By matching the column attributes with the appropriate system characteristics, the chromatographic results (Resolution [Rs] and Average Peak Capacity [PCAve ] ) for this mixture of steroids remain constant, but the overall run times and cost per analysis decreases as the method is scaled from HPLC to UPLC.

DISPERSION MATTERS: Rethinking LC separation categories

Dispersion – n. Broadening of an analyte band due to both on-column effects (diffusion and mass transfer kinetics which are both dependent on particle size and linear velocity) and system effects (tubing internal diameter [I.D.] and length, connections, detector flow cell volumes, etc.).

True separation performance is governed by the system dispersion, paired with a flow rate or linear velocity that yields the highest possible efficiency for a given analytical column. As the system dispersion increases, the column volume must be proportionately increased, or a loss in chromatographic resolution will occur. Systems can be categorized based upon the column attributes that are best suited to match their dispersion characteristics.
