2. Intended Purpose of Microscopes

The Leica DM6000 B microscope, to which thisuser manual belongs, is designed for biologicalroutine and research applications. This includesthe examination of samples taken from the hu-man body with a view to providing informationon physiological or pathological states orcongenital abnormalities, or to determining thesafety and compatibility with potential recipients,or to monitoring therapeutic measures.

The Leica DM6000 M microscope is designedfor examinations in the field of materialsresearch.

All the above-named microscopes comply withthe Council Directive 98/79/EEC concerning invitro diagnostics. They also conform to theCouncil Directives 73/23/EEC concerningelectrical apparatus and 89/336/EEC concerningelectromagnetic compatibility for use in anindustrial environment.

Caution!

The manufacturer assumes no liability fordamage caused by, or any risks arising fromusing the microscopes for other purposesthan those for which they are intended ornot using them within the specifications ofLeica Microsystems Wetzlar GmbH.In such cases the conformity declarationshall cease to be valid.

These (IVD) devices are not intended for usein the patient environment defined by DINVDE 0100-710. Neither are they intended forcombining with medical devices accordingto EN 60601-1. 3.1 General Safety Notes

Caution!

The power plug may only be plugged into anoutlet equipped with a grounding contact.

Do not interfere with the grounding functionby using an extension cord without a groundwire. Any interruption of the ground wire in-side or outside of the device, or release ofthe ground wire connection, can cause thedevice to become hazardous. Intentionalground interruption is not permitted!

Caution!

Through activating to the groundingconnection (earth screw on the back of theLeica CTR6000 Electronics Box) ancillaryequipment with its own and/or extra powersupply may be brought to the same groundwire potential. For connections without aground connector, Leica Service must beconsulted.

Caution!

Never use any fuses as replacements otherthan those of the types and the current rat-ings listed here. Using patched fuses orbridging the fuse holder is not permitted.

The microscope’s electrical accessory com-ponents are not protected against water.Water can cause electric shock.

Caution!

Protect the microscope from excessive tem-perature fluctuations. Such fluctuations canlead to the accumulation of condensation,which can damage the electrical and opticalcomponents.Ambient temperature: 15-35°C.

Before exchanging the fuses or lamps, beabsolutely certain to switch-off the mainpower switch and remove the power cable.

5. Unpacking the Microscope

The device is delivered in several boxes.

The stand box contains the following compo-nents:

• Stand with integrated incident light axis andobjective nosepiece

• Specimen stage with stage bracket

• Power cable and PC connecting cable

• CD with Leica DMControl software package

• Instructions and list of microscope defaultsettings („Identification Sheet“)

The system box contains the microscope acces-sories:

• Tube

• Eyepieces

• Objectives

• Condenser

• Lamp housings with accessories

• Fitting tool

• Depending on configuration, additional micro-scope accessories such as filter cubes, etc.

The CTR6000 Electronics Box, the remotecontrol element SmartMove and the externalebq 100 supply unit* are delivered in separatepackaging.

First, carefully remove all components from thetransportation and packaging materials.

Note:

If at all possible, avoid touching the lens sur-faces of the objectives. If fingerprints do appearon the glass surfaces, remove them with a softleather or linen cloth. Even small traces of fingerperspiration can damage the surfaces in a shorttime. See the chapter, "Care of the Microscope"→ p. 81, for additional instructions.

Caution!

Do not yet connect the microscope and pe-ripherals to the power supply at this point!

Installation location

Work with the microscope should be performedin a dust-free room, which is free of oil vaporsand other chemical vapors, as well as extremehumidity. At the workplace, large temperaturefluctuations, direct sunlight and vibrationsshould be avoided. These conditions can distortmeasurements and micrographic images.

Allowable ambient conditionsTemperature 15-35°CRelative humidity maximum 80% up to 30°C

Microscopes in warm and warm-damp climaticzones require special care in order to preventthe build up of fungus.See the chapter, "Care of the Microscope" → p. 81,for additional instructions.

Caution!

Electrical components must be placed at least10 cm from the wall and away from flamma-ble substances.

Transport

For shipping or transporting the microscopeand its accessory components, the originalpackaging should be used.

As a precaution to prevent damage from vibra-tions, the following components should be dis-assembled and packaged separately:

• Unscrew the objectives.

• Remove the condenser.

• Remove the stage.

• Remove the lamp housings.

• Disassemble the burner of 106 z lamp housing.

• Remove all moving or loose parts.

6. Assembling the Microscope

The microscope components are logically as-sembled in this order:

• Stage• Condenser• Tube• Eyepieces• Objectives• Light sources• Filter cubes/reflectors*

Only a few commonly used screwdrivers andkeys are necessary for assembly, which are in-cluded in the delivery package.

When using intermediate systems and opticalaccessories, the sequence may vary.In this case, read Chapter"6.10 Optional accessories" → p. 34.

Note:

For thicker specimens (Leica DM6000 M) thestage can be set to a correspondingly lowerlevel.

Caution:

This changes the pre-set focus position and thelower threshold. Both positions have to be setagain. See 8.3. Focusing, → p. 60.

