Preparation

Highly purified antigens are used to hyperimmunize host animals. Most ofthese antigens are isolated from plasma, serum or cell lysates, using salt frac-tionation and various chromatographic procedures (gel filtration, ion-ex-change and bioaffinity). Goats, mice, rabbits, rats, and/or sheep are injectedwith antigen suspended in Freund’s Adjuvant.

Specific Antisera - Prepared by delipidation of bleed pools andimmunoabsorption when necessary to insure specificity. Specific antisera areadjusted to standard titer, dialyzed into 0.02M sodium phosphate, 0.14M so-dium chloride, pH 7.3, with 0.05% sodium azide, followed by filtration through0.22µm filters and lyophilized.

Immunostaining Grade Antisera - Prepared by delipidation of bleed poolsand immunoabsorption when necessary. They are supplemented with 1% bo-vine serum albumin and 0.09% sodium azide. They are presented as liquid.

Forensic Antisera - Specific anti-whole serum antisera further processed byaffinity adsorption and neutralization to remove antibodies which cross-reactto other common species. These antisera are adjusted to a standard 60mg/ml, dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH7.3, supplemented with 0.05% sodium azide followed by filtration through0.22µm filters and lyophilized.

IgG Fractions - Prepared from specific antisera by ammonium sulfate precip-itation and anion exchange chromatography. IgG fractions are adjusted tostandard titer, dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride,pH 7.3, filtered and lyophilized. Preservative is added.

Fab Fragments - Prepared from specific IgG fractions by controlled digestionwith papain under partial reducing conditions. They are purified using anionexchange and bioaffinity chromatography, adjusted to standard concentra-tion, dialyzed into 0.02M sodium phosphate with 0.14M sodium chloride, pH7.3, filtered and lyophilized. Preservative is added.

F(ab´)2 Fragments - Prepared from specific IgG fractions by limited digestionwith pepsin and chromatographic purification using gel filtration and bioaffinitychromatography. These purified antibodies are adjusted to standard concen-tration, dialyzed, filtered and lyophilized. Preservative is added.

Affinity Purified Antibodies - Prepared from specific IgG fractions by posi-tive immunoadsorption onto solid phase antigens. Acidic elution is used to se-lect high affinity antibodies. The resulting antibodies are adjusted to standardtiter, dialyzed, filtered, and aseptically vialed. Preservative is added.

Monoclonal Antibodies - Supplied as ascites or tissue culture supernatantor purified monoclonal antibodies. Mouse hybridomas were produced by fu-sion of myeloma cells and spleen cells of Balb/C mice immunized with purifiedor partially purified antigen. Cells cloned from hybridomas are injected intomice to produce ascites or are grown in tissue culture to producesupernatants. Ascites and tissue culture supernatants are clarified bycentrifugation and filtration and generally contain added sodium azide andbovine serum albumin. Purified antibodies are prepared using protein A orprotein G bioaffinity chromatography and generally contain added sodiumazide and bovine serum albumin.

Specificity

Antigen Specificity - Most products are tested usingimmunoelectrophoresis for the appropriate specificity. Products are testedwith positive controls - antigen and related material with which they should re-act. Furthermore, they are tested against negative controls - material withwhich they should NOT react. Monoclonal antibodies and immunostaininggrade antisera are tested for specificity in immunoblotting, immunostaining ofcells or tissues, and/or ELISA.

Species Specificity - Products designated by “no cross species” demon-strate no cross-reactivity in immunoelectrophoresis to the IgG of the indicatedspecies. Forensic antisera demonstrate no precipitin traces in OuchterlonyDouble Diffusion or microtube precipitin assays to the whole sera of most spe-cies. Any other products must not be assumed to be cross-reactive unless in-dicated.

Potency

IgG Fractions and Specific Antisera - Demonstrate high antibody affinityand titer. Every lot is standardized to the same titer, but the standards mayvary by specificity. Most are from 2-12 mg/ml antibody protein. Antibody pro-tein is determined by measuring the immunoprecipitate produced at the anti-gen-antibody reaction equivalence point. Fab and F(ab´)2 Fragments are ad-justed to the protein concentration determined from the titer of the source IgGfractions.

Immunostaining Grade Antisera - Tested in immunohistochemistry usingindirect immunoperoxidase procedure for working dilution in the 1:50-1:100range.

