4 METHODS
4.1 NMR Spectroscopy
1H-NMR spectrum and
13C-NMR spectra were recorded to characterise the test items. Approx.
200 mg of each test item have been dissolved in 1 ml DEUTERATED SOLVENTS.
The actually used NMR parameters appear on the spectrum plot.
For quantification according to standard operation procedure SAA-MET001-02 appropriate amounts
of each test item and of internal standard have been exactly weighed (s. Table “Calculation of
content”, Chapter 5 - NMR Spectroscopy), dissolved and measured. Integrated signals of test items
and internal standard have been used for calculation.
To characterize the test item and to determine the content of the phospholipids "Nuclear Magnetic
Resonance Spectra“ of phosphorus (31
P-NMR spectra) were recorded. The quantifications were
carried out against a reference item according to the standard operation procedure SAA-MET002-02.
The procedure corresponds to „USP method <761> NUCLEAR MAGNETIC RESONANCE“.
For the quantification two measurements were performed for each sample: one with 1 whole capsule
and one with only the filling of 1 whole capsule. Each test item and 20 mg of the internal standard
(TPP) were exactly weighed (see tables “Calculation of content”, Chapter 6 - NMR Spectroscopy),
dissolved in 10 ml CDCl3 / MeOH / Cs-EDTA, aq. (2/4/4) and measured. The integrated signals of
test item and internal standard were used for calculation.
The ratio of integrals per atom corresponds to the molar ratio of the compared substances. For
calculation software Microsoft Excel 14.0 was used.

Analysis of KO by NMR
• Krill oil is a complex mixture of lipids • Krill oil can not be directly compared with regular Triglyceride oils
• Analytical methods needs to be adapted specifically for the KO matrix
• Regular methods for determination of oxidative status of the lipid mixture is not sensitive nor specific enough to be more than marginally useful (e.g. Anisidine or TBARS methods)
• Krill oils will always have more free fatty acids as well as water content than TG, thus are more prone to hydrolysis than TG-oils
• Methods that can distinguish between TG from KO and FO is desirable• Methods for direct (and sensitive enough) measurement of molar amount of
double bounds could be useful to investigate oxidative status and development
• Krill oil standards, both PL and TG are needed
