Tremendous strides have beenmade in eradicating infectious disease scourges such as smallpoxand polio that once killed and crippledmillions; still, about 15million deaths—or about one thirdof all deaths annually—result from infectious diseases worldwide, according to theWorld HealthOrganization. Of those, nearly half involve children under the age of 5 years, predominantly inpoorer countries. The ongoing hefty death toll, the pharmaceutical industry’s increasing interestin the research and development of vaccines, and plentiful funding from multiple sources allcombine to provide a range of opportunities for postdocs and graduate students in vaccineresearch. The field is high growth and, perhaps more important, the fruits of this work promiseto have a real impact on the health of the world’s population. By Emma Hitt

Arecent estimate of the global vaccine business by Lehman Brothers predicts that thevaccine industry will grow at a rate of 18 percent a year to $30 billion by 2011—significantly faster than the average of 4.4 percent for the pharmaceutical industryoverall. “Today we see more opportunity than ever to be gained from investing invaccine research,” says Keith Gottesdiener, vice president for vaccine and infectious

diseaseclinical research withMerck. “Thereasons for thegrowthof the industry include increasedfunding, from both governmental and private sources, as well as an increased scientificunderstanding of the immune system that will enable real breakthroughs in the field,” hesays. And increased growth translates into a stronger job market with more opportunities (see“Expansive Opportunities”).Recent blockbuster vaccines have included Gardasil and Cervarix, vaccines against certain

human papillomavirus strains that cause cervical cancer, and Prevnar, the pneumococcalconjugate vaccine. Currently, about 20 vaccines are in development for HIV and are producingmixed results. The recent phase 2 STEP trial of the HIV vaccine (V520) showed disappointingresults, but another vaccine made by GeoVax will be moving into phase 2 clinical trials inthe next few months based on promising phase 1 results. VaxInnate is testing a universal fluvaccine that would work against all strains of the disease by using a Toll-like receptor (TLR)technology platform. Numerous experimental vaccines designed to ward off infections such asJapanese encephalitis virus, herpes simplex virus, and B cell lymphoma, and even to countercocaine addiction, are under evaluation.While vaccine research is clearly a growing field, this has not always been the case, notes

Alan Shaw, CEO of VaxInnate, and the trend represents a stark contrast to the situation in the1970s and 1980s, whenmany pharmaceutical companies exited the field due to poor economicsand high risk of litigation. “A mere handful stayed in this business, but now, vaccines are seenas both economically and technologically interesting” he says. “A benefit of serving this areaof research and participating in the development of a successful vaccine is that it is immenselysatisfying. You can literally watch a disease disappear.”David Burt, who runs R&D in GlaxoSmithKline’s North American hub, started working

in the vaccine industry in 1989. “At the time, vaccines were thought of as being a low profit,low growth area but now pharmaceutical companies are viewing vaccines as a much moreprofitable area. New knowledge from the fields of immunology and microbiology has beenapplied to the development of vaccines in recent years. I think we have entered into a heydayfor vaccines,” he says.

Booster Shot for FundingIncreased funding has dramatically facilitated advances in the field within the past few years.To bring a vaccine through the R&D process to delivery can cost as much as $500 million, andthe probability that a preclinical vaccine candidate will fail to come to market is about five timeshigher than its likelihood of success, according to a World Health Organization

Since its inception in 2000, the Bill & Melinda GatesFoundation has contributed nearly $8.5 billion to global health, withHIV and tuberculosis (TB) receiving $2.29 billion and other infectiousdiseases about $1.96 billion. In the United States, the budget of theNIH’s National Institute of Allergy and Infectious Diseases (NIAID) isabout $4.5 billion for fiscal year 2008, and the WHO’s Initiative forVaccine Research budget was approximately $26.5 million for 2006-2007. “Funding and commercial support are increasing in the area ofvaccineR&D, especially because of the high investment by entities suchas the NIH and the Bill & Melinda Gates Foundation,” noted Marie-Paule Kieny, director of the WHO’s Initiative for Vaccine Research.

