What does this software release contain?
The ACQUITY UPLC Systems January 2010 Driver pack DVD contains software for both the ACQUITY UPLC H-Class system and the ACQUITY UPLC System.
This DVD also contains the ACQUITY UPLC Columns Calculator for methods transfer and the required software change note (SCN) 733 for the MassLynx 4.1 data application.
The ACQUITY Systems January 2010 DVD includes all current instrument drivers for ACQUITY UPLC and ACQUITY UPLC H-Class instrument modules. The H-Class modules are version 1.45, while the remainder of the ACQUITY UPLC modules remain at version 1.40.
Note: This release has only been tested for an ACQUITY UPLC H-Class System configuration, consisting of:

During installation of the H-Class modules, an upgrade of ACQUITY Console dynamic-link library (.dll) files occurs. The installation tool on this DVD performs the necessary upgrade on the target workstation to preserve compatibility between the version 1.40 modules and the version 1.45 .dll files.
โข If you need to install any additional instrument drivers later, for example to add driver version 1.40 for the ACQUITY PDA Detector, use the January 2010 DVD to do so.
โข If an earlier ACQUITY UPLC CD or DVD has been used to install a pre-version 1.45 instrument driver, use the January 2010 DVD to reinstall at least one of the required drivers, to reconcile any incompatibility that may have been introduced.

Installation of this software release is mandatory for all ACQUITY UPLC H-Class systems.
Installation of v1.40 included with this software release is optional for standard ACQUITY UPLC systems that have previous versions installed.

Supported Waters software for ACQUITY UPLC H-Class systems
This release supports MassLynxโข, Empowerโข 1154 (English and Japanese), and Empower 2 data system software (English, Chinese, Japanese, and Korean language versions).
This release supports the following Waters applications:
โข MassLynx 4.1 SCN 714 with SCN 733 (required for ACQUITY UPLC H-Class systems)
โข Empower 1154: ACQUITY service pack (SP) to Service Pack M
โข Empower 2: base release to Service Pack G
Note: If an SQ, TQ, or 3100 detector is part of the system, at least Empower 2 Feature Release 4 needs to be installed and the Empower 2 System Suitability option is required.
Limitation: The optional local console controller (LCC) is not supported in any system with a mass spectrometer.

Important: If you want to run the ACQUITY UPLC Systems January 2010 driver pack release with any versions of ACQUITY UPLC prior to v1.40, contact Waters Technical Service.
To install or upgrade the ACQUITY UPLC Systems January 2010 driver pack

1. Exit all applications and restart the computer.
2. Run Setup.exe on the DVD.
3. On the Welcome page of the setup wizard, select Install / Upgrade, and then click Next.
Result: The Deployment Manager - Option page for your system appears.
Tip: For a MassLynx-controlled ACQUITY UPLC system, the SQ and TQ detectors do not appear in the deployment manager. Instead, select the instruments during MassLynx installation.
4. Select the drivers for the instruments that you want to install, and then click Next.
5. Click Yes, and then wait for the process to finish.
6. Click Finish.
Result: The Waters Autoloader is automatically launched, and selects the instruments or devices that require firmware upgrades.
Tips: To see versions and checksums, scroll to the right. To see where on a hard drive the firmware is installed, hover the pointer over the instrument or device name.
7. Click Load Firmware. The Autoloader starts loading all selected firmware.
Tip: The Waters Autoloader simplifies installing the release. It automatically detects the presence of compatible software and installs the appropriate support for your ACQUITY configuration. By default, the installer appears in the language of your workstation settings.
For Empower installations, ACQUITY instrument support is installed in C:\Empower\Instruments.
For all other installations, ACQUITY instrument support is installed in C:\Program Files\Waters Instruments.
Result: When the status is Cycle Power, the firmware is installed.
Caution: Do not cycle power on any instruments or devices until firmware for all instruments is installed.
8. When the status of all instruments and devices is Cycle Power, restart each instrument or device.
9. When the status of all instruments and devices is Load Complete, click OK, and then restart the computer.

