THOMSON REUTERS INTEGRITY: LOG ON AND INTEGRITY HOME PAGE

Take the following steps to begin consulting the Integrity drug discovery and development portal:

Go to http://

integrity.prous.com

(or alternatively, https://integrity.prous.com for an SSL connection).


Log on by entering your User Name and Password and clicking Login.

This takes you to the Integrity Home Page.

Note: There is a Password Reminder feature for your convenience.


The Home Page provides access to: the Knowledge Areas for running searches, Quick Access links to recent and relevant content, a Quick Search utility and a series of Getting Started features.


Click Getting Started to access the FAQs which include useful definitions and to access the Learning Center which presents key features and training opportunities including Live Sessions, E-Learning and Use Cases

THOMSON REUTERS INTEGRITY: QUICK SEARCH

You can use the Quick Search feature to launch a broad search across all Knowledge Areas of Integrity.

Type a term in the Quick Search box and click Go to launch your search.


When the Quick Search results display, click any of the enabled bars to see results in that Knowledge Area, for example, to see a set of drugs related to the search term.


From the Knowledge Area results display, use Filter by Statistics to refine results or open the Options pull-down menu to select from a variety of display and data management options. Click Product List with Structures


The Product List with Structures display option facilitates review of the structure-activity relationship of compounds.

Click an Entry Number to display an individual product record.


The product record display provides general and chemical information on the product…


…a Product Summary, a Development Status Summary and links to Related Information in the other Knowledge Areas of Integrity

Click a Related Information link to display information such as...


...Pharmaco-kinetics / Metabolism values, each of which is referenced to its original source.

THOMSON REUTERS INTEGRITY: GUIDED SEARCH (SEARCH FORM)

To set up a Guided Search, go to a Knowledge Area -- Drugs & Biologics in this example.


Click the down arrow to open the pull-down of search fields.

and select one – Development Status -> Condition in this example.


Next open the Browse Index of search terms for the search field chosen, run a preliminary lookup search to locate a term in the index, and then click the term on the right to copy it to the Search Form.


Add other relevant criteria to the Search Form – in this example, select Development Status -> Under Active Development = Y – and click Start to launch the search.


From the list of active development programs for leukemia, you can retrieve the corresponding products selecting the Product List from the Options pull-down menu


Use the Filter by Statistics charts to further limit your search. From the Product List obtained, filter by Organization


Check the companies you are interested in and click View Subset(s)


Select the Full Record option from the Options pull-down menu


The product record for Imatinib mesylate shows that it acts on multiple targets, including Abl Kinase and Bcr-Abl Kinase


The product record for Nilotinib Hydrochloride Monohydrate shows that it acts selectively on Bcr-Abl Kinase. To further investigate selectivity of similar compounds a substructure search can be run.

THOMSON REUTERS INTEGRITY: STRUCTURE SEARCH

To set up a structure search, select a Knowledge Area – Drugs & Biologics in this example – and click the Structure Search button to go to the Structure Search Form.


Draw or paste a structure in the structure search window.


Combine a substructure search with a text search by opening the search field menu and selecting a field - in this example, Therapeutic Group.

Then open the corresponding Browse Index to select a term.


Use the Lookup box in the Browse Index to locate terms in the hierarchical Therapeutic Group index.

Then click a term -- Oncolytic Drugs in this example -- to copy it to the Search Form.


Click Start or press Return to launch a search for products with this Therapeutic Group classification and that share a similar substructure.


Compounds can be classified quickly and easily by using Filter by Statistics. In this example, select to filter by Molecular Mechanisms.


The chart shows that many of the compounds have more than one mechanism of action.

Using the checkbox function, select Bcr-Abl Kinase Inhibitors and click View Subset(s) to view the subset of compounds with this mechanism.


From the original list of product records retrieved that are Oncolytic Drugs and that have a similar structure, these are the compounds that are Bcr-Abl kinase inhibitors. Select the Product List with Structures from the pull-down menu


From this list, structure-activity relationships can be examined. Access to Experimental Pharmacology, Pharmacokinetics/ Metabolism and other data can be obtained through the All Related Information via Quick Search

For example, select the Targets & Pathways enabled bar


From the Targets & Pathways list obtained, select the Targetscapes option


Click on the Acute Myeloid Leukemia Targescape…


… to view the molecular landscape for this condition.

THOMSON REUTERS INTEGRITY: INFORMATION RETRIEVAL VIA DISEASE BRIEFINGS

Click Disease Briefings from the Home Page or select it from the pull-down menu of Knowledge Areas in other pages of Integrity.


In the Disease Briefings Knowledge Area, click a category to display a menu of related titles and make a selection. For instance, select Leukemia


Disease Briefings present information in a text-based format accompanied by multimedia images.

The Table of Contents appears on the left side of the screen. Click any block (Morbidity and Mortality in the example) to move to that chapter.


Chapters with arrows have sections. In the Treatment chapter, click a section to display information on specific mechanisms and compounds acting via those mechanisms.


The “higher-level” sections present information on marketed compounds...


...and the “lower-level” sections on developmental compounds.

Click any compound to display the product record…