6.1 Stage

Caution:

Before assembling the stage, make sure no ob-jectives are installed!

• Place the specimen holder on the stage andfasten it with the two screws (5.1).

• Using the condenser height adjuster (5.2), turnthe condenser holder completely upwards,i.e. as close to the stage as possible.

• Loosen the stage clamp (5.3) slightly.

• From above, set the stage clamp onto thedovetail guide (6.2) and push the stage down-wards until the upper end of the dovetailguide is tightly fastened to the upper end ofthe stage clamp.

• Firmly tighten the stage clamp (6.1).

6.2 Condenser

• Screw the condenser head into thecondenser.

• Using the condenser height adjuster (7.4), turnthe condenser holder (7.1) completely down-wards.

• Unscrew the clamping screw for the con-denser (7.3) far enough so that the condensercan be inserted from the front.

• From the front, insert the condenser into thecondenser holder as far as it will go. On theunderside of the condenser, there is an orien-tation pin (8.1), which must be located in theguiding notch (9.1).

• Pull the condenser’s clamping screw (7.3) sothat the condenser is locked in place.

• Connect the condenser by connecting thecondenser cable (10.1) with the stand.

6.3 Tube and Eyepieces

The tube is mounted to the stand either directly orwith the use of intermediate modules. It is fastenedin place with the side clamping screw (12.1).

• Only for the motorized tube MBDT:Remove the transportation lock (11.1) from thebottom side of the tube.

• Loosen the clamping screw (12.1). on thestand.

• Insert the tube in the circular receptacle(dovetail ring).

• Retighten the clamping screw (12.1).

• Only for the motorized tube MBDT:Connect the tube to the stand with the con-nector socket (13.1).

• The eyepieces are inserted into the eyepiecetubes on the tube.

Note:

For eyepieces that are not included in shipment,we recommend to learn them in with the Soft-ware Leica DMControl, module: Configuration.This ensures that the information about totalmagnification on the LeicaScreen is correct.

6.4 Objectives

The receptacles on the objective turrets arenumbered (Fig. 14). The individual objectiveshave already pre-assigned positions at thefactory according to their configuration.A list of the exact objective positions is providedin shipment (“Identification Sheet” ).

Cover unoccupied threads on the turret withdust protector caps!

107/2 Lamp Housing

This lamp housing is used with a 12V 100W halo-gen lamp, which is already mounted.In case the lamp has to be removed:

• Remove the fastener screw on the housing(Fig. 15).

• Remove the housing by pulling it upwards.

• Remove the lamp.

• Insert the new 12V 100W lamp (16.1) with thedust cover straight into the socket until itstops. Be sure that the lamp is insertedstraight.

• Remove the lamp’s dust cover.

Caution!

Do not remove the lamp’s dust cover untilyou have installed the lamp. Avoidfingerprints on the lamp.

• Replace the housing and fasten it in place us-ing the fastening screw.

• Place the lamp housing in the transmittedlight lamp housing receptacle (17.2) and fas-ten it with the clamping screw on the side.

• Connect the lamp housing to the power supplyfor transmitted light (symbol: ) (17.3).

• Now connect the lamp power cable of themicroscope (17.5) to the Leica CTR6000Electronics Box (18.1).

6.6 Light Sources for the Incident Light Axis

Caution!

Light sources pose a potential irradiationrisk (glare, UV-radiation, IR-radiation).Therefore, lamps have to be operated inclosed housings.

During assembly, always unplug the powersupply unit of the 106 z lamp housing from itssocket.

During assembly work on xenon burners, al-ways wear the supplied protective glovesand face protection (Fig. 19) (risk of explosion).

Never touch the glass parts of the burnerwith bare hands.Never look directly into the beam path(blinding hazard).

106 z lamp housing

This lamp housing is used with a 12V 100W halo-gen lamp or various gas discharge lamps.

Caution!

Make sure to follow the instructions andsafety notes of the lamp supplier.Before changing lamps allow at least 30 minsfor cooling down!

Inserting the 12V 100W halogen lamp into the106 z lamp housing

• Unscrew the fastening screws of the coverand lift up the cover (20.1).

• Unscrew the fastening screws of the lampmount (20.8) and pull out the mount (Fig. 21).

• Insert the lamp with the dust cover straightinto the socket until it stops.

Caution!

Do not remove the lamp’s dust cover untilyou have installed the lamp. Avoidfingerprints on the lamp.

• Remove the dust cover.

• Reinsert the lamp mount and retighten thefastening screw (20.8).

• Close the lamp housing and retighten the fas-tening screws.

• Place the lamp housing in the incident lightlamp housing receptacle (22.1) and fasten itwith the clamping screw on the side.

• Connect the lamp housing to the power supplyfor incident light (symbol ) (22.4).

• Now connect the lamp power cable of themicroscope (22.5) to the Leica CTR6000electronics box (18.1, p. 25)

Inserting the gas discharge lamps (Hg and Xe)into the 106z lamp housing

Hg and Xe lamps are powered by separatesupply units.Read the separate instruction manual providedwith these supply units.

