Asterand’s PhaseZERO® Services team supports your human tissue-based research needs with a broad range of service offerings. From scalable translational research solutions to proven delivery of high quality data, our team is committed to providing reliable services to help you reach your next breakthrough in cancer research.
With over 15 years of contract research experience, Asterand’s dedicated PhaseZERO Service team combines state-of-the-art technologies with the highest quality human biomaterials to provide expression profiling, molecular histopathology and cell-based assay services.
SuPerior quality ServiceS StartS with high quality tiSSue
The ability to deliver reliable and reproducible results, through PhaseZERO Services, begins with ensuring that starting material is of highest quality. Asterand’s XpressBANK™ biorepository consists of hundreds of thousands of fully consented tissue from donors with a broad range of disease states (>75% cancer tissue), extensive ethnic diversity, and corresponding comprehensive clinical data. XpressBANK tissue is obtained under strict, standardized procedures and rigorous QC-tests are conducted to ensure the integrity of molecular representation. Tissue sections from tumors and normal tissue controls are available in FFPE, flash frozen, mirrored pairs, matched pairs, and tissue array formats for molecular pathology analyses. Through our extensive worldwide network of tissue suppliers, we have immediate access to available unfixed, unpreserved tissue for use in cell-based assay services.

More predictable models for drug development
The physiological relevance of conventional 2D cell culture systems has increasingly been questioned. Our PhaseZERO expert scientists recognize the value of creating 3D cell culture systems that better simulate the cellular microenvironment and signaling pathways present in diseased tissue. With isletOrganDOT™ and hydrogel technologies, our service team can use fresh tumor tissue in a 3D setting to study the effect of therapeutic agents on proliferation, apoptosis, migration/chemotaxis, angiogenesis, cell-signaling and more.

figure 1. isletorgandot cell embedding technology. Fresh human tissue is dispersed into constituent cell types, which are seeded on to a membrane. Fed basolaterally with culture medium, the cells re-aggregate into 3D structures that retain functionality. Cells can be challenged with physiological and pharmacological agents and various end-points, such as proliferation or secreted mediators, and measured.

figure 2. the concentration-dependent effect of temozolomide on glioma cells in isletorgandot 3d culture. Temozolomide exhibits anti-neoplastic activity by interfering with DNA replication on glioma tumors in vivo . The graphs show the effects of temozolomide on the proliferation of LN18 glioma cells grown in culture. Concentration-dependent cytotoxicity is only apparent in vitro when these cells are culture in 3D. Data courtesy of Capsant Neurotechnologies Ltd.
