Immunoassay technologyWhat are ELISAs and multiplex immunoassays?
Both ELISAs and multiplex immunoassays are routinely used for the qualitative and quantitative assessment of cytokines,
chemokines, growth factors, phosphorylated targets, immunoglobulins, and other immunological markers. Detection
and quantitation of protein analytes from various biological samples, including serum, plasma, and other bodily fluids
(e.g., sputum, urine, etc.) as well as cell culture supernatants and cell lysates, indicate a multitude of biological and
pathological events. Immunoassays are used in areas of basic research for studying cell transformation and activation.
They have also proven to be valuable tools in drug development as they are commonly used for biomarker profiling of
disease versus normal states, as well as toxicity profiling.
eBioscience ELISAs and multiplex immunoassays use the sandwich immunoassay technique of two highly specific
antibodies that recognize two different epitopes on the same protein. This provides an extra layer of specificity
over competitive ELISA formats that employ only the use of a single antibody. To understand the advantages of
sandwich immunoassays, it is important to understand the basics of the assay.

1. The first antibody, specified as the capture antibody, is bound to either the well of the ELISA plate or to a capture bead in a ProcartaPlex Multiplex Immunoassay.
2. The sample of choice is incubated in a pre-coated ELISA well, or in the case of a ProcartaPlex assay, in a plate well with the capture bead mixture. During this incubation step, the specific antigen is captured on the solid surface (plate well or bead) through its specific binding to the capture antibody. The remaining sample is washed away.
3. A second biotinylated or direct horseradish peroxidase (HRP)-labeled, analyte-specific antibody is used for analyte detection. The detection antibody adds a layer of specificity for the sandwich immunoassays. Any unbound detection antibody is washed away.
4. A detection reagent used in the sandwich immunoassays allows for the detection and quantitation of the analyte. For ELISAs, the use of a horseradish peroxidase (HRP)-labeled, analyte-specific antibody triggers the colormetric conversion of the substrate and, hence, the development of color. For the ProcartaPlex assays, the detection reagent is a streptavidin-phycoerythrin (streptavidin-PE) conjugate, which allows the instrument to quantitate the analyte levels based on fluorescence intensity.
5. High-antigen concentration causes more substrate to be converted, which produces a stronger colorimetric signal for the ELISA and higher fluorescent readings for ProcartaPlex assays. Correlation of the color development or fluorescence intensity can then be used to calculate the concentration of an antigen in a sample when compared to a standard curve of known concentration.

eBioscience High Sensitivity ELISAs detect low expressing proteins by enhancing the
signal obtained from conventional colorimetric ELISAs. A conventional sandwich ELISA
signal development is accomplished only via the biotin/streptavidin-HRP amplification
system. eBioscience High Sensitivity ELISAs further enhance this signal with the additional
signal amplification step using a tyramide amplification reagent to generate multiple
biotin binding sites for additional streptavidin-HRP molecules. The amount of reacted
biotinyl-tyramide is proportional to the amount of HRP in the well. This amplification step
effectively multiplies the quantity of HRP molecules for the final TMB substrate reaction.

Easy one-hour protocol. InstantOne ELISA, the one-hour, one-wash ELISA for cell
signaling, detects total and phosphorylated targets with an easy-to-use assay that results
in sensitivity that exceeds industry standards.
The one-hour, one-wash phospho ELISA
InstantOne ELISAs use the traditional sandwich ELISA format with some significant
differences that allow for greater flexibility, ease of use, reduced assay time, and reduced
hands-on assay time. Unlike a typical sandwich ELISA, the target analyte binds to the two
sandwich ELISA antibodies in solution. As the capture antibody binds to the plate through
a proprietary mechanism, both the sample and the assay reagents may be added to the
InstantOne ELISA assay plate at the same time. Unbound assay reagents and non-specific
sample components are washed away (just as in a traditional sandwich ELISA), while the
specific analyte is detected through a colorimetric detection. The entire process takes a
little more than one hour to complete. Analysis of both total and phosphorylated targets
across family members or down pathways on the same plate has never been easier.
In addition to the simplicity the InstantOne ELISA provides, it also adds a layer of flexibility
not readily accessible with traditional sandwich ELISAs. Since the antibodies are not pre-
coated in the wells, several different targets can be analyzed simultaneously on the same
plate in different wells. Simply add the sample lysate to the plate wells and add different
antibody reagent cocktails to the different wells.