Forensic Antisera - Tested for potency in Ouchterlony Double Diffusion andmicrotube precipitin assays. They show precipitin reactions with highly dilutedwhole serum, but do not show precipitin traces with the whole sera of othercommon species. Test results are supplied with each lot of product.

Affinity Purified Antibodies - Demonstrate high titer and avidity. Most arestandardized to 1.0 mg active antibody per ml determined by equiva-lence-point precipitation with purified antigen. Some may be standardized byimmunoelectrophoresis using technical grade antigens.

Monoclonal Antibodies - Selected for high avidity. Products intended forimmunohistochemistry are tested for proper working dilutions inimmunostaining of blots, cell suspensions or tissue sections. Many are testedfor ELISA titer. The recommended working dilutions for tissue staining and/orELISA are reported in product inserts.

Purity

IgG Fractions - Tested for purity at 40 mg/ml by immunoelectrophoresis us-ing anti-host species serum and anti-host species albumin. There may betraces of IgA, IgM and other gamma proteins, but no trace of albumin is pres-ent.

Fab and F(ab´)2 Fragments - Tested for purity at 20 mg/ml byimmunoelectrophoresis using anti-host species serum and anti-host IgG Fc (γspecific). They demonstrate appropriate fragment precipitation, no trace ofwhole IgG, Fc or other serum proteins.

Affinity Purified Antibodies - These are IgG without traces of albumin asdemonstrated by immunoelectrophoretic testing of antibodies at 10 mg/ml us-ing anti-host species serum and anti-host species IgG.

Purified Monoclonal Antibodies - Tested for protein purity using laser scan-ning of reducing SDS gel electrophoretic patterns. Protein purities, whichrange from 97-99%, are reported on product inserts.

Storage

All Cappel monoclonal antibodies and immunostaining grade antisera have 1year expiration dates.

Antibody products not labeled with an expiration date have a shelf life of a min-imum of five years from manufacture date assuming the product is stored atthe recommended conditions appearing on the label and accompanying in-sert. It is recommended that the final user routinely employs the appropriatecontrols to verify that the reagents perform satisfactorily for the user’s specificapplications.

Unopened, lyophilized products are stable when stored at 2-8°C or lower.Once reconstituted, they may be stored at the same temperature range forseveral weeks with preservative added, but only for a few days if preserva-tive-free. They may be aliquoted and frozen at -20°C or lower for long termstorage. Repeated freeze-thaw cycles are NOT recommended.

Unopened liquid products are stable when stored at 2-8°C. Once opened,they may be stored at 2-8°C for several weeks with preservative added, butonly a few days when preservative-free. Immunostaining grade antisera,ascites and tissue culture supernatants may be aliquoted and frozen at -20°Cor lower for long term storage. Glycerol (40% v/v) may be added to affinity pu-rified antibodies, affinity purified fragments and purified monoclonal antibod-ies to prevent freezing and the products stored at -20°C for long term storage.

Monoclonal antibodies are presumed to be equivalent in epitope specificityand affinity since they are all derived from a clone of cells. However, themonoclonal antibodies provided in ascites contain some inherent antibodiesof the host animals. Even the monoclonal antibodies purified from ascites cancontain some inherent antibodies of the hosts. Use tissue culturesupernatants or monoclonal antibodies purified from tissue culturesupernatants to avoid this potential source of background.

Autoimmune Antigens

Small nuclear RNA’s (snRNA or URNA’s) are metabolically stable RNA’swhich contain a unique cap structure at their 5°-ends. They are organized intoribonucleoprotein particles (U snRNP’s) of which at least six different catego-ries have been identified (U1-U6). There are at least 26 polypeptide constitu-ents of the major snRNP’s, U1, U2, U5 and U4/U6 from Hela cells. Nine ofthese proteins present in each of the individual snRNP’s are designated as“Sm” or “core” proteins. These include the BB’ doublet, the D1/D2/D3 tripletand the E,F and G polypeptides. snRNP’s contain some characteristic pro-teins, i.e. U1 snRNP contains at least three unique polypeptides with apparentmolecular weight of 70 kDa (U1 70 kDa), 32 kDa (U1A) and 22 kDa (U1C).Characteristic for U2 snRNP particles is the presence of proteins of 29 kDa(U2A’) and 27 kDa (U2B’’). Ro and La RNP complexes are predominantly lo-calized in the cytoplasm. La protein is a phosphoprotein with a molecularweight of 47 kDa. Ro RNP particles consist of small Y-RNA’s, the “core”polypeptides, Ro60 and Ro52, and distinct analogous proteins depending onthe origin of the cells.

