EFECTO DEL pH• Principio de Le Chatelier : HA H+ + A-
• pH afecta el equilibrio de ácidos débiles (HA) o bases débiles(B)• pH no afecta a neutros o ácidos fuertes• pH puede afectar a la muestra y al empaque
1. FASE NORMAL – Debe ser usada para moléculas neutras y semi-polares; pH no es un factor relevante.
2. FASE REVERSA – Si aumenta el pH, HA más ionizado, tR más largos, coleo. Si aumenta el pH, B menos ionizado, tR más cortos, picos más simétricos.
3. INTERCAMBIO IÓNICO – El pH es un factor crítico.Recuerde – Si el pH afecta tR, se trata de un HA o B.

¿SE PUEDE ANALIZAR POR HPLC?1. Si es soluble en hexano, pruebe una columna corta y rápida,
5 o 10 cm de silica gel, 2 ml/min acetonitrilo. Si salen picos de la columna baje la fuerza del solvente hasta k’ = 5.
2. Si es soluble en metanol, trate con fases químicamente unidas, bien sea fase normal o reversa.
3. Si es soluble en agua:no-iónico: 5 o 10 cm RP-18, 2 ml/min MeOH (o acetonitrilo).iónico: pruebe ambas SCX y SAX
4. Dependiendo de lo que se busque con el análisis: Pruebe SEC.Seleccione el tamaño de poro basado en PM.
4. Si no salen picos después de 1-4 minutos, no será fácil de analizar por HPLCPuede haber problemas de detección.

SI SÍ, ¿CUÁL DETECTOR?1. UV/VIS (PDA) primera elección.
Obtenga un espectro para definir max.UV disponible, costo moderado, uso ~ 80%No es universal, pero provee buena sensibilidad.
2. IR segunda elección. Universal, detecta todos los compuestos, no es sensible (± 10 ppm). Estándar en GPC.
3. LC/MS, mejor elección.Sensible e universal, muy caro, no muy común.
4. Los detectores de fluorescencia y electroquímico ambos son sensibles y selectivos: los recomendables si se sabe que responden a los analitos.

VARIOS
1. La muestra debe ser soluble en la fase móvil, soluciones diluidas, volúmenes mayores si es necesario.
2. SIEMPRE filtre la muestra y los bufferes.
3. Con temperaturas bajas en la columna (40° C) por lo general N , , R son difíciles de predecir.
4. pH afecta a la muestra (si HA o B) y a la fase estacionaria