Hg 50 burner:After installation, the labeling must be upright.If a glass melt nipple is present (23a.4), posi-tion it by turning the burner so that the nippledoes not come in the way of the beam pathlater, but instead is positioned sideways.

Xe 75 burner:Remove the burner’s dust cover (23b.5) afteryou have installed the burner.

• To open the 106 z lamp housing, unscrew thefastening screws on the cover.

• Remove the transport anchorage (red plasticrod in place of the burner) in the lamp mount.To do so, remove the lower clamp (23.1). Pullup the cooling element (23.3) and turn it to theside. Detach the lower clamp system (23.2)and remove the transport anchorage.

• Install the burner in reverse order.

• Insert the lamp mount, with the burner in-stalled, into the lamp housing and tighten itwith the screws (24.8).

• Put the lid down again. Plug in the contactplug as far as it goes and retighten thescrews.

• Place the lamp housing in the incident lightlamp housing receptacle (25.1) and fasten itwith the clamping screw on the side.

• Connect the lamp housing to the power supply(26.1).

6.7 Equipping the Incident Light Turret Disc

The receptacles on the turret are numbered.According to your equipment, the individualfilter and/or reflector cubes have already pre-assigned positions. A list is provided along withyour shipment (“identification sheet“).

Insert the filter and reflector cubes in the fol-lowing manner:

• Equip the incident light turret only when themicroscope is switched off.

• Remove the front cover from the upper part ofthe microscope (Fig. 29). Push the retentionpin (29.2 or 30.2) to move the turret.Releasing the retention pin locks the turret.

• Insert the filter or reflector cube into themounting in front of you according to theidentification sheet provided.To do so, place the filter or reflector cube onthe right side and press it to the left into themounting (Fig. 31).

At the 4-fold and 5-fold filter turrets, thenumbers are right below the mounting. At the 8-fold turret, the numbers are on the bottom of thedisc.

• Push the retention pin (29.2 or 30.2) andcontinue to turn the filter turret until youreach the next locking position.

• Again make sure that the turret engages(retention pin unlocks) and insert the nextfilter and/or reflector cube as describedabove.

• When all filters and reflector cubes have beeninserted, close the front cover plate again.

Note:

At the Leica DM6000 M, 2 positions for brightfield and dark field reflector cubes may be set(depending on configuration).

6.8 Polarizer and Analyzer

ICT/P transmitted light polarizer

• Using the left clamping screw, fasten the ICT/Ptransmitted light polarizer to the underside ofthe condenser holder (Fig. 32).

• Make sure that the red index point on thefront of the polarizer is aligned with 0.

• If necessary, insert the compensators (λ- andλ /4 plates) into the polarizer’s receptacle(Fig. 33).

Incident light polarizers:

R/P polarizer, rotating polarizer

L/ICR, R/ICR polarizer

• Remove the plug cap on the right side of theincident light axis (Fig. 34).

• Insert the polarizer into the receptacle until itlatches in place.

Attention:

Insert the polarizer only in the front receptacle!

• A motorized polarizer is already installed andready for operation in the DIC condenser.

Transmitted light and incident light analyzer

• Remove the plug cap on the left side of thestand.

• Insert the analyzer into the receptacle until itlatches in place (Fig. 35).

Motorized analyzer

• Insert the analyzer cube as described in sec-tion 6.7 "Equipping the Incident Light TurretDisc " → p. 31, in the corresponding positionon the filter turret. See the list provided(“Identification Sheet”) for the correctposition.

6.10 Optional Accessories

Camera

Connecting a cameraA camera can be connected via c-Mount orVario-Mounts.

• Attach the c-Mount or the Vario-Mount to thetop port of the tube and fasten it tightly withthe side clamping screw.

• Screw on the camera.

Note:

The use of a c-Mount or a Vario-Mount shouldbe learned in with the Software LeicaDMControl, module: Configuration.

Connecting two camerasThe dual port enables you to connect twocameras (one digital and one analog) to themicroscope.

• When using a DC-type digital camera, thecamera is connected to the PCI-card of thePC.

• When using a DFC-type digital camera, thecamera is connected to the Fire Wire card ofthe PC.

For raising the eye level of the tube opening, theergomodule may be used.It is fastened in place with the side clampingscrew.

Mirror Housing

• Place the mirror housing directly onto thelamp housing receptacle on the back of thestand and attach it using the side clampingscrew.

• Place the lamp housing onto the mirror hous-ing and fasten it using the correspondingclamping screw on the side.

6.12 Connecting the Computer

Note:

To start the Leica DMControl, the COM1 serialinterface must not be occupied by another pro-gram or driver. However, this often happenswhen using palmtops or other electronicplanners and installing additional modems ordevices. The accessory devices always have tobe deactivated before using the LeicaDMControl software.

• Connect the COM1 serial interface of thecomputer to the RS232C (39.1) port on theback of the stand. Use the serial cable sup-plied.

6.13 Connection to the Power Supply

• After completing the assembly work, connectthe Leica CTR6000 electronics box to the po-wer supply using the power cable supplied(port 38.1).