Global CollaborationsVaccine research presents many opportunities for those interested intraveling to the far reaches of the globe. The GAVI Alliance (formerlyknown as the Global Alliance for Vaccines and Immunization) has beenin existence since 2000 and seeks to “align public and private resourc-es in a global effort to create greater access to the benefits of immu-nization.” According to a 2008 UNICEF report, the Alliance’s financialand programmatic support to vaccinate children around the world hashelped prevent 2.9million deaths since 2000 and decreased child mor-tality to less than 10million per year for the first time ever.“There has been a complete change in the attitudes of all involved in

believing that vaccine coverage worldwide is possible and also that wecan introduceand sustain that coverage indeveloping countries,” notesRosamundLewis,with theGAVIAlliance. Lewispointsout that theyhavebeen able to make relatively new vaccines, such as the pneumococcalvaccine, sold in theWest since 2003, available to developing countries.“So the time lag between vaccine licensing and vaccine introductionin developing countries has been reduced from 20 years to less than10 years for other products [Hib and Pneumo vaccines for example].”Another example is the rotavirus vaccine, which was put on the marketin the West just last year. “We are already introducing this vaccine todeveloping countries, so we have reduced the gap to two years, whichis our aim. The other aim is to introduce vaccines that are of specialinterest to developing countries, such as a malaria vaccine,” she says.The International Vaccine Institute, established under the Vienna

Convention of 1969 with the signatures of 40 countries and theWHO, is headquartered in Seoul, South Korea. The IVI also focuseson bringing new vaccines to developing countries. According to JohnClemens, the IVI’s director-general, its immunology programs havefocused on research and supply of vaccines against diarrheal diseases,bacterial meningitis and pneumonia, dengue fever, and Japaneseencephalitis. “Our immunology program has a special interest inmucosal immunology and the development of improved clinicalimmunological tests that can be used in field evaluations in developingcountries,” he says. “A particularly noteworthy accomplishment by theIVI has been the development of a sublingually administered vaccinewhich has demonstrated efficacy in eliciting mucosal immunity againstrespiratory and genital tract infections,” he notes.

According to Clemens, the IVI has a variety of programs andopportunities ranging from laboratory internships to pre- andpostdoctoral student and fellowship opportunities. The institute alsohas shorter term training opportunities lasting a few months. The IVIhas three divisions: the Division of Translational Research focuseson the treatment and prevention of pediatric dengue, Japaneseencephalitis, respiratory pathogens, and rotavirus as well as “diseasesof the most impoverished,” including typhoid fever, cholera, andshigella; the Division of Laboratory Sciences focuses on developingnovel formulations and delivery systems for vaccines; and the TechnicalAssistance and Technology Transfer Program provides assistance inmanufacturing practices and regulation in developing countries.The Aeras Global TB Vaccine Foundation, a nonprofit product

development partnership working in collaboration with private,government, industrial, and academic institutions, focuses ondeveloping new vaccines against TB and ensuring their availability toall who need them globally. In September 2007, Aeras received a $200million grant from the Bill & Melinda Gates Foundation to accelerateTB vaccine development. “We seek to make TB vaccine regimens forinfants and adolescents available and licensed in the next seven tonine years,” according to Yasir Skeiky, chief scientific officer of Aeras.“The disease of TB is probably worse than it was decades ago in termsof the health impact of drug resistant strains and coinfection with HIV,”he says, “and drug resistance is now extensive.”According to Skeiky, Aeras’s state-of-the-art manufacturing plant in

Rockville, Maryland, can produce ~200 million doses per year of therecombinant bacille Calmette-Guerin (BCG) vaccine against TB, enoughtomeet global need. However, the BCG vaccinewas developed 86 yearsago, and TB, with increasing drug resistance, now kills more than 1.5million people each year, second only to HIV/AIDS as the world’s most

Diverse Skill SetsThe need for skilled scientists at several levels is likely to increase inthis field. “The field of vaccine research intersects with areas of publichealth, economics, politics, ethics, and patent/intellectual propertyissues. Opportunities exist for people with expertise includinginfectious diseases, epidemiology, molecular biology, immunology,and biostatistics,” says Eileen Barry, with the Center for VaccineDevelopment at the University of Maryland in Baltimore.Notably, the basic skills needed for vaccine research are taught in

most universities: immunology, molecular biology, protein purificationand analysis, epidemiology, biostatistics, biochemical engineering.“There will always be a need for people who have a strong backgroundin basic science such as biology, chemistry, bacteriology, andimmunology. Molecular biology and bioinformatics will be two of theareas in vaccine research with the strongest needs,” says GSK’s Burt.So whatever your life science training, there could be a rewarding

work toward enabling the delivery of vaccines to the people who needthem. “Students interested in this area could enter into any numberof fields in addition to immunology research, including public health,epidemiology, and health economics,” she says.

