HISTORY OF THE ALOE

Some of the most ancient civilisations worshipped the Aloe like a god. The first therapeutic use of the aloe was found on a tablet belonging to the Su-merian civilisation of about 2110 BC. Drawings of the Aloe were discovered on the walls of the temples of Ancient Egypt. Various formulas on the use of the Aloe to treat internal and external illnesses were written on the Egyptian “Ebers Papyrus” dating back to 1550 BC.

The first person to describe the uses of the Aloe was a Greek doctor, Dio-scoride (41 AD), for the treatment of serious skin diseases. The Aloe was also greatly valued during the following centuries, in the 15th century it was discovered by the Jesuits who, having a deep understanding of its properties, planted it wherever their missions took them. The western world learned of the Aloe in the 18th century through the colonisation of North America

by the settlers. The red Indians used it in rituals and as a therapy. From a scientific point of view the first chemical analysis of this plant were carried out around the middle of the 19th century. It was classified as a laxative in the “Uni-ted States Pharmacopea”in 1922. From 1930 the United States, Japan and Russia have carried out research into the Aloe. In the 50’s, especially in Russia, the Aloe Arborescens and the Aloe Vera were te-sted on many patients having serious skin diseases and gynaecological illnesses. In the 70’s many patients with various ill-nesses were treated with remedies prepa-red with Aloe Arborecens and honey.

THE ALOE ARBORESCENS OF THE BOTANIC GENDER ALOE BE-LONGS TO THE LILIACEE FAMILY

Recently these plants have been clas-sified in their own specific family which is the ALOACEE.

This is due to the fact that around 300 have been classified in the entire world. The plants have evergreen fleshy le-aves of a sword-like shape, larger at the base and prickly edges. The lea-ves of the Aloe Arborescens are nar-row and long, have a thick outer skin, inside there is a layer which contains the lymph and in the centre a gel –like substance.

The Aloe grows naturally only in warm climates and resists in long pe-riods of dryness due to its capacity to retain water.

AN IMPORTANT INGREDIENT THE HONEY OF THE RED ALOE FLOWERS

Our organic honey comes from the bees on our plantation which is in flower from December to April. Our organic agriculture can produce an organic honey using natural methods.

No type of pasteurization or re-fining treatments are used in the preparation of the honey, it crystal-lizes naturally so that the natural ingredients are not changed. The honey is the result of the tran-sformation that takes place in the nectar during the transition in the honey extractor. This transformation, activated by

the digestive secretions, makes the nectar a mixture of invert sugar (dextrose and laevulose), of sucrose and of mineral salts. The honey is collected after the flowering period of the Aloe Arborescens.To extract it from the honeycombs the opercula, which close the cells, are removed and the combs placed in the extractor. The centrifugal force of the extractor causes the honey to flow out.The empty honeycombs leave the extractor intact and so can be re-turned to the bee colony to be fil-led again with honey during the new harvest. The honey from the red Aloe flower is a gift of nature which only bees can produce. EXPERIMENTAL Plant ExtractionAloe leaves were harvested in spring of 2008. Leaves were processed by two types of extraction procedures.EXTRACT A: obtained by 24 h juice collection from excised leaf.The leaf used to analysis was tied to the apex, it has been cut crosswise near the base (approximately 1 cm) and moreover it has been carried the excision of on the leaf and the dropped juice has been collected over night. Then, it was dried to obtain a crystalline of red-tawny powder that was dark stored into a drier. EXTRACT B: Achieved by extractions with methanol.The Aloe leaf was washed and homogenized. The homogenate was mixed with methanol (1:1), and that was underwent homogeneization again. The resultant homogenate was centrifuged at 5000 rpm x 20 min, and the surnatant was con-centrated to dryness and dark stored into a drier.

ANALYTICAL INVESTIGATION ON ACTIVE PRINCIPLES IN ALOE EXTRACTS

INTRODUCTIONAloe arborescens var. Etnea is a succulent plant that belongs to the family of Liliacee. Aloe species are currently the most studied and cultivated medicinal plants. Historically, the use of leaf extracts in medical traditions of ancient Me-diterranean populations has been described. Aloe leaves are rich in many con-stituents, i.e. phenolic compounds, polyamines, saccharides and proteins. The phytocomplex possesses an interesting biological activity, in particular it is of-ten used for its antioxidant properties. The overall concentration and activity of the components of the Aloe leaves have been investigated, indicating as prin-cipal components the polyphenols. The antioxidant activity would be related to the three major constituents of Aloe leaves, i.e. the antrones barbaloin and its stereoisomer isobarbaloin (1-2), the chromones aloesin ( 3 ) and aloeresin A ( 4 ) [1, 2]. Our goal was the determination of this specific phytochemical markers in Aloe arborescences var. Etnea. We decided to compare two different extraction procedures, i.e. a classic solvent extraction and a juice collection from excised leaf. The samples were analyzed by a gradient reversed-phase HPLC method coupled to DAD detection. We developed and optimized the analytical assay obtaining a high resolution and sensitivity to determine all studied compounds.EXPERIMENTAL HPLC apparatus and chromatographic conditionsHPLC system: Perkin-Elmer Series 200 pump Detector: LC-235C Diode Array Detector (DAD)Processor: Turbochrom Workstation software, version 6.1.2.Column: 5mm Hypersil ODS RP-18 column (250x4.6 mm; Supelco, Bellefonte, PA USA) fit-ted with a guard column Hypersil ODS RP-18,10x4.6 mm; 5mm particles; Supelco)A gradient elution program using: - Solvent A - acidified water at pH 2.8;- Solvent B - acetonitrile.