• When using the ebq 100 supply unit, thisalso has to be connected to the power supply(port 40.1).

7. Startup

7.1 Functional Principle

1. Intelligent automation

• Switch between different contrasting methods by pressing just one button. Light rings,DIC-prisms, etc. are inserted into the beam path automatically.

• The microscope recognizes the objective in use and the corresponding contrastingmethod. Therefore, the values for intensity (INT), aperture diaphragms (AP) and fielddiaphragms (FD) are always set appropriately.

• The information about INT, AP and FD is always given with regard to the currentlyactivated light axis (transmitted light or incident light).

• The values for INT, AP and FD can be changed individually. This overwrites the previoussettings. Actual settings are stored automatically and can be recalled when switching onthe microscope again.

2. Control elements

• LeicaScreenControls the microscope’s functions via touch screen.

• SmartMoveFor stage and focus control

• Fixed function keys at the standFor INT, AP, FD and to switch between transmitted light and incident light.

• Variable function keys at the stand and at the SmartMoveThe function keys are assigned logical functions before delivery, corresponding to theconfiguration of your microscope. These functions can be re-programmed according toyour individual requirements.

• Entire control of microscope and camera via software(Leica DMControl, Leica application software)

7.2 Switching on

• Start with the objective with the lowestmagnification.

• Switch on the Leica CTR6000 Electronics Boxat the on/off switch (41.1). When in operation,the pilot lamp will light up green (41.2). Allmotorized microscope components first und-ergo an initialization phase.

Note:

If you have connected a PC, please switch onthe electronics box first and the computerafterwards.

After initialization is completed, theLeicaScreen shows the current microscopesetting (Fig. 42).

If one of the components is not mountedproperly, e.g. the cover of the filter turret is notlocked in place, an error will show up on theLeicaScreen.See chapter ‘Trouble Shooting’, → p. 80.

The microscopic components such as dia-phragms, condenser, light and phase rings arealready pre-centered in the factory. However,re-centering may be necessary due to transpor-tation and assembly.Before proceeding with the necessary steps,first familiarize yourself with the LeicaScreenand control panel.

Caution!

After turning on the gas discharge lamp, theburner must be immediately adjusted. There-fore, do not turn on the power supply unityet. First, work in transmitted light in order tofamiliarize yourself with the microscope’scontrols.

7.3 The LeicaScreen

On the status page, the LeicaScreen shows thecurrent microscope settings. The displaydepends on the microscope’s configuration.Apart from this, the microscope can be operatedvia several menu levels on the LeicaScreen.Please see the abbreviation index for a list ofabbreviations used → p. 84.Read the separate instruction manual of theLeicaScreen.

The display represents different levels ofoperation:

Level 1: Navigation panelLevel 2: Menu barLevel 3: Information and control panel

After selecting a pictogram on the navigationpanel (left column on the display), thecorresponding menu bar opens in the upper rowof the display. The items on the control panelchange accordingly. The currently activefunctions are highlighted in black. .

Variable Function Keys at the standA factory preset is performed which fits yourmicroscope configuration. The function keysare labeled accordingly, and a separatedescription of the key occupation accompaniesthe microscope (“Identification Sheet“).Abbreviations are listed on p.41f.

7.4 The Function Keys at the stand

There is a row of function keys both on the rightand left side of the stand. Some of these keysare defined, and some of them are variable. Thevariable function keys have various meaningsdepending on the microscope configuration.

Defined Function Keys on the left side of thestandThe TL/IL key (43.1) switches between incidentlight and transmitted light. The last contrastmethod used is restored.The INT (43.3) keys adjust the light intensity indi-vidually. Settings can be made either in large orsmall increments. Pushing both INT buttons atthe same time switches between coarse andfine setting. If the fine setting has beenselected, the display indicates „Intensity fine“.The AP (43.2) keys for the aperture diaphragmand FD (43.4) for the field diaphragm are usedto open/close each diaphragm.

Note:

Changes in light intensity and settings ofaperture and field diaphragms are stored for thecurrent objective and contrasting method.

7.6 Köhler Illumination

7.6.1 Transmitted Light

For each objective, reasonable values for theaperture diaphragm and the field diaphragm arealready set. The condenser is also pre-adjustedin the factory.However, it may be necessary to re-adjust thecondenser in some cases. Therefore, check thecondenser centering.

The following procedure is provided for thetransmitted light-bright field illumination.

• Select an objective with moderatemagnification (10x-20x).

• If necessary, activate the transmitted lightaxis by pushing the TL/IL button (43.1).

• Choose "bright field" as the contrast methodby pressing the BF key (one of the variablefunction keys at the stand or at theLeicaScreen).

• Insert the specimen in the stage’s specimenholder (45.1).

• Focus on the specimen with the SmartMoveor focus wheel.

• Set the light intensity using the INT keys(43.3).

• Close the field diaphragm with the FD functionkey (43.4) until the edge of the diaphragm ap-pears in the specimen plane.

