●Users are recommended to carefully read the content of this manual before operating the TaKaRa PCR
Thermal Cycler Dice system. ●After familiarizing themselves with the information in this manual, users are recommended to store it in
secure and convenient location from which it is always available for reference
Introduction
All Takara scientific instruments are sold strictly for use in research applications and should not be
used by those who are not experts in this field.
Due to ongoing improvements, product specifications and the external appearance are subject to

(2)General Precautions Regarding Takara Scientific Instruments
It is important to observe the various precautions described below to ensure optimum system
performance and prevent accidents before they happen. Ensure safe and correct system operation
by carefully reading the precautions.
【Precautions for Installation Location】
Observe the following precautions in order to prevent fires and electrical shocks.
● Do not install the system on an unstable or sloped foundation or any
other location subject unstable conditions.
● If the system tips over or falls, immediately turn off power to the
system and all connected devices and disconnect the power supply
cables.
● Install the system away from direct sunlight or rainwater.
● Avoid blocking the paths to any air vents included on the product.
Blocked air vents can lead to internal overheating that may result in
physical burns or fire.
● Avoid locations subject to corrosive or flammable gases.
● Avoid installing the system in locations subject to dust or strong
vibrations.
● Avoid installing in locations subject to direct sunlight and wide
temperature fluctuations as this may result in internal overheating and
may cause physical burns or fire.
● Do not place objects on top of the system.
● Do not install the system in locations other than research institutions or
related facilities.
【Electrical Precautions】
Observe the following precautions when using electrically powered scientific instruments in order to
prevent electric shocks and fires.
● Avoid operating system with power sources that do not conform to
system power supply specifications.
● Avoid sharply twisting, altering, marking or otherwise damaging the
power supply cable. Avoid applying stress to, heating or placing
objects on the power cable.
● Never use a multiple outlet extension plug.
● Always ground the system to prevent electric shocks due to leakage
current.

● Be sure to compare system power supply requirements with the rated
power capacity of the power source to ensure system current
requirements do not exceed power socket capacity. Confirm that the
total power requirements of all connected equipment do not exceed
the rated power supply capacity of the common power source.
● The system body and power supply cable are subject to high voltage
levels(The maximum voltage is a line voltage AC100V~240V). Avoid
inserting metal or flammable objects inside the system or handling it
with wet hands, as this can result in electric shock and/or other
dangers.
● When moving the system, be sure to first disconnect the power cable
as well as any peripheral devices.
● Be sure to disconnect the power cable prior to cleaning or servicing
the system.
【Other precautions】
Observe the following precautions in order to prevent electric shocks and fires.
● Avoid inserting metal objects, flammable materials or other foreign
objects into the various openings on the system case.
● Never remove the system cover or attempt to alter it in anyway.
● Avoid disassembling or making any direct contact with internal parts of
the system.
● Should noxious fumes smoke or other abnormal signs appear,
immediately terminate operation, turn off system power and
disconnect the power cable.
Wait until conditions stabilize before contacting the nearest sales
agent or the company technical support line.
● Should water or other foreign matter enter the system, immediately
terminate operation, turn off system power and disconnect the power
cable.

The main features of the Thermal Cycler Dice™
Real Time System Lite are as follows:
【 TP700/760 】
This model has the 48-well Sample Block Assembly. Use a 48 well plate, 24well
plates, 8 strips tubes and individual tubes for your situation.
The reaction block of copper with a high thermal conductivity is use. Temperature
uniformity has improved better than the reaction block of aluminum.
Original calibration processing eliminates the need for calibration using secondary
fluorescent dye. This reduces running costs and reagent preparation time.
The miniature design saves space by using the smallest width and height possible.
With an internal mirror and built-in tilt and sliding lid mechanism, the device can be
stored on a shelf, but is still easy to operate.