Installing this release in an Empower Enterprise environment
Important: If you want to run the ACQUITY UPLC Systems January 2010 driver pack release with any versions of ACQUITY UPLC prior to v1.40, contact Waters Technical Service.
On an Empower Enterprise (Client/Server) system, install the driver/firmware according to the preceding instructions. You must do so on every computer (LAC/Eโข32 module, client, and Citrix application server) that interacts with the instrument or device. For consistency, install the driver/firmware on all clients, LAC/E32 modules, and Citrix application servers. You need not install the driver/firmware on the database server unless Client software is loaded on the server to interact with the instrument or device.
In workgroup/enterprise environments, MS acquisition is supported via acquisition clients only. LAC/E32 acquisitions are not supported.
For information on Empower Enterprise server systems or Citrix server systems, refer to the Waters Empower 2 Feature Release 5 Release Notes, Waters Empower 2 Service Pack D Release Notes, or Empower 2 Feature Release 4 Release Notes.
Troubleshooting and uninstalling If the installation fails, uninstall previous versions as follows:
1. Exit all applications and restart the computer.
2. Run Setup.exe, on the DVD.
3. Remove the previously installed instruments and devices by selecting "Remove", and then click Next.
4. Select all of the instruments and devices you want to remove, and then click Next.
5. Click Finish.
6. Restart the computer.
7. Repeat the installation procedure.
Restoring an SQ and TQ Empower 2 Project for use with IntelliStart
This section applies to SQ and TQ detectors.
Notes:
โข During installation of the instrument driver, a template project is installed to:
<Drive Letter>\Empower\Instruments\Projects
โข The installer creates a new 'Projects' subdirectory under the <Drive Letter>\Empower\Instruments file path to which it copies the supplied project.

To use the IntelliStart functionality, you must then restore the supplied project for your detector in Empower 2. This project contains the appropriate methods for the detector to run IntelliStart.
Requirements:
โข The supplied project must be used to clone all future projects.
โข The supplied instrument methods are based on a chromatographic system consisting of either the TQ Detector or SQ Detector, along with the ACQUITY Quaternary Solvent Manager and the ACQUITY Sample Manager โ FTN. If your configuration is different, the instrument method must be modified to include all modules configured in the current chromatographic system. Additional system hardware must be added to the instrument method using the Add Instrument option. Refer to the Empower online Help for information on adding instruments to a method.
โข Once the project has been restored, right-click the project and select Project Properties. From the Project Properties dialog box, uncheck the Dissolution, GPC, GPCV, and Chemical Structures options. Leave all other properties at the default.

MassLynx data application, v4.1, SCN 733 MassLynx SCN 733 is based on and requires MassLynx v4.1 SCN 714. Refer to MassLynx SCN 733 release notes on this DVD.

Known Issues in this Release The following table contains a list of the known issues and workarounds (where applicable) for this release. The PCS (Product Change System) numbers identify issues that Waters personnel monitor within a system change request tracking tool.

Caution: When installing or removing a column, be sure that you turn the column itself and not the fitting. If the fitting is turned, the collet could get stuck and cause the active pre-heater tubing to be turned with the fitting (compression screw and ferrule), potentially damaging the active pre-heater assembly.
Precautions for sample vials and plates
The sample manager-FTN is compatible with the ANSI standard well-plates, vial-trays, vials, and cap-mats/seals that are approved for use with the ACQUITY UPLC H-Class system.
Recommendations:
โข Use pre-slit cap-mats/seals and vial caps.
โข Use foil covers on vial plates whenever possible.

Cautions:
โข Use of non-pre-slit cap mats and vial caps can cause clogging in the wash lines.
โข Vial holders must conform to ANSI/SBS standards.
โข To prevent sample spillage or needle damage, use only Waters-approved covers on the sample vials.
โข Do not put plates in a centrifuge.
โข Plates containing samples with high concentrations of organic solvent can give inconsistent results at room temperature.
Choosing needle-height settings for vials and sample containers
The default setting for needle penetration depth does not allow you to reach the bottom of the vial.
Caution: To avoid damaging the needle, follow the guidelines in this section, and use the appropriate needle-height setting for your sample plates or vials.
You can change the default needle placement (needle height) setting in the software in two places: Dilution tab of the Sample Manager instrument method editor and Advanced Settings dialog box. Set consistent values in both places.
To aspirate more sample from the vial, decrease the value, so the needle tip is closer to the bottom of the vial.
Notes:
โข The default needle-height setting for 48-vial plates is "automatic", which is set at 4.0 mm.
โข The default needle-height setting for all other plates is 2.0 mm.

Recovering maximum sample from vials
The current ANSI plate (48 vials) definition for the 2-mL max. recovery vials can leave some sample in the vial. If you want to recover the maximum amount of sample, change the needle placement setting.
Tip: To modify needle placement for vials, click Instrument Method Editor > ACQ-FTN > General tab > Advanced, and change the "Needle Placement (from bottom)" value.