• Using the condenser height adjuster (45.2),adjust the condenser until the edge of thefield diaphragm appears in sharp relief.

• If the image does not appear in the middle ofthe field of view (47c), the condenser must bemoved into the middle of the field of view withthe help of the two centering bolts (46.1).

• Open the field diaphragm just enough for it todisappear from the field of view (47d).

Caution:

The light of the condenser depends on thethickness of the specimen. It has to be adjustedfor each specimen.

7.6.2 Incident Light

For each objective there are reasonable valuesassigned to aperture and field diaphragm. Theincident light module is also pre-centered at thefactory.

Due to transportation and setup of the stand,however, you might have to re-center theincident light module. Please check the apertureand field diaphragm centering.The following procedure refers to the incidentlight bright field illumination.

• Select an objective with moderatemagnification (10x-20x)

• If necessary, activate the incident light axisby pressing the TL/IL key (43.1).

• Activate the bright field contrasting methodby pressing IL-BF/Fluo (one of the variablefunction keys at the stand or at theLeicaScreen).

• Insert the specimen in the stage’s specimenholder (45.1).

• Focus on the specimen with the SmartMoveor focus wheel.

• Adjust the light intensity with the INT keys(43.3).

Adjusting the field diaphragm

• Close the field diaphragm with the FD keys(43.4) until the edge of the diaphragm (roundor rectangular) appears on the specimen pla-ne.

• If the boundaries of the field diaphragm arenot in the center of the field of view, thediaphragm has to be moved with the twocentering bolts (48a.1) on the right side of thestand.

• With the function keys FD (43.4) on the leftside of the stand, open the field diaphragmuntil it just disappears from the field of view.

• When using a digital camera, rectangularfield diaphragms are recommended. Adjustthe size of the diaphragm to the chip size ofyour camera.

Adjusting the aperture diaphragm

(DM6000 M only)

• Remove one eyepiece.

• Close the aperture diaphragm with the APkeys (43.2) until the edge of the diaphragmappears on the exit pupille of the objective(aperture field plane).

• If the image does not appear in the center ofthe field of view, the aperture diaphragm hasto be moved with the two centering bolts(48b.1) on the upper left side of the stand.

• Open the aperture diaphragm so it coversapprox. 2/3 of the field of view .

7.7 Checking Phase Contrast Rings

If your microscope is equipped for the use ofphase contrast, the light rings that fit the objec-tives are built into the condenser.The light rings are already centered in thefactory. However, the centering should berechecked.

Note:

Every objective is assigned its own light ring inthe condenser disc. Therefore, a check must beperformed for each objective. When swivelingin a suitable objective for phase contrast, thecorresponding light ring is set automatically.

• Press the BF (Bright Field) button (one of thevariable function keys at the stand or at theLeicaScreen).

• In the place of an eyepiece, insert the focus-ing telescope (Fig. 49) into the observationtube.

• Select the phase contrast objective with thelowest magnification (one of the variablefunction keys at the stand or at theSmartMove).

• Focus on the specimen with the SmartMoveor focus wheel.

• Focus the ring structure (50a) by slightly loos-ening the clamping ring (49.2) and moving theeyelens (49.1).

• Retighten the clamping ring.

• Press the PH (Phase Contrast) button (one ofthe variable function keys at the stand or atthe LeicaScreen). The ring diaphragm in thecondenser is pivoted in.

• If the light ring and the phase ring are notshown as arranged in Fig. 50c, the light ringmust be centered.

• Insert the centering key through the corre-sponding openings (51.1) in the condenserholder.

• Turn the centering screws until the dark ring(phase ring in the objective) is congruent withthe slightly narrower bright ring (light ring incondenser) (50 c).

Caution!

During change of objectives the centeringkeys must not remain in the openings of thecondenser. They can block the condenser.

• Repeat the process for all other phase con-trast objectives.

• Remove the centering keys after thecentering procedure.

• Select POL contrast method (one of the varia-ble function keys at the stand or at theLeicaScreen).

• Inserting centering key through the corre-sponding opening (51.2).

• Turn it until maximum darkness is reached.

7.9 Adjusting the Light Sources

Transmitted Light Axis (TL) with 107/2 Lamphousing

The 107/2 lamp housing with 12 V 100 W halogenlamp has a defined presetting. The lamp neednot to be centered.

Incident light axis (IL) with 106 z lamp housing

• When a supply unit is used, it is turned onfirst.

• Activate the incident light axis using the TL/ILfunction key. FLUO (Leica DM6000 B) or IL (Lei-ca DM6000 M) appears in the LeicaScreen.

• Insert the reflector cube for lamp adjustment(Fig. 52) into the filter turret in place of a filtercube. (See → p. 31).Note the name of the exchanged filter cube.

Note:

To avoid incorrect adjustment, we recommendto remove the filter cube on the left side of thereflector cube.

• Turn the reflector into the beam path.The reflector has reached the correct positionwhen the name of the exchanged filter cube isshown in the upper right of the LeicaScreen .

For the 106z lamp housing, the direct filament im-age (for halogen lamps) or direct arc image (forgas discharge lamps), and its mirror image arefocused separately and adjusted to each other.