The CCD camera system gives no difference in measuring time between wells.
The system includes automatic compensation for stable detection, maintenance
functions and easy optical calibration functions.
Expandability is provided for by allowing installation of a maximum of four (4) types
of filter unit. Two fluorescent filter units are installed as standard (FAM and ROX).
The application software is user-friendly. Basic settings are easy, and application
settings provide a high degree of freedom. It is possible to set the fluorescent
measurement point and to view data even with other computers. The software
uses a unique data format that prevents alteration of the data by general-purpose
software.
The application software have the Absolute Quantification method, the Relative
Quantification mode and added the Plus/Minus Assay program and the SNP
Genotyping Assay program.
The application software has the List manager of Target and Sample, making text
files for RDML(Real-time PCR Data Markup Language) and changeable graphic
colors patterns.

● Chapter 2: Installation Requirements and Maintenance 1. Installation Conditions
To ensure optimum performance and long and stable product life, observe the
following precautions when selecting an installation location.
Note that installing the system in a location that does not meet the requirements
listed below can result in malfunctions and/or system breakdown. Replacement
of components and system repairs for any deterioration in performance or
damage caused by failure to adhere to the following conditions will not be
covered by warranty.
1. Install system in a comparatively dust-free location.
2. Avoid locations subject to corrosive or flammable gases.
3. Avoid locations subject to either strong shocks or continuing low levels of
vibration.
4. Install the system away from direct sunlight or rainwater.
5. Install the system as far away as possible from electrical equipment that
generates strong magnetic fields, electrical fields and high frequencies. Avoid
locations near equipment that generate noise caused by electrical motors and
transformers.
6. Choose a flat location capable of withstanding the design weight of the
system.
7. Do not put it on the slippery seat. Avoid to slip down when the earthquake.
8. The installation environment must be within 15 to 30ºC temperature range and
a 20 to 80% relative humidity range.
9. Avoid locations subject to sharp temperature fluctuations, exhaust from
air-conditioning systems and condensation caused by sharp humidity
fluctuations.
10. Avoid blocking air vents located on the right and left sides, front and rear of the
system.
11. Choose an operating location with good ventilation.
12. Do not operate the system in a cold room.
13. Select a room that can provide sufficient power (frequency, voltage and
current) for system operation.
14. Never use a multiple outlet extension plug.
15. Do not attempt to move the system while holding it by the lid handle or the
control panel, as this can damage to key components. Doing so can also
result in personal injury due to part breakage resulting in the system falling
down.

2. Opening the Packaging
This equipment comes packaged in carton, as shown below. The power cord is bundled at
the bottom of the carton box. The gross weight with packaging is 21.5 kg, and the equipment
itself weighs 18 kg. Take sufficient care when removing the equipment from the packaging.

3. Installation
Use the following procedures when installing system.
1. Confirm that the system power switch is set to off.
2. Connect the system side of the power cord to the power connector on the rear
of the system.
3. Plug the AC power plug of the cord into a power outlet.
The system is designed to operate normally with a 100 to 240 V AC, 50/60Hz
power line.
Make sure that the power outlet is designed for a 3-pronged plug with ground
line.
4. Install the Control Computer in a location near the main unit.
5. Connect the USB cable provided to the connector
on the back of the main unit, and connect the other
end to the USB port on the computer.
Note: Connect the cable to one of all computer USB ports of which the drivers
are installed.
6. After turning the main unit power ON, turn the computer power ON and start
the software. “Instrument: Connected” should be displayed at the bottom right
of the initial screen.
Caution: Connect the AC adapter as a power source for the computer. If
battery power is used alone, experiments may be cut off when
battery power runs out.
7. Please refer to the Software Manual for information about real time PCR and
analysis.
8. When experiments are complete, close the software by File -> Exit.
9. Shut down the Control Computer.
10. Turn the power source for the main unit to the OFF position.
In the interests of safety, and avoiding electric shocks, be sure to connect
the system to a properly grounded power outlet. ! CAUTION

