Complete digestion with various restriction enzymes
Restriction analysis of pUC18 derivative (3.65 kbp, A), pBluescript KS+ (2.96 kbp, B), pcDNA3.1/His B/lacZ* (8.6 kbp, C) and pCMVβ**(7.2 kbp, D). Plasmid DNA was purified using NucleoSpin® Robot-96 Plasmid Kit on MultiPROBE II (Packard BioScience) from 1.2 ml LB overnight grown cultures of E. coli DH 5α (T. Lins, University of Duesseldorf, Department of Botany III, Duesseldorf, Germany). Cells were grown in a 96-well flat-bottom block with shaking at 300 rpm. 10/85 µl of eluted plasmid DNA were used for digestion with the indicated enzymes (1-5 units) each, and were analyzed on a 1% TAE agarose gel. APPLICATION DATA Reproducible yields of high-quality total RNA
Agarose gel analysis. HeLa cells (5 x 105) from a homogeneous cell culture were pelleted in each well of a 96-well cell-culture plate and total RNA was isolated using NucleoSpin® Robot-96 RNA on Genesis 150 (Tecan). 20 µl of each eluate (100 µl elution volume) + 5 µl 5x sample buffer were loaded and analyzed on a 1.2 % formaldehyde agarose gel.
Removal of genomic DNA contamination
PCR detection of genomic DNA (362 bp fragment of GAPDH). 2 µl of each eluate (80 µl elution volume, total RNA preparation from 5 x 105 HeLa cells) was amplified with "LightCycler - DNA Amplifi- cation Kit Hybridization Probes" (Roche) (0.5 µmol GAPDH primer, 0.15 µM LC-Red 640 hybridization probes, 50 cycles). + DNase: total RNA preparation with DNase treatment - DNase: total RNA preparation without DNase treatment Standard: 24 pg, 240 pg, 2400 pg genomic DNA purified from human blood by NucleoSpin® Blood L was used for PCR detection. Negative control: 2 µl water was used for PCR detection

Detection of gene-specific RNA by Northern Blotting
HeLa cells (5 x 105) from a homogeneous cell culture were pelleted in each well of a 96-well cell-culture plate and total RNA was isolated using NucleoSpin® Robot-96 RNA on Genesis 150 (Tecan). 15 µl of each eluate (100 µl elution volume) were used for a 1% denaturing agarose gel.