On the left side of the microscope, there is anadjustment window (1.14, p. 16) for mapping thelight source.

While observing the light source in the adjust-ment window, the lamp is adjusted as follows:

Centering the 12 V 100 W Halogen Lamp

• In the adjustment window, you see the directfilament image and the mirror image, which inmost cases are not aligned.

• Focus the direct filament image with the col-lector (53.6).

• Use the adjusting buttons on the rear side ofthe lamp housing (53.2, 53.4) to pivot the lampfilament’s mirror image to the side or com-pletely out of the beam path. The lamp fila-ment’s focused image remains visible (Fig.54).

• Adjust the direct filament image using the ad-justing knobs (53.1) and (46.5) so that thecentering surface is halfway covered (Fig. 55).

• Then pivot the lamp filament’s mirror imagewith the adjusting knobs (53.2 and 4), andfocus it using the reflector (53.3).

• Align the mirror image symmetrically to the fil-ament image (Fig. 56). To do so, use the ad-justing knobs (53.2) and (53.4) again.

• Defocus the image with the collector head(53.6) until the filament image and mirror im-age are no longer recognizable and the imageis homogeneously illuminated.

• Exchange the reflector cube for lampadjustment for the original filter cube.Note:Turn off the microscope before exchangingthe reflector cube.

• Then pivot the arc’s mirror image with the ad-justing knobs (53.2 and 4) and focus it usingthe reflector (53.3).

• Use the adjusting knobs (53.2 and 4) to orientthe mirror image symmetrically to the directimage (Fig. 59).

• Defocus the image with the collector knob(53.6) until the arc image and mirror image areno longer recognizable and the image ishomogeneously illuminated.

• Exchange the reflector cube for lampadjustment for the original filter cube.

• Focus the direct image with the collector(53.6).

• Use the adjusting buttons to pivot the arc’smirror image on the rear side of the lamphousing (53.2,53.4) to the side or completelyout of the beam path. The arc’s focused im-age remains visible (Fig. 60).

• Use the adjusting buttons (53.1 and 5) to placethe direct arc image in the middle of thecentering plane, whereby the bright tip of thearc, the focal spot, should lie slightly outsidethe center (Fig. 61).

• Then pivot the arc’s mirror image with the ad-justing knobs (53.2) and (53.4) and focus it us-ing the reflector (53.3).

• Use the adjusting knobs (53.2 and 4) to orientthe mirror image symmetrically to the directimage (Fig. 62).The V-shaped irradiation of the direct imageand mirror image arcs can be superimposed.

Caution!

The bright tips of the arcs, the focal spots, mustnever be projected onto each other, as this re-sults in a danger of explosion by overheating.

Caution:

In older lamps, the structure of the arc is nolonger clearly recognizable. The image isthen more like that of a HG 50 lamp. The im-age and mirror image can no longer be su-perimposed exactly. In this case, align bothimages.

• Using the collector, defocus the image withthe knob (53.6) until the arc image and mirrorimage are no longer recognizable and the im-age is homogeneously illuminated.

• Exchange the reflector cube for lampadjustment for the original filter cube.Note:Turn off the microscope before exchangingthe reflector cube.

8.1 Switching on

When using a gas discharge lamp, the ebq 100external supply unit must be turned onseparately (63.1).

Switch on the Leica CTR6000 Electronics Box atthe on/off switch (64.1). When in operation, thepilot lamp will light up green (64.2).

During initialization, the stage will move to apre-set position. The stage is lowered. Themotorized tube will be set to the 100% VISposition. All most recently used settings will berecalled.

Caution:

The focus position (DM6000 B only) and thelower threshold will also be stored whenswitching off the microscope.

After the initialization is completed, the statusmenu of the LeicaScreen (Fig. 65) shows thecurrent microscope setting.

Note: (Reset-Function)

The microscope can be reset to the defaultsettings:

• When the microscope is switched off,press all 3 variable function keys on theleft stand section.

• Switch on the stand.

• Hold the keys pressed down untilinitialization is completed.

• The standard information is shown in theLeicaScreen.

• Switch off the instrument and switch it onagain. The settings are stored now.

Note:

Parfocality has been learned in at the factory.Mounting the objectives, however, can changethe values so that parfocality has to be learnedin again.We recommend to check parfocality beforesetting the thresholds. If necessary, it can belearned in with the Software Leica DMControl,module: Fine Tuning.

Image Focus

You can focus by using the turning knob (66.3)on the remote control element SmartMove.

Alternatively, you can use the focus wheels onboth sides of the stand.

The current Z-position is displayed on the statuspage of the LeicaScreen. When switching onthe microscope, before initialization the z-drivewill automatically go to a position which is 3mmbelow the most recently used Z-position.

Setting thresholds

The lower threshold can be set with theLeicaScreen and with the Software LeicaDMControl.The lower threshold is the same for allobjectives and can not be overrode.

Leica DM6000 B only:

An additional focus position, which can not beoverrode, can be set. This position should be setfor the DRY objective with the highestmagnification. For all other objectives it will beset automatically, regarding parfocality andworking distance.

