warranty.
Cleaning
Cleaning should only be performed by personnel trained in such
work, and who are aware of the possible dangers involved. The
microscope (and all the associated hardware) has not been
designed for sterilisation or use with an autoclave.
Returning Equipment
Equipment which has been contaminated with, or exposed to, body fluids,
toxic chemicals or any other substance hazardous to health must be
decontaminated before it is returned to Dolomite-Microfluidics or its
distributor.

3 Using the High Speed Digital Microscope
Overview The Meros High Speed Digital Microscope is designed specifically for microfluidics and is ideal for observing droplets during high speed droplet production. The high speed, high resolution imaging and the convenient stage greatly simplify microfluidic experiments. The easy to use stage securely holds a wide range of microfluidic equipment for reliable viewing including microfluidic chips and. With the benefit of extra-long working distance, the High Speed Digital Microscope enables optical access to samples which would normally be difficult to view. Illumination from above leaves the underside of the microfluidic device free for temperature control and is compatible with the Meros TCU-100 (part no. 3200428). High magnification optics and the zoom lens ensure that micro-scale features can be clearly visualised. The integrated coaxial lighting system offers exceptional brightness for image capture at very low exposure times. Coupled with the high speed USB3.0 camera, the lighting and optics provide the capability to capture still images or videos of microfluidic material moving at kHz rates. The High Speed Digital Microscope is fully compatible with the Flow Control Centre software (downloadable for free from Dolomite website) which enables observation, image adjustment and image capture. Flow Control Centre also includes advanced functions (accessible with a purchased licence key) which allows complex protocols to be run with the capability to control Dolomite imaging systems, pumps, sensors and temperature control devices simultaneously”. Figure 1 shows the fully assembled High Speed Digital Microscope System

Assembly Instructions Screw the upright post into the base using the 3 Allen
screws and Allen key provided. See Figure 2.
Fit the microscope assembly onto the stand and tighten the
lock knob. See Figure 3.

Position the light control stage under the microscope, plug
the power supply/power cord into the light control stage. See
Figure 4.
Plug the light control stage cable coming from the
microscope into the socket on the light control stage. See
Figure 5.
Plug the USB 3 cable from the microscope into your
PC/laptop.
Fit the diffusor/reflector glass on the top of the light control
stage. See Figure 6.
NOTE: The best performance will be achieved when the microscope is plugged into a USB 3 socket. If
plugged into a USB 2 socket, performance will be reduced as the data transfer rate is slower with this
set up.

Fit the chip clips if required. The clips screw into a choice of
positions on the deck of the light control stage to allow for a
variety of possible configurations. See Figure 7.

Installation of Flow Control Centre Software
The High Speed Microscope can be controlled via our Flow Control Centre
software. This is available to download FOC from the Dolomite Microfluidics
downloads area, http://www.dolomite-microfluidics.com/en/downloadsmain
and click on “software downloads” from the drop-down menu. In order to
download this, you must register on the website, it only takes a few minutes
and no credit card details are required.
Click on the downloaded file and follow the on-screen instructions.
Attach the microscope to your PC via the USB 3 cable.
For user instructions on how to use this software, select ‘help’ from
the top menu bar.
Your High Speed Digital Microscope is now ready to use.
NOTE: Flow Control Centre Software is only compatible with Windows 7.
NOTE: Please note that versions of FCC earlier than 3.x.x are not compatible
with this microscope.
NOTICE: When starting up your PC, the USB lead between the Microscope and the PC MUST be
disconnected. This applies even if the Microscope is turned off. Failure to do this may result in the
Microscope not being recognised as a USB device and will not be able to communicate with the Flow
Control Centre Software.
NOTE: We have a Droplet Monitoring Software that will allow you to analyse droplets from a saved
.avi file. The link below will take you to our software downloads page. Please take care to download
the correct version for your PC as 32 and 64 bit versions are available. http://www.dolomite-
microfluidics.com/webshop/software_downloads
You will require a licence to run this software. Please contact our support team support@dolomite-
microfluidics.com to request a licence key.

4 Operating the Microscope
Physical Microscope Adjustments Coarse Height Adjustment
Whilst supporting the microscope, turn the lock knob anti-clockwise
to loosen the Microscope. With the Droplet Monitor Software
running, lower or raise the microscope to the required height so that
it is roughly in focus. See Figure 9.
Fine Height Adjustment
NOTE: You also have access to additional software tools including Microfluidic Flow Calculator and
Droplet Volume Calculator, to help you set up your microfluidics system. Contact Support@dolomite-
microfluidics.com if you require any of these aids.

Moving either of the Fine Height Adjustment knobs will allow
the Microscope to be raised or lowered with fine control to
achieve a sharp focus. See Figure 10.
Tension Adjustment
The force required to move the fine
adjustment knob can be varied. Hold the
right hand knob (plain) still whilst adjusting
the left hand knob (marked) clockwise to
increase the tension, or anti-clockwise to
reduce the tension. See Figure 11.

Magnification Adjustment
Adjust the dial to the mark to change the magnification, 1 is the
lowest magnification, 5 is the highest. See Figure 12.
Light source Adjustment
Adjust the dial to change the brightness at the light control
stage. See Figure 13.
Securing the chip
Position the clips to suit the chip. Push down of the plastic
arm of the clip to secure the chip. See Figure 14.

