1 Product Description Your new Biohit mLINE is an autoclavable air displacement pipettor. The attractive and ergonomical design of the mLINE pipettor together with its very low pipetting forces reduce the risk of repetitive strain injuries (RSI). The pipettor is designed for both right- and left-handed use. The special tip cone design of the pipettor offers the possibility to use replaceable Safe-Cone filters to help prevent the contamination and damage of the pipettor. The operating button of the mLINE allows safe and easy filter ejection. The pipettor utilizes disposable tips.

2 Protective tip cone filters and filter ejection The tip cone design of the Biohit mLINE pipettors (> 10 µl) allows the use of Biohit Safe-Cone filters in the tip cones. These removable filters prevent liquids and aerosols from entering the pipettor and thus protects the pipettor from contamination and damage. The Safe-Cone filters are available as Standard or Plus versions. It is recommended to use the Standard filter for general applications and the Plus filter for more demanding applications such as cell culture, bacterial and virological work and molecular biology. Filters need to be changed regularly. The interval for a filter replacement is application dependant but the recommendation is to change the filter daily (after 50 – 250 pipetting cycles) and always in case of over-aspiration. The Safe-Cone filters can be removed with the unique filter ejector without touching the filters by hand. Remove the operating button cap and eject the filter by pressing the operating button down. Make sure that the filter is disposed into a suitable waste container. Clean the tip cone if needed and put the new filter in place.

3 Maintenance Biohit mLINE pipettors have been designed for easy in-house service. If the pipettor is used daily it is recommended to clean/decontaminate and check the performance of the pipettor every three months. Biohit also provides complete repair and recalibration service for your pipettor including a service report and performance certificate. Note: Please make sure that the pipettor has been decontaminated before you send it to us or your local representative. Please advise if any hazardous material has been used with your pipettor. Note: The use of the tip cone filters may prolong the interval of the service. Change the filters regularly. Note: It is recommended to always use gloves when cleaning the pipettor.

3.1 Daily cleaning of the outer surface of the pipettor Your Biohit mLINE pipettor should be checked every day for cleanliness. To clean and decontaminate the outer surface of your pipettor use Biohit Proline‚ Biocontrol (Cat.no. 724004, 5 litres) and a soft lint-free cloth. It is also possible to use ethanol (70%), isopropanol (60%) or a mild detergent as a cleaning agent. Gently clean the surface of the pipettor with moistened cloth and wipe dry. Pay special attention to the tip cone. Change the tip cone filter.

3.2 Cleaning and decontaminating the lower parts If your pipettor is in daily use it is recommended to clean/decontaminate and grease the pipettor every three months. To clean and decontaminate the lower parts of the pipettor follow these steps:

3.2.1 Single channel pipettors: disassembling, regreasing and cleaning 1. Eject the tip cone filter 2. Unscrew the tip ejector collar counter

clockwise and remove it.

3. Pipettors m10, m20, m100, m200 and m1000:

Unscrew the tip cone holder counter clockwise and carefully remove it with the tip cone.

Pipettor m5000: Unscrew the tip cone cylinder counter clockwise and remove it.

4. Remove the piston (if needed to replace, otherwise it is not

required to remove it during basic maintenance). Turn the pipettor upside down, push it against a table, hold the white collar with one hand and unscrew the piston counter clockwise with another hand.

5. Clean the tip ejector collar, the tip cone holder, the tip cone (cylinder) and the piston

with Biohit Proline Biocontrol, ethanol (70%), isopropanol (60%) or mild detergent and soft lint-free cloth.

6. Clean the interior of the tip ejector collar and the tip cone (cylinder) with a cotton

swab. Be careful with the pipettors m10, m20 and m100 so that the seal inside the tip cone will not damage.