Needs and Emerging ChallengesPressing needs in vaccine research will ensure opportunities in thefield for years to come. “Obviously, we still have HIV, tuberculosis, andmalaria staring us in the face,” says VaxInnate’s Shaw. “Most peoplein the industry would say that the easy vaccines have been developedalready,” he says.“Understanding the host response will be a critical area in vaccine

research,” says George Kemble, vice president of research anddevelopment for MedImmune Vaccines, recently purchased byAstraZeneca. According to Kemble, many of today’s vaccines werecreated by relying on the natural immune response mechanisms. “Ourability to control these reactions and use them to form a response tothe vaccinewill be critical in designing the next generation of vaccines,”Kemble says.Bioterrorism is another important and well-funded area in vaccine

research. Concerns for national and international security havesupplied impetus for an influx of funding for biodefense vaccines andemerging infection vaccines in the past five years. The Centers forDisease Control and Prevention in Atlanta, Georgia, is a pioneer ofresearch on organisms such as anthrax, botulism, plague, smallpox,and viral hemorrhagic fevers.

The VRC brings together a diverse group of scientists with the goal ofadvancing vaccine-related research as well as initiating and advancingvaccine candidates through clinical trials. Crucial to the basic andpreclinical efforts are immunological evaluations in the nonhuman primate(NHP) model. In terms of basic research, the VRC is actively involved instudying innate and adaptive immune responses, lymphocyte traf�cking,the generation and maintenance of memory responses in systemicand mucosal sites, as well as pathogenesis following SIV infection. Intranslational research, the VRC is optimizing immunogenicity of a varietyof different platforms and vectors by studying the effects of adjuvants,schedules, and delivery platforms for vaccine vectors. Finally, the VRCconducts preclinical testing of vaccines in the NHP model; these data areused to support moving clinical products forward to human testing andcan be critical to regulatory �lings.

To support these efforts, the VRC has created the NHP ImmunogenicityCore (NIC). Currently, we are recruiting a Staff Scientist (Core) tolead the NIC. This individual will oversee all NIC operations, including(1) consulting on the design and implementation of NHP studies;(2) coordinating all studies, sample collection, and analysis with theVRC Laboratory of Animal Medicine, including writing and defendingAnimal Study Protocols before the Institutional ACUC; (3) developingstandard operating procedures for, and validating, cellular and humoralimmunogenicity assays; (4) implementing all testing according to GLP

The Department of Hematology at St. Jude Children's Research

Hospital is seeking to fill two faculty positions at the level of

ASSISTANT and/or ASSOCIATE MEMBER. We are specifically

laboratory-based research in Hematology. Candidates with interests in stem cell

The Hematology Department currently consists of eight faculty members actively engaged

in clinical and laboratory research in sickle cell disease, thalassemia, gene therapy for

an NHLBI-funded Comprehensive Sickle Cell Center, an ongoing NHLBI-funded Program

Director of the Health Sciences and Technology DivisionThe Massachusetts Institute of Technology (MIT) is seeking a dynamic, innovative leader to direct the Harvard-MIT Division of Health Sciences and Technology. We are looking for a distinguished researcher in a field ofscience, engineering or medicine with demonstrated accomplishments in a major leadership role. The Director isresponsible for overall Division leadership and administration, and works closely with the HST Director counterpartat Harvard Medical School (HMS). The MIT and HMS Directors of HST jointly oversee a longstanding, multi-institutional program that includes MIT, HMS, Harvard University, several Boston area teaching hospitals, and anumber of research centers that, collectively, form a unique collaboration that integrates science, medicine andengineering to solve problems in human health.

HST currently trains some 400 graduate students in the fields of science, medicine, engineering and management.A hallmark of the HST educational process is that innovation and entrepreneurship are encouraged throughside-by-side training of outstanding students with diverse backgrounds for careers in the biomedical sciences andengineering.