4. Moving the System
Follow the procedures described below when moving the system to a new location after it has been installed. ● New Location Within the Same Site Follow the precautions listed below when moving the system to a new location
within the same site.
1. Thoroughly clean the system and make sure that there are no tubes or plates remaining in the sample block.
2. Avoid any shock during movement that can affect the operation of this highly sophisticated system.
3. When lifting and carrying the system, be sure to place both hands on each side of the system and lift and carry in a balanced way.
4. Make sure that the new location conforms to the installation requirements described in section “1. Installation Conditions” in Chapter 2.
5. Follow the installation procedures outlined in section “2. Installation” in Chapter 2.
● Moving to a New Site
When moving the system to a new site, clean it carefully and repack it in the packaging in which it was delivered. Note that damage sustained by the system during transport due to improper packaging will not be covered by Warranty even if the Warranty period is still valid. Follow the instructions provided in this chapter when reinstalling the system at the new location.

① Lid Handle To open the Lid, lift the Lid Handle, slide the Lid towards the
back and push it into the main unit. To close the Lid, pull the Lid
Handle forward and push it down.
② Lid The Lid prevents evaporation by heating the top of the Reaction
Plate. The Lid also allows through path of excitation light and
reflected light.
[Caution]: The equipment becomes very hot inside (108-110°C).
Do not insert fingers.
③ Sample Block Place the Reaction Plate or Tube on the Reaction Block.
[Caution]: Be careful to avoid injuries from burns. The block
heats up to a maximum of 100°C.
④ Lid Lock While the program is in operation, the Lid becomes locked so
that it cannot be opened.
⑤ Ventilation Intake (Front Side)

● Chapter 4: Explanation of Screens
1. System Startup and Shut Down
1) Startup
① Verify that the Thermal Cycler Dice™
Real Time System Lite main unit is connected
to the computer via the USB cable.
② Turn the Power Switch located at the bottom right of the Thermal Cycler Dice™
Real
Time System Lite main unit to the ON (|) position.
When the power is turned on in the main unit, The Lid begins to warm up. It takes
about 5 minutes for the Lid plate until its temperature is greater than about 100°C.
The Standby LED blinks until Lid have warmed up sufficiently. When warm-up is
complete and the equipment is ready for use, the Standby LED changes to ON.
③ After the Thermal Cycler Dice™
Real Time System Lite main unit is powered, turn
on the computer.
④ At the Login Screen, input User Name and Password if necessary.
⑤ When the computer has finished starting up, double-click the Thermal Cycler Dice™
Real Time System Software icon on the desktop.
Alternatively, the user may select [All Programs]→[TaKaRa Thermal Cycler Dice]→
[Thermal Cycler Dice Real Time System ] from the Start menu at the bottom left of
the computer screen.
⑥ The splash screen is displayed, and the Thermal Cycler Dice™
Real Time System
Software launches.

2) Shut Down
① Select Exit from the File Menu in the Thermal Cycler Dice™
Real Time System
Software.
Caution: If there are experimental files
that have not been saved, a
dialog box will be displayed
asking whether or not the files
should be saved before shutting
down. Save the experiment files
if necessary.
② Select “Turn Off Computer” from the Start menu at the bottom left of the computer
screen.
③ The Shut Down Screen is displayed.
④ Click “Turn Off”. The computer shuts down.

the OFF (○) position.

2. Initial Screen Menu
The connection status can be verified using the Instrument Status and the Camera Status displays in the Status Bar (bottom of screen). “Connected” is displayed when the computer and any units are connected, and “Disconnected” is displayed if they are not connected, or if power to the main unit is OFF.

● Chapter 5: Maintenance
1. Self-diagnostic Function
If the PCR reaction does not proceed well, it possible to determine whether the
temperature control for the main equipment is operating normally using the self-diagnostic
function for Sample Block temperature efficiency.
In addition, the status of the temperature control function can be tracked by performing
periodic self-diagnostic tests. There are 3 items in the diagnostic test:
① Rate Test
Tests the heating and cooling rates.
② Cycle Test
Tests the cycle time, overshoot and undershoot .
③ Up-Down Test
Measures the heating and cooling time required to go up 98°C and then down to
10°C.