There are two step sizes: Fine and Coarse.In the Fine mode, the value is adjusted to theobjective in use. The values are pre-defined.The settings can be changed with the SoftwareLeica DMControl, module: Fine Tuning.In the Coarse mode, the value is the same for allobjectives. 8.5 Eyepieces

Note:

The eyepiece’s aperture protector must beremoved or at least folded back, duringmicroscopy while wearing eyeglasses .We recommend to take off glasses with bifocalor progressive-addition lenses when workingwith the microscope.

• For the adjustable tubes with documentationoutput, choose the 100% VIS position.

Eyepieces with Inlaid Reticle

• Focus the reticle by adjusting the eyelens.

• Focus on the object through this eyepiece.

• Then, close that eye and focus on thespecimen by adjusting only the secondocular.

Correction for Vision Problems

• With your right eye, look through the righteyepiece and bring the specimen into sharpfocus.

• Then, with your left eye, view the same speci-men and rotate the left eyepiece tube until theobject is brought into sharp focus. This doesnot affect the Z-position.

Note:

For eyepieces that are not included in delivery,we recommend to learn them in with the Soft-ware Leica DMControl, module: Configuration.This ensures that the information about totalmagnification on the LeicaScreen is correct.

8.6 Objectives

Selecting objectives

Objectives can be selected and rotated into thebeam path via motorized functions, by pressingthe variable function keys at the stand or at theSmartMove. They can also be selectedmanually. In this case be sure that thenosepiece turret locks into place.

The objective’s position in the turret is factory-set and must be adhered to while screwing inthe objectives (see Objective Assembly → p. 24)

When you rotate the objective into position, themicroscope automatically recalls:

• the optimal setting for the field diaphragm

• the optimal setting for the aperture diaphragm

• the settings for light intensity for the chosencontrast methode.

The objective magnification and the total magni-fication appear in the LeicaScreen → p. 58.

Changing the operation mode DRY or IMM(Immersion)

Each objective belongs to one of the followingcategories:1) Dry objectives (DRY)2) Immersion objectives (IMM)

Note:

It is possible to assign one objective to bothoperation modes, especially with dry objectiveswith >2mm working distance. The setting can bechanged with Software Leica DMControl,module: Fine Tuning.

Changing the operation mode

• First select the operation mode (IMM or DRY)

on the LeicaScreen, navigation item .The operation mode can also be selected withthe Software Leica DMControl.

• The stage is lowered by 3mm. This enablesyou to apply the immersion oil when changingfrom dry to immersion objectives. In thereverse case, the oil can be removed.The current objective remains in the beampath.

• Now press the button for the objective youwant to select. You can also press the graybutton for the currently activated objective.The objective will be rotated into the beampath.

• Start with a small level of magnification. Thenswitch to the next higher objective.

• For immersion objectives use the appropriateimmersion medium.OIL: only use optical immersion oil

according to DIN/ISO standards.Cleaning → p. 81

W: Water immersion.IMM: Universal objective for water,glycerol, oil immersion.

8.7 Magnification Changer

Optionally, a coded magnification changer canbe used, which is manually operated.On the knurled ring (72.1), the followingmagnification factors can be set:

The selected factor is indicated in theLeicaScreen or in the corresponding window ofthe Leica DMControl software and included inthe total magnification.

8.8 Light Sources

• The brightness is set using the function keys(73.5). Then, the INT function keys are as-signed to the currently active axis for trans-mitted light (TL) or incident light (IL).

• For TL and IL:Settings can be made either in large or smallincrements. Pushing both INT buttonssimultaneously switches between coarse andfine setting. The display on the LeicaScreen,menu page ‘Light’ (fig. 74), changesaccordingly.

Coarse adjustment: 0-20Fine adjustment: 0-255

• The light intensity is set and storedindividually for each objective and eachcontrast method.

Both diaphragms are already factory-set to theoptimum setting for the current objective andcontrasting method.

• The AP (73.2) keys for the aperture diaphragmand the FD keys (73.4) for the field diaphragmmay be used to change each diaphragm’s set-ting at any time. The display on theLeicaScreen, menu page ‘Light’ (Fig. 74),changes accordingly.

Caution:

When doing so, old values are overwritten andthe new values are stored!

• The function keys are always assigned to thecurrently active axis for transmitted light (TL)or incident light (IL).

9.1.1 Bright Field (TL)

• Insert the analyzer into the left side of thestand (76.1).

• Bring the polarizer and analyzer into cross po-sition until they reach maximum darkness.

• Insert a specimen and select an appropriateobjective.

Motorized procedure:

• After selecting the POL contrast method, thecondenser automatically switches to the posi-tion of the polarizer. The analyzer cube is alsoautomatically brought into the light path.

Combined procedure:

• For the Leica DM6000 B microscope, it ispossible to combine mechanical andmotorized components.

Alternatively:• Manually rotate the polarizer on the under-

side of the condenser into the light path (Fig.75).

