AbstractStrokeisthethirdleadingcauseofdeathinindustrializedcountriesandthemostimportantcauseofacquiredadultdisability.Manyevidencessuggestthatinflammationaccountsfortheprogressionofcerebralischemicinjury.Arctigenin,aphenylpropanoiddibenzylbutyrolactoneligninisolatedfromcertainplants,hasshownantiinflammatoryactivityagainstdiabetesandAlzheimer'sdisease.Inthisstudy,wetestedwhetherarctigenincanprotectmiddlecerebralarteryoccluded(MCAO)rats.MaleSpragueDawleyratswerepretreatedwitharctigeninorvehiclefor7dbeforebeingsubjectedtotransientocclusionofmiddlecerebralarteryandreperfusion.Ratswereevaluatedat24hafterMCAOforneurologicaldeficitscoring.Furthermore,themechanismoftheantiinflammatoryeffectofarctigeninwasinvestigatedwithafocusoninflammatorycells,proinflammatorycytokines,andtranscriptionalfactors.Arctigeninsignificantlyreducedcerebralinfarctionandimprovedneurologicaloutcome.Arctigeninsuppressedtheactivationofmicrogliaanddecreasedtheexpressionofinterleukin(IL)1andtumornecrosisfactor(TNF).Theseresultsrevealedthatarctigeninhasapromisingtherapeuticeffectinischemicstroketreatmentthroughanantiinflammatorymechanism.
