Our skin cells die and are replaced by new cells every21 days. This renewal is made possible by stem cells.Although stem cells ensure the daily renewal of ourskin, they are only present in small numbers with ashort life span and remain protected in the deepestlayers of our tissues. As they are precious and fragile,
they act via “adult stem cells” which are predisposedto renew one or more precise cell types and are the
main cells responsible for skin cell balance and theirperpetual renewal.
With age, stress and external aggressions, adult stem cells become older,lose their vitality and capacity to divide, so that skin renewal becomes
slower and wrinkles appear and deepen.
Rather than act on stem cell multiplication or proliferation processes(generation of an unspecified and uncontrolled number of daughter
cells), CODIF Recherche et Nature is developing a new approach involvingthe division capacities (generation of two daughter cells alone) of ageing
stem cells in the dermis (Phycojuvenine) and epidermis (PhycosaccharideAI), by returning them to their youthful state.
Phycojuvenine acts on adult stem cells in the dermis in 3 steps:
1 - It restores ageing adult stem cells of the dermis with their capacity for division,thereby stimulating their action.
2 - It protects adult stem cells from ageing by preserving and prolonging their efficacy.

In-vitro test: Revitalizing effect of 1% Phycojuvenine on adult stem cells in the dermis.
Protocol:
Growth of adult stem cells for 24 hours under different culture
conditions:
- Young skin condition (about 20 years): FCS= 10% - bFGF= 1ng/ml
- Mature skin condition (about 40 years): FCS= 10% - bFGF=0.2ng/ml
- Old skin condition (about 60 years): FCS= 2% - bFGF=0.2ng/ml
FCS = Fetal calf serum - bFGF= basic Fibroblast Growth Factor
Stem cell vitality is evaluated from the reduction inMTT (tetrazolium salt) by a mitochondrial enzymeduring cell respiration. The reduction of MTT is monitoredby measuring the optical density (purple compoundwhen reduced).

In biology, a stem cell is a cell capable, on the one hand, of giving riseto any type of specialized cell and, on the other, virtually reproducingitself indefinitely…Virtually only, as stem cells are by definition dormant cells whichrapidly exhaust their division capacities and therefore their life spanif they are stimulated too often. As their number is limited, once theyare destroyed, they are not renewed and the skin loses its capacity toregenerate itself…
The key to perpetual cell renewal resides in the vitality of adultstem cells.
These adult stem cells are produced by stem cells themselves withthe role of proliferating. Unlike stem cells which are able to reconstituteany tissue of the human body, adult stem cells are already specializedaccording to the tissue in which they are produced. Hence adult stemcells in the dermis terminate their differentiation by forming fibroblastswhich are responsible for collagen synthesis.In the skin, the activity of adult stem cells is stimulated by growthfactors, such as bFGF (basic Fibroblast Growth Factor) in the dermis,the concentration of which gradually falls with age. The vitality, activityand capacity for division of ageing adult stem cells thereforedecreases with time. This ageing leads to a reduction in collagensynthesis and an accentuation of wrinkles.
1% Phycojuvenine significantly revitalizes adult stem cells in mature and old skin. Itsaction is optimal on mature skins, which recover a cell vitality equivalent to that ofyoung skin. As adult stem cells from young skin already has an optimal vitality,Phycojuvenine does not have a significant effect on this cell population.

Protocol:Microscopic examination of adult stem cell cultures incubated for 24 h under different culture conditions:

By its revitalizing action on adult stem cells, 1% Phycojuvenine restores theircapacity for division. Ageing skins recover similar levels of adult stem cells asyoung skins.
In-vitro test: Anti-ageing effect of 1% Phycojuvenine on dermal adult stemcells.
Fibroblast morphology changes with ageing and up to 7 differentmorphotypes may be defined. The age of fibroblasts may then beevaluated from their appearance. This method was used to evaluatethe effect of Phycojuvenine on human fibroblast cultures subjectedto an ageing process.
Protocol:Ageing of fibroblasts was induced by UV exposure in the presence or absence of 5%Phycojuvenine.
The addition of Phycojuvenine to the culture mediumdisplaces the average age of fibroblasts frommorphotype V to morphotype II to III.Phycojuvenine has a rejuvenating effect on fibroblastsby rejuvenating adult stem cells and then preventingthem from ageing.
In-vitro test: Rejuvenating effect of 5% Phycojuvenine on fibroblastmorphology
In-vitro test: Reinitialization of the division capacities of adult stem cells ofthe dermis by 1% Phycojuvenine.
Protocol: the cell ageing process is simulated by UVB radiation. Adult stemcell cultures were incubated for 24 hours before and after UVB radiation (20to 30 mJ/cm2). Cell vitality was evaluated from the reduction of MTT (tetrazoliumsalt) by mitochondria during cell respiration. Reduction of MTT is monitored bymeasuring the optical density (purple compound when it is reduced)

From 1%, Phycojuvenine has a significantanti-ageing effect on stem cells from youngand old skin. This anti-ageing effect is more visible on olderskins as cell ageing weakens the internal defense systems.

Protocol: the effect of Phycojuvenine on humanfibroblast collagen synthesis was evaluated byfollowing the incorporation of proline which is themain amino-acid in collagen molecules.

Clinical test: Rejuvenating effect of 1% Phycojuvenine measured on crow's footwrinkles.
Protocol: 15 volunteers aged from 40 to 55years; 2 applications per day for 28 days on theface.
Change in depth of main wrinkle:
After 28 days of twice-daily use of 1%Phycojuvenine, 80% of volunteersobserved a significant reduction inthe main wrinkle of -6.2% on averageand up to -15.6%
This reduction in the depth of wrinkleswas accompanied by a smoothingeffect, with a reduction in meanroughness of up to -29.6%.