• Likewise, manually insert the analyzer into theleft side of the stand (Fig. 76).Objective and condenser prisms areautomatically moved into the light path aswell.

11. Trouble Shooting

Problem

Stand

The microscope does not respond.

Illumination

The image is completely dark.

The image is unevenly or not uniformly illumi-nated.

The illumination "flickers."

The lamp does not illuminate immediately uponbeing switched on.

Cause/Remedy

Make sure that voltage is impressed.Make sure that the CTR6000 electronics box isconnected to the power supply.Check the cable connections.Inform service technician to change thefuses.

Open the shutter (→ p. 84).Check the connection of the lamp houses tothe microscope.Transmitted axis:Reflected (Fluo) axis:Make sure that the lamps are connected tothe power supply and that they are not defect.Inform service technician to change the fusesof the ebq 100.

Remove all unneeded filters from the lightpath.Center the lamp (→ p. 51ff).Replace the old lamp (→ p. 24ff).

Be sure that there is no loose connection atthe power supply.Replace the old lamp (→ p. 24ff).

The ebq 100 must be switched-on repeatedly.Hot Hg lamps should cool down beforeswitching on again.

Bright Field

The specimen can not be brought into focus.

Dark Field

No definite DF contrast is possible.

The image is unevenly or not uniformly illumi-nated.

Undesirable stray light

Phase contrast

No phase contrast is possible.

Cause/Remedy

Use the correct immersion medium.Lay the specimen with the cover glass to-wards the top.Make sure that the cover glass thickness iscorrect and that it suits the indication on theobjective.

Be sure that a DF objective is being used.The objective aperture setting is too high(maximum 0.75). If necessary, reduce the ob-jective aperture using the iris diaphragm onthe objective.Check the condenser centering.

The magnification is too weak. Use a highermagnification.

Clean the specimen and neighboring lenses(→ p. 81).

The specimen is too thick.The cover glass is not placed evenly.Check the centering of the light rings (→ p. 49).

Polarization

No polarization contrast is possible.

Fluorescence

The image is completely dark (no fluorescence).

The fluorescence is too weak.

Init Error!

Cause/Remedy

Bring the polarizer and analyzer into cross po-sition until they reach maximum darkness(without specimen) (→ p. 70, 75).

Open the shutter (→ p. 72).Select the incident light axis (IL) (→ p. 45).Check the antigen-antibody combination.Insert a new lamp (→ p. 26f).

Insert the Booster Lens (→ p. 35).Center the lamp (→ p. 51ff).Insert a new lamp (→ p. 26f).

Check the cable connections.Check if the front cover of the filter turret islocked in place.Check the currently used objectives, filtercubes, etc.Restart the microscope.

Caution!

Unplug the power supply before performingcleaning and maintenance work!Protect electrical components frommoisture!

Microscopes in warm and warm-damp climaticzones require special care in order to preventfungus contamination.The microscope should be cleaned after eachuse, and the microscope optics should be keptstrictly clean.

12.1 Dust Cover

Note:

To protect against dust, cover the microscope andaccessories with the dust cover after each use.

Caution!

Let lamps cool down before covering thestand with a dust cover. The dust cover isnot heat-resistant. In addition condensationwater may occur.

12.2 Cleaning

Caution:

Residual fiber and dust can create unwantedbackground fluorescence.

Cleaning Coated Parts

Dust and loose dirt particles can be removedwith a soft brush or lint-free cotton cloth.

Clinging dirt can be cleaned with all commerciallyavailable water solutions, benzine or alcohol.For cleaning coated parts, use a linen or leathercloth that is moistened with one of these sub-stances.

Caution:

Acetone, xylene or nitro-containing thinnercan harm the microscope and thus may notbe used.

Test cleaning solutions of unknown compositionfirst on a less visible area of the unit. Be surethat coated or plastic surfaces do not becomematted or etched.

Cleaning the Stage

Remove light-colored spots on the stage by rub-bing with paraffin oil or acid-free Vaseline.

Cleaning Glass Surfaces

Remove dust on glass surfaces with a fine, dryand fat-free hair brush, by blowing with a blowbag or vacuum suction.

Carefully remove stubborn dirt on glasssurfaces with a clean cloth moistened withdistilled water. If the dirt still can not beremoved, use pure alcohol, chloroform orbenzine.

Cleaning Objectives

Caution!

The objective may not be unscrewed duringcleaning. If damage appears on inner sur-faces, the objectives must be sent to yourLeica subsidiary for repair. We also adviseagainst cleaning the inside surfaces of theeyepieces.

The front lenses of objectives are cleaned asdescribed under "Cleaning Glass Surfaces". Theupper lens is cleaned by being blown off with apneumatic pump.

Removing Immersion Oil

Caution!

Follow safety instructions for immersion oil!

First, wipe off the immersion oil with a cleancotton cloth, and then re-wipe the surfaceseveral times with ethyl alcohol.

12.3 Handling Acids and Bases

For examinations using acids or other aggres-sive chemicals, particular caution must betaken.

Caution:

Be absolutely certain to prevent the opticsand mechanical parts from coming into con-tact with these chemicals.
