Using  the  Nimrod  toolkit,  a  set  of  tools  that  allows  for  inves6ga6ng  highly  complicated  parametric  systems,  my  goal  is  to  op6mize  Excita6on-­‐Contrac6on-­‐Transcrip6on-­‐Coupling  Model  (Shannon-­‐Bers-­‐Michailova  Model)  for  a  ventricular  cardiomyocyte  in  rabbits  and  run  sensi6vity  analysis  in  order  to  elucidate  how  the  model  behaves  under  various  s6muli.  

The  model  will  be  op6mized  and  fiIed  for  4  kine6c  measurements  of  calcium:  

§  Systolic  (mM)  §  Diastolic  (mM)  §  Time-­‐to-­‐peak  (ms)  §  Res6ng  6me  to  50%  peak  calcium  concentra6on  (ms)

Progress:  Conducted  Sensi6vity  Analysis  aRer  Perturbing  Parameters  10%    

•  Ran  sensi6vity  analysis  and  determined  effects  of  perturba6on  on  kine6c  parameters:  •  Those  parameters  were  selected  for  op6miza6on  based  off  which  produced  a  significantly  nega6ve  effect  (those  to  the  leR  of  0.0  and  below  a  z-­‐score  of  -­‐1.5)  

•  These  parameters  selected  will  be  fed  into  Nimrod/O  to  find  op6mal  parameter  values.

Future  Plans

•  Consider  the  addi6on  of  a  sodium  buffering  equa6on  in  the  nucleus  in  order  to  correct  driR  within  the  model.  

•  Will  con6nue  fi[ng  various  ranges  of  parameter  values  aRer  inves6ga6ng  a  feasible  range  of  parameters.  •  Conduct  sensi6vity  analysis  by  perturbing  parameters  in  the  nucleus  by  ±10,  30,  50,  and  100%.  

•  Run  addi6onal  sensi6vity  analysis  in  order  to  iden6fy  mechanisms  that  regulates  nuclear  Ca2+
