TaqMan® hPSC Scorecard™ Panel Setup Guide for the 7900HT Real-Time PCR System

Use the following step-by-step instructions to support the TaqMan® hPSC Scorecard™ Panel on your real-time PCR instrument.

Downloading the Template File

In order to run your 7900HT Fast Real-Time PCR instrument, you will need to obtain a template file from the Life

Technologies website that contains all of the necessary experimental parameters to run your instrument. To do

this:

1. Download the template file (.sdt) file (depending on the block type you are planning on using) from the

Life Technologies website at lifetechnologies.com/scorecardinstrument.

2. Save the template file (.sdt) to your computer.

How to Initiate the Run from the Template File

Initiating a Run using the 7900HT’s Sequence Detection Systems (SDS) Software You can use the template file to run an experiment on the 7900HT SDS Software:

1. Load your samples and prepare the Scorecard.

2. Open the SDS software on your instrument’s computer.

3. Log in to the SDS Software and open your template file that was saved to your computer.

4. Assign sample names by selecting your sample and assigning it a sample name.







Note: There is no need for you to modify any of the experimental parameters as this information is already

contained within the downloaded template file. In addition, if you do not wish to name your samples, your

sample names will be automatically assigned (Sample01, Sample 02, etc.).

5. Before running your experiment, you must save your experiment as an .sds file. a. .sds files are experimental files and .sdt files are template files.







6. After saving your .sds file, close the experiment and reopen the .sds file. 7. Load your plate into the 7900HT instrument. 8. Proceed to running the experiment (by clicking Start Run) and saving the data.







9. Once the run has finished, select Analysis menu > Analysis Settings.







10. Select the Automatic Ct option button.

11. Click OK to accept the analysis settings and exit.







12. Select all wells in the plate if they are not already highlighted. Select Analysis > Analyze.

13. The SDS software analyzes the run data and displays the results in the Results tab

14. IMPORTANT! Verify that the baseline and threshold were called correctly for each autocalled detector following the analysis. If a CT value was called incorrectly, it is highly recommend that you omit the well from the experiment rather than manually adjust the baseline.

15. Finally, save your experiment. You are now ready to export your data for upload into the hPSC Scorecard™ Analysis Software.

How to Export Your Data and Upload It to the hPSC Scorecard Software To export your data:

1. Click on File > Export

2. In the export window, select “Results Table,” “All Wells,” “SDS 2.3 And Above,” and “Current Table Only.” Then click “Export” to export your data in a .txt format to your desired location.

3. You are now ready to upload your data into the hPSC Scorecard™ Analysis Software for analysis.
