The Bentley NexGen represents the latest in state-of-the-art technology for automated milk analysis at 500samples/hour. Engineered in accordance with BentleyInstruments’ rigorous design principles, the NexGenprovides precise and accurate measurements. TheBentley NexGen comprises two separate modules—theBentley Flow Cytometer (FCM), and the Bentley FourierTransform Spectrometer (FTS). The FCM measures thesomatic cell count of the milk samples while the FTSsimultaneously measures the milk composition,including fat, protein and lactose. After first beingstirred, the milk is withdrawn from a sample vial anddelivered to both measurement modules. Thesampling, sequencing and identification of the samplevials are functions performed by the Autosampler.

Technology Executive Summary

The Bentley FTS component analyzer uses an industrialFourier Transform Spectrometer (or FTIR) that capturesthe complete infrared absorption spectrum of the milksample for component analysis. Collecting thecomplete spectrum allows the calibration to be basedon all spectral characteristics of each particularcomponent. The infrared spectrum is permanentlystored so that at any time in the future othercalibrations may be retroactively applied to your data.

The Somacount FCM is the next innovation in theevolution of our world-renowned instruments forsomatic cell counting. Based on a highly stable solid-state laser induced fluorescence, this module is capableof delivering an accurate and timely warning formastitis onset.

With the Bentley NexGen, control of both the FCMand FTS modules is integrated into a singleWindows-based software program. The simple andintuitive graphical user interface is designed for easyoperator use. The software’s sophisticated dataexport and reporting functions and network accesscapability allow lab supervisors to monitorinstrument status and analyze results remotely.

Extensive diagnostics are collected on the NexGen sothe user can be assured that the instrument wasperforming optimally when the sample data wastaken. And because a full set of diagnostic readingsis stored with each sample run, an audit trail existsto validate the state of the instrument during datacollection. Prior to sampling, the sample temperatureis read and recorded while the sample is beingstirred. An external temperature probe is alsoprovided for the user to deploy in their water bath.That temperature is logged for each sample run aswell. Other examples of the diagnostics include avariety of critical temperatures, optical systemalignment, laser power, the pump’s characteristicpressure curve, humidity level and air pressure withinthe FTS optical deck, and the presence of shock orvibration on the instrument. Should a problemdevelop with the instrument, immediate assistancefrom Bentley Instruments’ service personnel can begreatly facilitated by reviewing these instrumentdiagnostics. Bentley Instruments’ service staff canalso log onto the instrument remotely to further aidin troubleshooting.

BENTLEY NEXGEN DESCRIPTION

The NexGen incorporates the latest technology for automated milk

component and somatic cell testing.

A wide variety of data output options are available,including diskette, serial, parallel and throughnetwork connections, which can be easily configuredto be compatible with existing systems.

The NexGen exceeds the IDF 148A standard andICAR requirements for somatic cell counting andIDF 141C:2000 and ICAR requirements forcomponent measurement employing AOACapproved methodology.

Applications

Primary applications include the measurement offat, protein, lactose, solids, other components andsomatic cells.

The system is engineered for dairy laboratories thatrequire a highly reliable and accurate instrument forperforming up to 500 individual tests per hour. Themid-infrared Bentley FTS is designed with speed andthroughput in mind, and the interferometer is ahardened industrial module that still retains its rootsin a research grade spectrometer. REAL TIME MILK QUALITY ANALYSIS AND REPORTING

The NexGen presents results and diagnostic information in a clear, concise

manner. Customized analysis reports can be generated for routine output.

The raw data for milk component measurement isthe complete FTIR absorbance spectrum as shownhere. Sophisticated multivariate calibrationalgorithms are used to extract the compositionalinformation. The spectrum is permanently archivedfor future reference, and the user can export thespectra for further analysis.

Real time diagnostic information is provided foreach sample. Here the infrared spectrum and thepump pressure characteristic plots are shown. Tabselection at the top of the screen allows theoperator to easily switch between various views.

The pulse height and width histograms for theSomacount FTS shown here are used to derive the somatic cell count.

This report shows a summary of the details of aparticular sample, including the batch and sampleidentifiers, and the measured component values.

Component repeatability and stability are easilyverified using the IDF funnel plot.

Accuracy is verified using this plot showing aregression analysis of the measured vs. thereference values for a set of calibration samples.

At the heart of the Bentley FTS is a Fourier TransformSpectrometer (or FTIR) that captures the completeinfrared absorption spectrum of the milk sample forcomponent analysis. Collecting the complete spectrumallows the calibration to be based on multiple spectralcharacteristics of each particular component. Theoptical deck of the FTS is housed in a hermeticallysealed enclosure, ensuring enhanced temperaturestability and environmental robustness. The air in thesealed deck is pumped out, leaving the deck undervacuum. The combination of the hermetic seal andremoving the humid air greatly reduces the humidityload on the enclosed desiccant, virtually eliminatingthe preventative maintenance task of frequentdesiccant regeneration.

This complete infrared spectrum is permanently storedso that at any time in the future, other calibrationscould be retroactively applied to your data. The user isprovided full access to the complete spectrum; it canbe viewed in the software or the data points can beexported for use with external programs. With thisaccess to the spectrum, the user can easilyaccommodate measurements of new components asthey become important in the future. Novel calibrationdevelopment can be undertaken independently by theuser or in conjunction with Bentley Instruments.

The FTS has a temperature-regulated fluid reservoir for the dual purposes of providing for regular andautomatic zero (or background) scans for theinstrument, as well as automatically rinsing the cell and plumbing lines after running milk samples.

The FTS exceeds the IDF 141C:2000 Standard andICAR requirements for Milk Component Measurementand uses AOAC approved methodology.

BENTLEY FTS MODULE

“Future-proof” Bentley FTS offers complete milk

analysis today with the capability to incorporate

novel component calibrations tomorrow.

The Somacount FCM uses a proprietary process based on theprinciple of laser-based flow cytometry to determine thesomatic cell count in raw milk. Flow cytometry is anextremely powerful and versatile technique and is themethod of choice in the medical field for detecting,analyzing and sorting cells. The Somacount FCM is the resultof 15 years of experience and research in flow cytometry; thefirst instrument was developed in 1991.

The milk sample is first treated with a proprietary buffersolution that stains the milk somatic cells with afluorescent dye. This solution is then injected into the flowcytometer where hydrodynamic focusing ensures that thestained somatic cells intersect an intense laser beam, whichcauses the cells to emit fluorescent light. This fluorescentlight is then collected and detected. Post-analysis ofhistograms that show the heights and widths of theelectronic pulses results in the total somatic cell count. Thehistograms are recorded and archived so they may berecalled in the future in the event that further data analysismethods become available.

The FCM consists of two identical flow and optical channels.This dual-channel construction provides redundancy. Should aproblem arise in one channel, the operator can simply shutdown that side and continue to test samples at a reducedspeed of up to 300 samples/hour. A more powerful andreliable diode-pumped solid-state laser replaces the gas laserof previous generations. Laser power is monitored andrecorded for stability.