2. Optical Calibration
Optical calibration is necessary for calculation of baseline values to ensure that the
obtained 48-well fluorescent display quantities are uniform. When the two
standard-specification wavelength filters are used, use the FAM and ROX calibration plates
which were specially designed for use with the Thermal Cycler Dice™
Real Time System
Lite. It is recommended that calibration be carried out once a year. Check for any soiling or
contamination of the Block before performing optical calibration, and clean if necessary.
Prepare the Test Reagent and the following tools and equipment:
One (1) FAM calibration plate for TP700
One (1) ROX calibration plate for TP700
Cotton swab (for cleaning the Block)
Ethanol (for cleaning the Block)
Plate Centrifuge
Powder-free gloves
1) Cleaning the Sample Block
If the Sample Block is dirty, it may affect the strength of the fluorescence. In order to
check whether the Sample Block is soiled/dirty, tighten the Lid without putting anything
on the Sample Block, and then photograph in FAM and ROX modes and verify that
fluorescence is not visible. If dirty, clean using alcohol and a cotton swab.
Verification of Sample Block Soiling
① Verify that there is no plate on the Sample Block, and close the Lid.
② Open [Instrument]→[Calibrate]→[Optical] from the Initial Screen Menu.
③ Using operation system Windows® 7,
the Use Account Control message appears.
Click “Yes” on the screen.

④ Close the initial screen menu and appear again.
In this condition, allow to access the program.

2) Optical Calibration
Preparation ① The FAM and ROX Calibration Plates each perform 3 cycles of PCR
in the Thermal Cycler Dice™
Real Time System. Melt the calibration
reagent and stir by convection.
PCR program: (95°C/5 seconds, 60°C/30 seconds) x 3 cycles.
② For the 48-well plate, perform the centrifugal process for 3 minutes at
1500 to 1800 rpm to remove Reagent and air bubbles adhering to the
well wall surface. Verify by visual inspection.
Caution: After the centrifuging process, verify that there is no liquid adhering to the
plate wall surface and that no bubbles are present.
If there is still liquid adhering to the walls or bubbles remaining, continue
the centrifugal process until the liquid drops solidly into the tip.
Caution: Wear powder-free gloves for experiments when conducting Optical
Calibration, and be careful not to touch the bottom surface of the plate or
the top surface of the seal.
When the Calibration Plate becomes dirty, Optical Calibration cannot be
performed accurately, and this may affect all test data for experiments
performed afterwards.
Procedure
① Verify that the main unit has completed warming up and the Standby LED is ON.
② Select [Instrument]→[Calibrate]→[Optical] from the Software Screen.
If appear the User Account Control message,
click “Yes”. The initial screen close and open again.
Select [Instrument]→[Calibrate]→[Optical].

③ The Optical Calibration Screen appears.
Verify that only "SearchWellCenter" is at the top
and select FAM from the drop-down menu.
④ Slide the Lid into the main unit and insert the FAM
Plate into the Sample Block.
Caution: Adjust the direction of the plate and insert
securely into the Sample Block.
⑤ Close the Lid securely and wait for 1 minute.
Caution: When the Calibration Plate sheet becomes
cloudy, this influences calibration.
Therefore, when the sheet is heated,
wait approximately 1 minute until the
cloudiness disappears.
Caution: During calibration, the Lid does not lock.
Therefore, be careful not to open the Lid
while calibration is being performed.
⑥ Click the SearchWellCenter button.
⑦ A message is displayed confirming that the Calibration Plate (FAM) is set on the
Sample Block.
Verify that the FAM plate is in place, and click OK.
The right screen is displayed when the
center of each well is detected.
Click OK. The center points of the detected wells are displayed.

⑧ A red spot is displayed in the center of each well. Verify by visual inspection that
this is the center of the Well.
Click the Save button.
Caution: If the red spot is not displayed in the center,
click the SearchWellCenter button again.
Caution: Data is not retained if Save is not selected.
Always be sure to click the Save button.
⑨ When the Save operation is complete, the screen below appears. Click the OK
button.
The diameter of the red circle becomes large, and a confirmation message
is displayed in the center.
⑩ Click the Calibration tab to proceed to Calibration.
Perform Calibration.
Verify that the fluorescent
pigment is FAM, and click
the Calibrate button.