7. Rinse the parts with distilled water if needed and let the parts dry. 8. Pipettors m10, m20, and m100: Put a thin layer of grease on the piston:

Pipettors m200 and 1000: Put a thin layer of grease around the seal:

Note: Avoid excess grease. Use only the grease provided with the pipettor (731140, 2g tube). Note: The spare part code for the grease is 731141 (tube 20g). The 731140 (2g tube) provided along with the pipettor package is not available as a spare part. Note: Before reassembling check that no lint or particles are on the surface of the piston. Decontamination: For complete decontamination of the lower parts place the tip ejector collar, tip cone holder, tip cone and tip cone cylinder (only the model m5000) into a beaker containing Biohit Proline‚ Biocontrol and leave for at least 30 minutes. Wipe the piston with Biohit Proline‚ Biocontrol and the lint-free cloth. Rinse the parts with distilled or sterile water. Let the parts dry. Grease the piston and seal according to the instructions given earlier in this chapter.

3.2.2 Single channel pipettors: reassembling 1. Reinstall the piston (if it has been removed, removal of the piston

is not necessarily needed during normal maintenance). Turn the pipettor upside down, push it against a table, hold the white collar with one hand and screw the piston clockwise with another hand.

2. Pipettors m10, m20, m100, m200, m1000:

Carefully place the tip cone on the piston and attach it by screwing the tip cone holder clockwise.

Pipettor m5000: Carefully place the tip cone cylinder on the piston and screw clockwise. Make sure that the tip cone (cylinder) is properly tightened. Avoid over tightening.

3. Attach the tip ejector collar by screwing it clockwise. 4. Place the new tip cone filter on its place with forceps

5. Press the operating button several times to ensure that the grease has spread

evenly. 6. Check the performance of the pipettor. Note: It is always necessary to check the performance of the pipettor after in-house service or maintenance.

3.2.3 Cleaning stop surfaces If there is a problem with the accuracy of the pipettor or the pipettor is very dirty, the cleaning of the critical stop surfaces might be required.

1. Remove tip ejector collar, tip cone holder, tip cone and piston as described above 2. Turn the volume to the maximum. 3. Insert a 1000ul tip into the machinery: push upwards and simultaneously turn

clockwise. The gray plastic part is connected to the machinery with a bayonet type of connection. Piston spring comes out with a metallic sleeve that acts as a stop surface.

4. Clean the stop surface thoroughly with ethanol and apply some compressed air inside the machinery.

5. Insert the piston spring and metallic sleeve into the machinery with the help of a tip. Push upwards and turn counter clockwise.

6. Reassembly the rest of the parts as described above.

3.2.4 Multichannel pipettors: disassembling, regreasing and cleaning 1. Unscrew the dispensing head by turning it counter clockwise and remove it.

2. Push down the piston support plate as far as it goes (for example with a pen) Note: It is very important that the pistons are at the lowest position during service. Otherwise the reassembly of the pistons and tip cones is tricky. 3. Push down the tip ejector bar locking piece (731145) with one hand and simultaneously hold the tip ejector bar at one end with another hand. Remove the tip ejector bar by pulling it out. 4. Attach a tip onto the tip cone. Push the tip cone upwards (it is spring loaded) and turn it counter clockwise. Pull the tip cone out of the tip cone housing

5. Pull out the piston. It is connected to the piston support plate with a magnet. Please insert the piston into the tip cone if you store them on the table. That way piston won’t get damaged and collect dirt. Remove all tip cones and pistons.

6. Clean the tip cone housing, the tip ejector bar, the tip cones and the pistons with Biohit Proline Biocontrol, ethanol (70%), isopropanol (60%) or mild detergent and soft lint-free cloth. 7. Disassembling tip cones: Models m10 and m100: Unscrew the o-ring support nut (m10, 731156) or seal support nut (m100, 726167).

Model m300: The tip cone can not be disassembled. The assemblies include the following parts:

Clean the parts with Biohit Proline Biocontrol, ethanol (70%), isopropanol (60%) or mild detergent if needed. Rinse the parts with distilled water if needed and let the parts dry 8. Regreasing. Models m10 and m100: Place the parts in an order as indicated in the pictures below. It makes regreasing and reassembly easier. Apply a thin layer of grease on the seals and pistons only.

Model m300: Apply a thin layer of grease on the sealing only.

Note: Avoid excess grease. Use only the grease provided with the pipettor (731140, 2g tube). Note: The spare part code for the grease is 731141 (tube 20g). The 731140 (2g tube) provided with the pipettor package is not available as a spare part. Note: Before reassembling check that no lint or particles are on the surface of the piston. 9. Reassembling Models m10 and m100: Tighten the o-ring support nut (m10, 731156) or seal support nut (m100, 726167). Please make sure the nut is properly tightened but avoid over tightening. Keep the piston inserted all the time.