The mobile phase was daily prepared and filtered through filters.Time of analysis of our method was 35 minutes.

Then continued with a linear gradient up to 100% acetonitrile – 0% water for about 10 minutes, follo-wed an isocratic step (5 minutes) to 16% acetonitrile – 84% water in 10 minutes to re-equilibrate the co-lumn, and bring it to the initial conditions before the next injection. The detection wavelength was 293nm. The DAD acquisition wa-velength was set in the range of 200-600nm. The injection volume was 208 (ɥL).

THE ELEMENTS CONTAINED IN THE ETNA ALOE ARBORESCENS

RESULTSThe developed gradient HPLC method allows simultaneous determination of the studied polyphenolic compounds of Aloe Arborescens var. Etnea leaf. The identity of the peaks was confirmed on the basis of the corresponding UV spectra (data not shown), and by the analysis of standard compounds.Chromatograms obtained from Extract A and Extract B samples. Table 1 reports the identified peaks of polyphenols compounds.Table 2 reports the results of the total polyphenols concentration using the Folin-Ciocalteau method. CONCLUSIONA new analytical method to identity four different polyphenolic compounds into leaf of Aloe Arborescens var. Etnea is described. Two different extraction procedures allowed us to observe: - Aloesin concentration obtained by both extraction procedures is comparable.- Aloeresin is present in more concentration in Extract A; - Aloin B and aloin A was shown to maintain the same concentration ratio as deter-mined in the commercial extract obtained from Aloe Barbadensis latex.

It is important to stress that this vitamin is present mostly in meat, milk and its derivatives, but is rarely found in plants. For this reason the consumption of Aloe juice can be profitable for vegetarians, for those allergic to milk and its

MONOSACCARIDES AND POLYSACCARIDES:

Monosaccharides (in particular glucose and mannose) and polysaccharides are found in the ALOE in a high percentage. As their function is of prime importance a short explanation is necessary.The long list of monosaccharides and polysaccharides contained in the Aloe plant are the elements which mainly contribute as to why so much is said about the extraordinary powers of the Aloe. They can be summarised as follows:-- CELLULOSE,GLUCOSE,MANNOSE,ALDONETOSE,- ACEMANNAN- ALOERIDINE: a powerful immune stimulant.A large quantity of ACEMANNAN is present, mucopolysaccharide which can strengthen the organism’s immune system, it is also bactericidal,germicidal, and antifungal. It is absorbed into the intestine without changing the digestive process. It’s function is to form a sort of mucus which protects the walls of the digestive organs. As the immune and the digestive systems are closely linked these elements are of fundamental importance.

ESSENTIAL AMINO ACIDS

The amino acids represent the constitutive tissues of the human organism. One must distinguish between “essential amino acids “ which are not produced natu-rally by the body and must be taken in externally, and “non essential amino acids” which the body itself produces.

The Aloe contains 7 out of the 8 amino acids classified as “essential” ISOLEUCIN, LEUCINE, PHENYLALANINEMETHIONINE, LYSINE, THREONINE and VALINE. The Aloe also contains 11 out of the 14 amino acids classified as “NON ESSEN-TIAL”. ASPARTIC ACID, GLUTAMIC ACID, ALANINE, ARGENI-NE, CYSTINE, GLYCINE, HISTIDINE, HYDROXYPROLINE, PRO-LINE, SERINE and TYROSINE.

THE BENEFICIAL PROPERTIES OF THE ALOE

Our product is made from the Etna Aloe Arborescens, cultivated in Si-cily and has a higher quantity of polysaccharides than those of other plants as, for example, the Aloe Vera.From the comparative analysis carried out the average quantity of polysaccharides at a high molecular weight of 21.000 Dalton (which is the fraction containing acemannan, mainly responsible for the results attributed to the Aloe), varies from 0.6% to 2.5%. Unlike the Aloe Vera our gel, made from Etna Aloe Arborescens, is obtained by a complete pressing of the leaves and an important result of this is that the anthra-quinones do not exceed 195pp of our product.

ADVERSE EVENTS AND CONTRAINDICATIONS

It has been scientifically experimented that the ALOE is not toxic. In a few cases some phenomena have been observed such as increased defecation, rashes and very rarely nausea and vomiting. These are due to the to the de-toxifying powers of the ALOE. If necessary the doses of ALOE should be stopped for a few days and then reassumed. In persons with multi allergies or with an established allergy to liliaceae, an allergy test should be carried out. People suffering from serious kidney diseases should refrain from ta-king the ALOE as it is very rich in mineral salts. For pregnant women or during the menstrual cycle there are no contraindications regarding taking the ALOE as it regulates the intestine and has never caused dysentery or diarrhoea. The recommended dose must be followed.

GEL AND OINTMENT 2 PRODUCTS FOR EXTERNAL USE

The therapeutic virtues of the Aloe Arborescence (superior to other plants or the ALOE family) have been recognised by numerous clinical studies published in recent international literature regarding this subject. So an ideal use has been found in the preparation of dermo cosmetics suitable in the treatment of skin problems. From ancient times the capacity of the ALOE to cure dermatological pro-blems has been recognised, even the more serious problems like wounds that won’t heal and dermatitis which resists all other treatments.
