Estimulación del complejo TCR induce fosforilación de tirosinas de ITAMs en CD3 y reclutamiento de moléculas efectoras. CD4 (unido a lck) se acerca a TCR por interacción con residuos no polimórficos en MHC. Removal of the antigen leads to rapid recovery of B-cell receptor signaling, and might result in autoimmunity due to activa tion by crossreactive immunogens.30 Naive T cells respond to continued antigen presentation by initial expansion and subsequent death of a subset of these cells, but also develop functional unresponsiveness in the remaining cells, which frequently cannot be reversed by the applica-tion of IL-2. This phenomenon might be caused by intracellular blockade of IL-2 receptor signaling by CTLA4-derived signals and a failure of cell-cycle progression.28,31
The existence of two distinct (though possibly overlapping) types of anergy have been
proposed, which differ in their mechanisms of induction, dependence on continued antigen presentation and responses to IL-2. Thorough reviews of the different types and mechanisms of anergy have been published.28,32 Despite these differences, all potential subtypes of lympho-cyte anergy share the common characteristic that they represent an active process rather than merely a passive state of unresponsiveness, as indicated by the requirement for calcineurin activity and protein synthesis.33,34
Early studies indicated a defect in signaling path-ways upstream of IL-2 gene transcription,35–39 as well as inhibition of IL-2 gene expression at the promoter level40,41 and selective upregula-tion of expression of general receptor of phospho-inositides 1.42 More recent mouse studies have identified certain E3 ubiquitin ligases as impor-tant anergy factors. These include the RING finger domain E3 ligases Cbl-c, Cbl-b, and GRAIL

Specific antigens can be targeted to immature dendritic cells in vivo by coupling them to a modified antibody molecule, which binds the dendritic cell marker DEC205. These modified dendritic cells efficiently present the loaded antigen to the T-cell receptor of antigen-specific T cells in secondary lymphoid organs.
