In this article, you will reach the correct diagnosis of the bacteria that cause diseases of peri-implant. There are two major diseases: peri-implant and peri-implantitis mucosis, the latter causing bone loss caused by stress and / or bacteria.

INTRODUCTION
Studies will be used primarily for PCR and DGGE, which help determine the type of bacteria staying there (differentiated as the genetic material and its molecular weight), and further, the more bacteria associated with the disease, to offer patients this a more specific diagnosis.

PERI-IMPLANTE
A las personas con perdida dental se les ofrece una alternativa de implante. La mucosa alrededor del sitio del implante se conoce como Peri-implante, en la cual se puede dar crecimiento bacteriano por falta de higiene principalmente y también por la profundidad del sitio de implante.

La microbiota hace referencia a la flora bacteriana normal que tenemos en las diferentes partes del cuerpo, en especial las zonas húmedas.
La microbiota tiene una relación simbiótica con el hospedero, antagonismo microbiano (Lactobacilos).
Es de las mas complejas y heterogéneas.
Puede verse alterada con la llegada de bacterias oportunistas, causando biofilm.

Método para amplificar una secuencia de DNA para poder estudiarla posteriormente, se realiza antes de la DGGE.

Es un método de rastreo molecular, por el cual se desnaturaliza en DNA, pasando de una doble cadena a una sencilla por método químico.El punto de desnaturalización aumentará con el tamaño de la secuencia de nucleótidos y por los nucleótidos de la secuencia (suele ser altos en G y C).

Es en gradiente para que se de la desnaturalización de todos los fragmentos, aun los que tienen energía de Gibbs mas negativa.
Cada banda representa diferentes especies microbianas, ya que actúa como un cebador identificando los rRNA 16S bacterianos pudiéndolos diferenciar uno a uno.

La microbiota oral se puede ver afectada por el ingreso de bacterias en el área de peri-implante, dichas bacterias pueden causar estrés en exceso hasta producir perdida ósea. Por tal motivo se pide exámenes de PCR y DGGE, para identificar cada una de las bacterias involucradas, para poder ofrecer un mejor tratamiento y detectar a tiempo y posible daño a nivel óseo.

The aim of this study was to identify bacteria associated with peri-implant diseases using Denaturing Gradient Gel Electrophoresis (DGGE) as a method for microbio- logical assessment.

MUESTREO: Todos los 22 sujetos dieron su
consentimiento informado, con la aprobación del Comité de Ética (FCE) de la Facultad de Medicina y Odontología de la Universidad de Bristol
Ninguno de los sujetos que participaron en el estudio habían recibido tratamiento para la enfermedad peri-implante.

Ninguno de los sujetos tenía una condición médica que requiere el uso de antibióticos locales, antibióticos sistémicos o fármacos anti-inflamatorios, o de agentes anti-microbianos, dentro de los últimos 3 meses.
Malestar y / o con enfermedad infecciosa en el momento del muestreo, el embarazo o la lactancia se consideraron como criterios de exclusión de la investigación.

TOMA DE MUESTRA
El sitio de implante fue aislado utilizando rollos de algodón para evitar la contaminación con saliva. Entonces, la placa supragingival se eliminó, y la placa se obtuvo de la parte más profunda del surco implantes utilizando una cureta estéril.

Amplificación del DNA por la PCR usando los diferentes componentes y usando agua pura para el marcador negativo.

DGGE
Luego de la PCR, los fragmentos se somenten a DGGE, para indentificar cada uno de los microorganismos presentes en la muestra.
Los cebadores abrazadera GC fueron confirmados en el apoyo de la amplificación por PCR de 16S rDNA, productos a partir de DNA extraído de una variedad de patógenos periodontales, incluyendo Treponema denticola y forsythia Tannerella.

SECUENCIA DE LOS PRODUCTOS DGGE
Las bandas fueron extirpadas y luego estraidas del gel para rectificar la prueva usando de nuevo la PCR utilizando los mismos cebadores de DGGE.99% de identidad se utilizó como punto de corte para la identificación positiva de una especie bacteriana. El número promedio de pares de bases (pb) utilizados para el análisis de la secuenciación fue de 116 pb.

ESPECIES PCR ESPECIFICA
MATERIALES Y METODOS
Se realizó usando cebadores específicos de especie dirigidos a regiones específicas en Aa, Porphyromonas gingivalis, Prevotella intermedia, o Staphylococcus aureus. Los productos de PCR se separaron a través de agarosa al 1,5%, se tiñeron. Por lo menos dos bandas independientes obtenidos para cada PCR específica de la especie se cortaron de los geles y se secuenció para confirmar las especies auténticos.

El porcentaje global de éxito secuencias obtenidas fue del 68,2%, que van del 43% al 100% por tema, y aproximadamente 26 especies fueron identificadas.
Los porcentajes de las diferentes especies bacterianas se muestra en la figura. 1, en esta figura la presencia de más de una cepa bacteriana de la misma especie en una muestra se registró como una sola incidencia.

Figura 3: Correlaciones de bacterias diferentes con profundidad de la bolsa (PD), mostró el porcentaje más alto de patógenos (rojo y naranja grupo) bacterias (82%) en los bolsillos más superficiales (<3 mm). Disminuyo con la profundidad de la bolsa en aumento (51%), pero aumentó de nuevo (63%) para los bolsillos más llenos. Otro factor notable fue la relación positiva entre los porcentajes de bacterias altamente patógenas (rojo grupo) con profundidad de la bolsa.
