AutosamplerThermo Scientific Accela Autosampler features temperature-controlled sampletrays (for vials or microtiter plates), injectionvalve, and an integrated column oven tomaintain sample integrity before and during analysis. The “Total TemperatureManagement” system ensures the mobilephase, sample loop, injection valve, and column remain at the same temperatureproviding maximum retention time repro-ducibility and minimizing thermal broadening.Method development is easily optimizedusing one of the multiple injection modeswhich include full loop, partial loop, and no waste. Accela High Speed LC uses Thermo Scientific Hypersil GOLD smallparticle technology to achieve sharper, narrower chromatographicpeaks with increased efficiency and throughput. Our patentedLightPipe™ technology further increases data quality by providing five

times the sensitivity of conventional photodiode array detectors forHPLC applications. The fiber optic beam shaper technology in theAccela PDA collects and focuses the transmitted light from thelightpipe, maximizing spectral resolution without light-reducing slits.

The Accela PDA Detector offers up to five times the sensitivity ofconventional photodiode array detectors. This is achieved by usingour patented LightPipe technology, which provides a 5 cm opticalpath length at a cell volume of only 10 µL for HPLC applications. Forfast UHPLC separations a flow cell of only 2 µL volume (1 cm opticalpath length) is employed to minimize undesired dispersion. The uniqueflow path maintains the integrity of the peak shape as it travelsthrough the flow cell to subsequent detectors. Along with unbeatablesensitivity, the fiber optic beam shaper in the Accela PDA offersabsorbance measurements with excellent spectral resolution andincreased throughput over the entire wavelength range of 190 – 800 nm.The pre-aligned deuterium and tungsten lamps ensure high spectralintensities for high signal-to-noise absorbance measurements, whilethe high data rate of 80 Hz at 1 nm wavelength increments providesexcellent spectral and time resolution for fast chromatography andhigh-speed HTS (High Throughput Screening) applications. TheAccela Plus ensures wavelength accuracy with an integrated filtercuvette filled with a certified holmium oxide solution for calibrationover the entire UV-Visible spectrum.

The inherent complexity of metabolic mix-tures employs chromatographic and accuratemass separation techniques to provide structural information of individual compo-nents. The broad range of chemistries oftenrequires multiple injections with different chromatographic or ionization conditionsusing long columns to achieve adequateseparation, taking hours to complete. By coupling the Accela System with the LTQ Orbitrap™ series and Exactive massspectrometers fast separations are easilyachieved on long columns providing sharperpeaks while greatly reducing run times.

Hypersil GOLD columns utilize the very latestin highly pure silica technology, along witha new novel proprietary bonding and end-capping procedure. The new bonding andend-capping delivers outstanding peakshapes for basic analyte types leading toincreased sensitivity of analysis, improvedresolution between critical pairings, andenhanced throughput. The outstanding peakshapes provided by the new bonding technology increases confidence in the stationary phase. This provides enhancedresolution and resolves impurities from theparent ion due to increased peak heights,which allows accurate qualitative and quantitative data.