⑪ The Calibration Verification Screen is displayed.
Verify that the Calibration Plate (FAM) is on the Sample Block and click OK.
⑫ FAM calibration starts.
⑬ When FAM calibration ends, a verification screen is displayed. Click OK.
FAM calibration is now complete.
After leaving to cool, open the Lid and remove the FAM Calibration Plates.
Caution: The Calibration Plate becomes hot due to proximity to the Lid heater.
Remove only after it has cooled off.
Next, perform ROX calibration in the same way as FAM calibration only installed
ROX filter.

⑭ Insert the ROX Calibration Plate into the Sample Block and close the Lid.
Click the SearchWellCenter tab A in the area on the left and designate the
fluorescent pigment as ROX in the drop-down menu B . Click the
SearchWellCenter button C , verify that it is the ROX plate, and click OK D
when the confirmation message is displayed.
Caution: Verify that the pigment is ROX in the Fluorescent Pigment Selection
Screen.
Caution: The Lid does not lock during calibration. Therefore, be careful not to
0pen the Lid while calibration is being performed.
⑮ The task of detecting the central position of the wells is complete. Click OK.

⑯ When the central points of the wells are displayed, verify by visual inspection that
they represent the well centers, and click the Save button.
Caution: If the Save button is not selected, data is not retained.
Always be sure to click Save .
⑰ When saving is complete, the screen below appears. Click OK.
Then click the Calibration tab to proceed to Calibration.
⑱ Select ROX as the fluorescent pigment type from the drop-down menu and click
the Calibrate button.
When the confirmation screen appears, verify that the ROX Plate is set,
and click OK.

3. Daily Inspections
Perform the following inspections and cleaning periodically in order to maintain the
operational performance of the equipment,
Recommended Once-Weekly Maintenance
Clean the Ventilation Intakes and exhaust (front/back/left/right).
Wipe clean with a cloth, or if there is significant accumulation of dust, clean with a
toothbrush.
* The main equipment, including the Sample Block, LED’s light source and
Electronic circuits, need to be cooled by air. Dirty ventilation intakes may cause
decreased functional capability and shortening of operating lifetime.
Clean the Lid and the main unit.
Wipe the outside of the equipment with a cloth soaked a little in a neutral detergent.
Then wipe with a dry cloth.
Verify the sliding action of the Lid. Wipe off any dirt or debris around the rails.
* Wiping the Lid rails with a dry cloth is sufficient. It is not necessary to lubricate.
Disinfect by wiping with a 10% bleach solution. Allow to leave for 10 minutes, then
wipe with a 70% alcohol solution in the same place. Lastly, wipe with a dry cloth to
dry the surface.
Caution: If bleach solution remains on the equipment, it may lead to corrosion.
Therefore, always wipe with alcohol after using bleach.
Recommended Once-Monthly Maintenance (or occasionally based on frequency of
use or soiling conditions):
Clean the Sample Block.
* For the cleaning method, refer to 3. Optical Calibration 1) Sample Block Cleaning
in this chapter.

⑬ Install a USB – Serial Bridge driver.
Double-click on the ATEN “cvrs9_1.0.075.exe” file inside the Driver CD.
⑭ Right-click on “Computer” icon on the
desktop ( or Start > Computer ) and select
Properties from the menu. Open the
“Device Manager”.
⑮ Under Ports (COM & LPT), set “ATEN
USB to Serial Bridge” (COMx) to COM1.
If OK, go to the item ⑲.
⑯ To change the settings, right-click on
“ATEN USB to Serial Bridge (COM x) ”
and open “Properties” from the menu.
⑰ In the Properties box, under Port
Settings, click on Advanced .
Caution: If “ATEN USB to Serial Bridge”
is not set to COM1, then change to “COM1”
as the following procedures.