For complete decontamination of the lower parts place the tip cone housing, tip cones, pistons and tip ejector bar into a beaker containing Biohit Proline‚ Biocontrol and leave for at least 30 minutes. Rinse the parts with distilled or sterile water. Let the parts dry. Grease the seal and piston as described above.

3.2.5 Multichannels pipettors: reassembling 1. Please make sure that the piston support plate is at its lowest

position. 2. Attach a tip on the tip cone and insert the piston assembly complete (piston+tip cone) into the tip cone housing. Push it upwards as far as it goes and turn the tip cone clockwise. Make sure that the tip cone is properly placed.

3. Reassemble the tip ejector bar. Push down the tip ejector bar locking piece (731145) with one hand and simultaneously hold the tip ejector bar with another hand and push it upwards. Make sure the snap fit will connect properly. 4.Insert tip cone filters. Attach the dispensing head to the handle and tighten it properly. Please avoid over tightening. Please make sure that the dispensing head can rotate freely.

3.3. Sterilization of the pipettor The entire mLINE pipettor can be sterilized by steam autoclaving at 121 oC, (252 oF), 1 atm for 20 minutes (please ref. instruction manual for details). Remove the tip cone filter (if fitted, Chapter 7.3). Put the pipettor into the sterilization bag and place it into the autoclave. After autoclaving the pipettor must be cooled down at least for two hours before use. It is recommended to grease the piston/seal after every 10 autoclaving cycle. It is recommended to check the performance of the pipettor after every autoclaving cycle.

4 Testing the performance and recalibration It is recommended to check the performance of your Biohit mLINE pipettors regularly (e.g. every 3 months) and always after in-house maintenance. However, the user should establish a regular testing routine for their pipettors with regard to accuracy requirements of the application, frequency of use, number of operators using the pipettor, nature of the liquid dispensed and the acceptable maximum permissible errors established by the user. (ISO 8655-1.)

4.1 Testing the performance Performance testing should take place in a draught-free room at 15 – 30 OC, constant to +/- 0.5 OC and humidity above 50%. The pipettor, tips and the test water should have stood in the test room a sufficient time (at least 2 hours) to reach equilibrium with the room conditions. Use distilled or deionised water (grade 3, ISO 3696). Use an analytical balance with a readability of 0.01 mgs. (ISO 8655-6.) If the results fall within the specifications, the pipettor is ready for use. Otherwise check both systematic and random errors and, when necessary, proceed to the recalibration procedure . Note: Systematic error (inaccuracy) is the difference between the dispensed volume and the selected test volume. Random error (imprecision) is the scatter of the dispensed volumes around the mean of the dispensed volume. (ISO 8655-1) Note: Biohit specifications are achieved in strictly controlled conditions (ISO 8655-6). The user should establish own specifications based on the field of use and the accuracy requirements placed on the pipettor (ISO8655-1). Note: This method is based on ISO 8655.

4.2 Recalibration The calibration of your mLINE pipettor has been factory checked and certified at 22 OC using grade 3 distilled water according to ISO 3696. The calibration is based on ISO 8655-6: Gravimetric test method for volumetric instruments. The pipettor’s specifications are guaranteed only with genuine Biohit tips. If you find the pipettor to be inaccurate after performance testing, please follow the instructions below: 1. Remove the lid of the calibration nut, located at the backside of the handle, with the

aid of the calibration tool. 2. Place the hexagonal head of the calibration tool into the hole of the calibration nut. 3. Turn the adjustment lock counter clockwise to decrease and clockwise to increase

the volume. 4. Repeat testing the performance procedure. Continue until the results are correct. Note: Biohit offers accredited calibration service. Please contact your local Biohit representative for further information. Note: Please do not use the plastic mLine calibration tool provided with the pipettor package for calibration purposes in technical service. The plastic tool has not been designed for continuous professional use. Please use a standard metallic tool.

5 Disassembling and reassembling the handle The upper part of the pipettor (handle) must be disassembled if, for example, there is a problem with the counter or any other inner part. The disassembly of the upper part is not a normal service routine!