Flow Control Centre Microscope Settings Within Flow Control Centre, click on the “Devices” tab to access a
Microscope specific and the pop-up window, as seen below.in Figure 15.
This window gives access to parameters that will change the
appearance and quality of the image that can be captured. There are
8 Icons in total that allow access to the control settings, image capture
settings, file save settings and Region of Interest (ROI) selection.
4.2.1 Individual Icons Function
Pop-Up
Opens a screen where just the live image can be seen
without clutter.
Settings
Opens a new window, see Figure 16 below.
Image
Click to capture a single image.
Video
Click to capture video, time can be set between 1 and 600
seconds.
Open
Open the default folder where images are stored.
Change
Change the default folder where images are stored.

ROI
Click and draw a box on the image window to select the ROI.
Re-set ROI
Click to re-set the ROI to full size.
Exposure Time
This is the equivalent to shutter speed in a conventional camera.
Click on "Exposure" slider and drag to obtain the desired
exposure time setting. We suggest that you initially select the
shortest possible 0.05ms (50µs). The sharpest images will be
taken with the smallest exposure time and this is especially true
when looking at droplets being produced in the order of kHz.
Alternatively, select the tick box to automatically determine the
exposure time
Gain
This is the equivalent to film speed in a conventional camera.
This can be useful when lighting levels are low. Increasing the
gain boosts the signal and hence appears brighter. However, it
should only be necessary to adjust this setting if working under
low light conditions or with high speed droplet production. Please
note the increasing the gain will also increase noise levels
causing the image to be less clear.
Frame Rate
This is the number of frames captured per second (FPS) during
video capture. Increasing the FPS will slow down the apparent
movement of the droplets.
Gamma
This setting increases contrast e.g. improves the definition.
Image Flip
Depending on the Droplet Imaging Software you are using, this
allows the direction of the droplet flow to be oriented correctly. Our
Droplet Monitor Software requires that the droplets are moving from
left to right.
Region of Interest (ROI)
Use this feature to select the region that you wish to view/capture.
Note that the larger ROI, the larger the file size and the lower the
frames/second rate.
Undo Region of Interest
Returns the ROI to full screen.
Quick Start Summary
Set the exposure time as low as possible, 0.05ms
recommended.
Increase the lighting brightness.
If more light is still required, increase the gain.
5 Support
Servicing There are no user serviceable parts in the high speed microscope.

The LED light source brightness will be reduced over a period of time. We
suggest that your microscope is refurbished when the brightness too low for
normal use (approximately 3000 hours of use). Please contact
[email protected] for further details.
Cleaning The lens can be gently wiped using a lint free wipe (clean room wipe).
Cleaning liquids should not be used as these can damage the optical coating
on the lens.
All other components of the microscope and should be kept free of dust and
fibres. Disconnect the microscope from the mains power supply before
cleaning and also ensure that is completely cool. The surfaces can be
cleaned with a lint free wipe and a small amount of iso propyl alcohol.
FAQ Q. All I can see is a white screen, what do I need to change?
A. There are 3 settings that could cause the screen to appear white, you will
need to adjust 1 or more of these to obtain the desired lighting level.
The exposure is too high, click on the auto button or manually select a lower
value. The recommended starting value for exposure time is 0.05ms
The light source is to bright, reduce the brightness adjustment knob.
The gain is too high, select a lower value.
Q. My image is too dark, and I can’t see droplets. What can I
do?
A. Increase the light to maximum, and increase gain until image is bright
enough.
Q. My chip won’t sit properly on the stage.
A. You can clip the chip on with the clips on the stage (see Figure 11 above).
If the clips are missing, you can order replacements (3200430).
Q. I think there should be droplets, and everything seems fine,
but I can only see a blur or lines at the junction.
A. There are several possible causes.
The droplets may actually be blurred. First, decrease the exposure time to
the minimum (0.05ms), and adjust the image so you have a bright, focussed
image.
Check that the image is live (communication hasn’t ‘hung’) by moving the
microscope state – the image should also move.
If you can see what looks like jetting flow through the junction, decrease first
the droplet phase flow rates, and then the carrier phase until the jetting
changes to ‘dripping’.

Check that you actually have flow going through the chip. If there is a
blockage, check the chip, fittings and tubing until you find the blockage.
Q. My droplets appear to be moving backwards, why is that?
This is a stroboscopic effect due the number of Frames per Second and the
Hz rate of the droplets being produced. You may have seen this before in a
movie where the propeller of a plane appears to move backwards. You can
change whether the droplets appear to move forwards, backwards or remain
stationary by adjusting the Frames per Second. This will not affect the
quality of the data.

Blacktrace software is not Windows 8 or 10 compatible

Recommended: Super VGA 1920 x 1080 or better.
Video Card: 24-bit or 32-bit with at least 64MB of video memory.
Native USB Ports Varies based on system, but the minimum is 1 x USB 3 ports and 3
x USB ports. We strongly recommend all Blacktrace hardware is
connected directly to the PC.
Additional USB ports can be added to a PC using only Blacktrace
approved powered USB Hubs.
USB Cables Shielded USB Cables can be no more than 3 meters in length from
PC to device.
Third Party Software The PC must be dedicated for Blacktrace software and no third
party software should be installed.
Anti-Virus Recommended: Symantec
Other Anti-Virus and Firewall software can prevent the Blacktrace
software from running properly
Regional Settings ‘UK English’ must be selected in Regional Settings, not just
language settings
