• Centrifugation of the cell culture and resuspension in a liquid containing calcium chloride

• Addition of the engineered pGLO plasmid

• Incubate on ice

• Heat shock at 42°C

• Return to ice

• Recovery in liquid LB

• Plate on selective medium