Revising a plate type
The 'ANSI-48Tube0.65mLHolder' plate type is defined in Empower so that the needle does not use sample near the bottom of the vial, greatly increasing the residual volume. The plate type contains 0.65 mL tubes.
To create a new plate type and correct the depth value
1. In Empower, open the Configure System window.
2. From the Empower Configuration tree, select Plate Types.
3. Select the plate type "ANSI-48Tube0.65mLHolder".
4. From the menu, select File > Properties.
5. Under Plate Type, type a suitable name for the plate.
6. Change the Depth parameter to 28.5.
7. Click OK.
Result: A copy of the plate definition is created with a new name and revised needle depth.

Loading plates or vial holders on the sample tray
Caution: To avoid damaging the sample needle, the sample plates must be positioned correctly and the sample tray must be fully engaged.
After pulling the sample tray out, load the sample plate so that well position A,1 is at the rear, left-hand corner and the forward edge of the plate is behind the spring inside the front of the sample tray.
Slide the tray in until it clicks into place.
Ensure the plate is sitting in full contact (flush) on the bottom of the tray.

Column heater
Caution: When installing or removing a column, be sure that you turn the column itself and not the fitting. If the fitting is turned, the collet could get stuck and cause the active pre-heater tubing to be turned with the fitting (compression screw and ferrule), potentially damaging the active pre-heater assembly.

To avoid inconsistent results due to temperature fluctuations, ensure the column heater's door latch is completely closed.
If an active column preheater is installed in the CH-A, in the console system tree, click Sample Manager FTN > Configure > Preheater, and then select Active > OK.
Alternatives:
โข If an active preheater is installed but has a problem, select Active, Disabled.
The column heater's door sensor operates as follows.
โข If you are controlling the column temperature and/or preheater to a set point, a column heater door alarm is raised if the heater's door is opened for over one (1) minute during a chromatographic run. The run stops and a "Column heater door open while running" message appears. Close the door, and restart the run.
โข If the column temperature is off, a door sensor check is not performed and an error does not occur.

Operating the Detectors
SQ and TQ detectors
When specifying an SQD shutdown method in Empower, always add the shutdown method to a method set and specify it as the last line of the sample set. Otherwise, if you create a sample set method in Run Samples, click Run, and then specify the shutdown method in the Run Samples dialog box, the flow path never switches to waste when the shutdown method runs. (The SQ is put into standby and the gas flow is turned off, irrespective of whether the sample set is interrupted or goes to completion.) This could lead to flooding of the source.
Example:
Create a shutdown method that puts the SQD in standby, turns off gas flow in the Shutdown tab, and specify an event that switches the flow path to waste in the Events tab. Add the shutdown method to a method set and specify it as the last line of the sample set. The flow path switches as expected, the SQD goes into standby, and gas flow switches off.
ACQUITY UPLC System Qualification
With ACQUITY UPLC Systems January 2010 Driver Pack
As with any software installation or removal in a GxP environment, Waters recommends that you follow your organization's standard operating procedures for managing software changes. We also recommend that you run the Verify Files utility or the ConnectionsSQT for Empower Installation Qualification (IQ), and review the resulting file for an entry that states, "No installation changes were detected".
For MassLynx, we recommend that you perform the IQ from the MassLynx IQ qualification workbook and ensure the IQ Checker program displays PASS for each file.
To verify that the database table changes were completed successfully, install and launch the ACQUITY UPLC Console software. If no errors occur, the update was performed properly.
To verify that installation of the ACQUITY UPLC January 2010 release did not affect any data, reprocess a data set before and after you install the software, and then verify that the results are equivalent.
Tips:
โข Conduct the qualification tests with the columns and column heaters for which the tests were designed.
โข For Empower software installation, run the Verify Files utility, and review the resulting file (File Verification Results) for an entry that indicates which service packs are installed. The installation log states, "Installed Empower ACQUITY UPLC Service Pack on mm-dd-yyy hh:mm:ss".
โข Installation of the ACQUITY UPLC Columns Calculator does not affect file verification.

Requalification guidelines
Customers must follow best practice under GMP, GLP, GCP, etc., and comply with their own standard operating procedures (SOPs). Requalifying the software and system when adding this ACQUITY UPLC Systems January 2010 release is not covered under the customer's Total Assurance Plan with Qualification unless it is time for yearly requalification under the plan, or if loading the release is required for operation of a new module or system. For situations not covered under plan, you may contract Waters to perform the qualification at additional charge.