5.1 Disassembling single channel models m10, m20, m100, m200 and m1000: 1) Remove the tip ejector collar, tip cone holder and tip cone 2) Next remove the tip ejector and handle collar by pushing the tip ejector upwards with your thumb. 3) Turn the pipettor upside down, push it against a table, hold the white collar with one hand and unscrew the piston counter clockwise with another hand. 4) Now the following parts have been removed:

5) Remove spring lock, primary and secondary springs and tip cone locking ring with the mLine tool (726201). Push the tool upwards and simultaneously turn counter clockwise. The spring lock is locked with a bayonet type of connection.

6) Pull out the machinery assembly (counter and operating button). Place your fore finger into the handle. Remove the display by pushing it with the fore finger.

5.2 Reassembling single channel models m10, m20, m100, m200 and m1000

1) Please note that there are two small slots on the machinery body and two small ribs on the calibration wheel inside the handle (please see the pics below).

2) Insert the machinery into the handle

3) Make sure that the two ribs on the calibration wheel go into the slots on the machinery body. 4) The machinery has been reassembled in a right way when the lines of the upper most counter wheel are visible

5) Reassemble the tip cone locking ring, primary spring, secondary spring and spring lock in a right order. Lock the spring lock with the mLine tool by pushing the spring lock upwards and simultaneously turning it clockwise. Please place your thumb on the counter to keep the machinery body still. Otherwise it may slip out of the handle.

Note! Please make sure that the counter lines are clearly visible. 6) Reassemble the window and counter release nut; the upper snap fit must go first. If the window is not sitting properly, then the calibration wheel might not be sitting well. In that case push the wheel back into its groove. Replace the display sticker (used for attaching the display), if needed. 7) Reassemble the tip ejector and handle cover. 5.4 Reassembling model m5000

5) Reassemble the tip cone locking ring, primary spring, secondary spring and spring lock in a right order. Lock the spring lock with the mLIne tool by pushing the spring lock upwards and simultaneously turning it clockwise. Please place your thumb on the counter to keep the machinery body still. Otherwise it may slip out of the handle.

6) Reassemble the tip cone locking nut by screwing it counter clockwise by hand until it “clicks”

7) Reassemble the window and counter release nut; the upper snap fit must go first. If the window is not sitting properly, then the calibration wheel might not be sitting well. In that case push the wheel back into its groove. Replace the display sticker (used for attaching the display), if needed.

8) Reassemble the tip ejector and handle cover. Screw the piston

9) Reassemble the tip ejector collar and tip cone. 10) Make sure the operating button and counter works properly

5.5 Disassembling MCP-models 1) Remove dispensing head 2) Next remove the tip ejector and handle collar by pushing the tip ejector upwards with your thumb.

3) Turn the pipettor upside down, push it against a table, hold the white collar with one hand and unscrew the magnet holder (726166) counter clockwise with another hand. Note: Do not hold the white part with forceps; it might damage the part! 4) Unscrew the locking nut (726165) with the mLine MCP tool (726204). Insert the tool and turn counter clockwise. Remove all parts and pull out the machinery

5) Place your fore finger into the handle. Remove the display by pushing it with the fore finger.

7) Clean, repair and replace parts if needed. Clean the piston spring (726071) and stop surface (metallic sleeve) thoroughly with ethanol and apply some compressed air inside the machinery (ref chapter 3.2.3).

5. Please place your thumb on the counter to keep the machinery body still. Otherwise it may slip out of the handle. Insert the piston spring (726071).

6. Insert the tip ejection adapter (726163) so that the flat end goes first. Then insert tip ejection collar (726163) and connecting collar (726164) 7. Insert the locking nut (726165) and tighten it with the mLine MCP tool (726204). Push down the spring loaded locking nut and simultaneously turn the tool clockwise. Please make sure the locking nut is properly tightened but avoid over tightening. Make sure that the connecting collar can be rotated freely. If not, then the nut is over tightened.

8. Reassemble the window and counter release nut; the upper snap fit must go first. If the window is not sitting properly, then the calibration wheel might not be sitting well. In that case push the wheel back into its groove. Replace the display sticker (used for attaching the display), if needed.
