Charles River provides Next Generation Sequencing (NGS) of prokaryotic 16S ribosomal RNA (rRNA) in the microbiota of laboratory animals. Please note, it is important to follow aseptic technique when collecting samples, as contamination of samples with bacteria from the environment could confound experimental findings. Supplies • 2.0 mL (mouse) or 5.0 mL (rat) PCR microcentrifuge tubes (Safe-lock vials). • <= -20oC freezer for interim storage • Dry ice for shipping • Insulated shipping container • Shipping label • Zip-top bags Method 1. For every sample you intend to collect, record a unique ID, the species, and the number of fecal pellets per sample vial.
2. Use the same sample ID for paired sample types from the same animal(s). 3. Create vial labels with the sample ID and collection date. 4. Charles River-provided individually wrapped PCR microcentrifuge tubes are sterile; they can be disinfected and transferred
into an isolator according to your standard procedure. Alternatively, the unwrapped PCR tubes can be sterilized by autoclaving along with other isolator supplies.
5. Following aseptic technique, collect fecal pellets directly from the animals. 6. Freeze PCR microcentrifuge tubes within an hour of collection and keep frozen until shipment, as it is critical to preserve the relative abundance of
bacteria in the sample. 7. When shipping, schedule for next-day arrival. 8. For shipment, place sample tubes and the submission form in separate zip-top bags, in a labeled insulated shipping container with dry ice. Additional considerations for collection • Because the microbiota might vary according to the time of day, samples collected on different days should be collected at the same time. • Samples whose microbiota will be compared (e.g., treated vs. untreated) should be collected within an hour of each other. • Other key variables to consider include age, sex, and strain.