⑱ Set COM Port Number to COM1
and click OK.
⑲ Close the Device Manager, and open it again to verify that the settings are correct.
Under Ports (COM & LPT) Settings, verify that the “ATEN USB to Serial Bridge
(COM1)” setting is in effect. For other devices, verify that “?” is not displayed. If a
question mark is displayed, double-click on that device, open Properties, and click
Reinstall Driver. The driver is then reinstalled.
⑳ If the computer has multiple USB ports, set up the driver according to the
procedure described above after changing the USB cable connection. Also, make
sure that the port setting is COM1.
Caution: Make sure to connect the main unit to the USB port on the computer that
has been set. If there are multiple ports and all of them are set in the same
way, then every time the computer is used, the connected USB cable will
be automatically detected. If this setting has not been performed, the Install
Shield Wizard appears and the driver settings are requested. Please set
the driver settings if they have not already been carried out.
21. Eject the CD and reboot the computer.

B.Software Install / Uninstall
If the Thermal Cycler Dice Real Time System Software is being installed for the
first time, perform the steps described in section 1 (Install).
If the software version is being upgraded, first perform the steps in section 2
(Uninstall), and then start the procedure from section 1.
1. Install
① Insert the Thermal Cycler Dice Real Time System Software CD in to the CD drive.
② Make sure the OS is Windows 7
English version. Double-click on
the English folder in the CD drive.
③ Double-click on the setup.exe file
inside the open folder to launch the
installation program.
④ In the initial screen, click the Next
button.

⑤ Click the Next button in the screen
that is displayed below.
○ The installation folder can be
changed, but please use the
default folder unless there are
special circumstances.
○ Use default (“Just me”) for the
user settings.
⑥ Click the Next button in the screen
below. The installation process
begins.
⑦ The Installation Execution screen
is displayed.
On the way, when
“User Account Control”
appear ,click on Yes .
⑧ When installation is complete, the right screen is displayed. Click “Close” button.
And then The restarts window appear, click on “Yes”.

2. Uninstall
① From the Start menu, go to Control Panel and double-click on the Programs icon.
② When the “Uninstall or Change a Program” window is displayed, select “Thermal
Cycler Dice Real Time System Software ” from the Currently Installed Programs list.
③ Click the Uninstall button.
When the confirmation dialog box is displayed, click Yes.

④ The uninstall operation starts.
On the way, when the User Account
Control dialog
appear, click on “Yes”.
⑤ When complete, a dialog box inquiring whether to restart now or later is displayed.
If additional software is to be installed, select “No”. The following screen is
displayed. After verifying that the Thermal Cycler Dice Real Time System program
has been removed, click Close. Then proceed to the installation steps detailed in
section 1.Alternatively, click Yes to restart the computer and continue settings.

C. Computer Basic Setup
Computer Settings
① In the Start menu, right-click on
Computer.
Click “Show on Desktop” to display
the Computer icon on the desktop.
This icon can then be selected when
needed for setting purposes.
② Right-click on the screen and open
“Personalize”. Under the Screen
Saver tab, set “None” for Screen
Saver and click on “Apply” .

③ Under Power Options, select “High performance” and “Change plan settings”.
If “High performance” is not appeared, click on the arrow (Show additional plans).
In the Edit Plan Settings, set as shown below and click on the Save changes.
④ In the same way, Power Options >
High performance > Change plan
settings > Change advanced
power settings. Set as follows.
Dim the display
Turn off the display all “Never”
Put the computer to sleep
Click on the Save changes.

⑤ Right-click on the Computer icon
and click Properties to enter the
System Properties window.

settings” to enter the system
properties, click Advanced tab,
then “Settings...”.
⑥ Under the Visual Effects tab,
remove the checks from the
following two boxes and click OK.

⑧ Hardware Verification
Right-click on the Computer
icon on the desktop and select
[Properties]→[Device Manager].
Verify that the following devices
are installed normally. If a
question mark is shown for an
item, right-click on that icon and
select [Properties]→ [Re-install]
to re-install the drivers.
・ Imaging devices: Lumenera
USB 2.0 Camera is normal.
・ Ports (COM & LPT) is normal
and COM 1 is selected.
・ All the USB drivers are
normal.