Antibodies to these autoimmune antigens are characterized inimmunofluorescence and immunoblotting. Intended for use inimmunofluorescence and/or immunoblotting. All autoimmune antibodies pre-cipitate the antigenic proteins against which they are directed with the excep-tion of the anti-La antibody 4B6 and the anti-Ro antibodies 4C6 and 1D8.They are supplied as clarified, lyophilized tissue culture supernatant with 1%BSA and 0.09% sodium azide.

CytokeratinsCytokeratins are a family of intermediate filament proteins which are represented in hu-mans by at least 19 different polypeptides ranging from 40 to 68 kDa. Thesepolypeptides occur in cell type-specific combinations that vary with the state of differen-tiation. The cytokeratin expressions in epithelial tumors are usually identical or closelysimilar to the pattern in the cell of origin.Monoclonal antibodies to cytokeratins are supplied as clarified tissue culturesupernatants or ascites. Polyclonal antisera are clarified and, if necessary,immunoabsorbed to remove cross-reactivates. They are clarified with 1% bovine serumalbumin and 0.09% sodium azide.The specificity and potency of products are tested by several screening methods includ-ing immunodiffusion, ELISA, immunoblotting and/or immunohistochemistry. Lung Cancer Markers

Monoclonal antibodies for immunostaining of cytological specimens andfrozen sections fixed by Nakane procedure and microwaved, formalin-fixed,paraffin-embedded tissue sections (item number 10829 and 10831). The an-tibodies were categorized in different “clusters” of reactivity patterns duringthe First International Workshop on Small Cell Lung Cancer Antigens in Lon-don, April 1987.

Cluster 1 Antibodies: MOC-1, MOC-21, MOC-32 and MOC-52. These anti-bodies are reactive with neural tissues, endocrine tissues, and in the lungpreferentially with small cell lung cancers and carcinoids.

Cluster 2 Antibody: MOC-31. This antibody reacts with all lung carcinomasand most epithelia tissues. MOC-31 is unreactive with normal and malignantmesothelia, and therefore, it is especially useful for the detection of carcinomacells in ascites or pleural effusions.

Multidrug Resistance-Related Proteins

Exposure to one natural product drug can render tumor cells resistant to otherstructurally and functionally unrelated drugs known as Multiple Drug Resis-tance (MDR). Classical MDR results from P-glycoprotein, a protein inserted inthe plasma membrane which functions as an ATP-driven drug efflux pump.Moreover, Multiple Drug Resistance-related Protein (MRP), a 180-195 kDaglycoprotein, and the 100 kDa lung MDR-related protein (LRP) can also pro-duce MDR.

The following monoclonal antibodies to MDR proteins are intended for use inimmunostaining of cell suspensions, acetone-fixed frozen tissue sections,and formalin-fixed paraffin sections (JSB-1, MRPm6 and MRPr1) andimmunoblotting (MRPm6 and MRPr1). They are supplied as clarified cell cul-ture supernatant with 0.7-1% BSA and 0.1% sodium azide.

Prosomes are cellular particles (MW 720,000) of variable protein compositionpresent from archeobacteria to the human species often associated to mes-senger RNA, higher order protease complexes and the intermediate filamentsof the cytoskeleton. They are identical to the Multi-Catalytic Proteinase (MCP)Complexes or Proteasomes and the Low Molecular mass Protein (LMP) com-plex apparently involved in antigen presentation within the MHC-1 system.

Antibodies are characterized in immunoblotting of purified prosome antigens.They are generally cross-reactive with prosomes from eukaryotic cells of sim-ple plants to human tissue. They are intended for immunoblotting andimmunostaining of cell suspensions, frozen sections and paraffin-embeddedsections. It is supplied as clarified ascites with 0.02% sodium azide.

Prosomes

Antibodies are characterized in immunoblotting of purified prosome antigens.They are generally cross-reactive with prosomes from eukaryotic cells of sim-ple plants to human tissue.