ProcartaPlex Multiplex Immunoassays use the Luminex xMAP (multi-analyte profiling)
technology that enables the simultaneous detection and quantitation of up to 80 protein
targets in a single 25–50 µL sample of plasma, serum, cell culture supernatants and other

ProcartaPlex® technology overview
xMAP technology ProcartaPlex assays use xMAP magnetic bead technology that uses the Magplex™
superparamagnetic 6.5-micron microsphere beads with a magnetic core. The beads
are internally dyed with precise proportions of red and infrared fluorophores. Varying
proportions of the red and infrared fluorophores result in 100 spectrally unique
microspheres, which are identified by the Luminex xMAP detection systems. The
conjugation of a distinct monoclonal antibody to a distinct bead allows for analysis of
multiple analytes in a single well.
Luminex instrumentation Luminex instrumentation is comprised of three instrument models: MAGPIX,
Luminex100/200 and FLEXMAP 3D. These instruments are based on either the principles
of quantitative fluorescent microscopy or fluorescent flow cytometry. The multiplex
quantitation capability of an assay is dependent upon instrument model and ranges from
50 analytes with the use of the MAGPIX, to 500 analytes with the FLEXMAP 3D. Used in
combination with ProcartaPlex assays, the Luminex platform delivers fast and cost-effective

Why ProcartaPlex assays?
Flexibility
Get exactly the panel you want because more than than
90% of ProcartaPlex assays are combinable with one
another. ProcartaPlex assays allow the creation of highly
individual and high-plex panels. Affymetrix currently offers
the largest published panel with a 63-plex.1,2,3,4
Reproducible results
ProcartaPlex assays are based on the principles of sandwich
ELISA where two highly specific antibodies recognize
different epitopes on the same protein. Reproducible
results come from reliable manufacturing. Affymetrix
owns and operates a world-class hybridoma facility for the
manufacturing of antibodies, and these antibodies are used
in ProcartaPlex assays, enabling the delivery of consistent,
reproducible, and high-quality assays.
Correlation to ELISA
Switch easily from ProcartaPlex assays to ELISA and vice-
versa with reliable results. A majority of our ProcartaPlex
assays use the same antibody pairs as our Platinum
ELISA, resulting in high correlation (R2>0.9) between
the two assays. As shown in Figure 1, equivalent results
(R2>0.9) were obtained using the ProcartaPlex Multiplex
Immunoassays and traditional plate-based ELISAs for
human and mouse IFN gamma. ProcartaPlex assays achieve
quantitative results comparable to traditional ELISA results
with the added benefits of reduced sample volume, shorter
assay time, and lower cost.
Results of an experiment measuring IFN gamma and TNF alpha in stimulated human and mouse PBMCs. Supernatants were serially diluted two-fold in normal human or mouse serum, to allow for differential cytokine levels in a serum matrix. Additional cytokines for human and mouse analytes were compared including IL-beta and IL-17A, and similar performance was obtained (data not shown).

ScalabilityThe ability to scale the number of analytes investigated in multiplex experiments is critical for the progression of many projects. For example, it is not uncommon to begin a project by analyzing a large, comprehensive 35-plex cytokine panel on fewer samples (i.e., 100) to determine which analytes are affected by a particular disease or drug treatment. The resulting list of analytes can then be investigated further, for instance, using an 11-plex panel, on a larger set of samples. Thus, there is a great need for scalability in multiplex panel assays in order to enable researchers to correlate all their data across the different stages of their investigation. Many commercially available multiplex assays have not been developed with this level of consistency in mind. However, this specification is a key design requirement incorporated during the ProcartaPlex assay development. Data in Figures 2 and 3 illustrate the high correlation between similar analytes in a large and small panel and the scalability when using ProcartaPlex assays. Similar results are obtained when investigating other analytes.

High Sensitivity ProcartaPlex Immunoassays High Sensitivity ProcartaPlex Immunoassays offer accurate quantification of key cytokines significantly below the detection limit of conventional Luminex assays.
High Sensitivity ProcartaPlex Immunoassays enable the multiplexed analysis of human and mouse analytes in very low
concentrations in various matrices including serum, plasma, bodily fluids, and cell culture supernatant samples. These high-
sensitivity multiplex assays are ideal when sample input is limited, and protein expression is in the low pg/mL range.
Accurate quantification below conventional limits is achieved through a proprietary signal amplification technology. The signal
amplification has negligible effects on the assay background, thereby increasing the signal-to-noise ratio of the cytokine
standards on the lower end of the standard curve. This also allows for more accurate and reproducible measurement of
cytokines with low expression levels.
All analytes listed above are also available as Simplex Kits. Additional ProcartaPlex® High Sensitivity Basic Kit that includes all non-target specific reagents is needed to perform assay.

Platinum ProcartaPlex® Immunoassays
Platinum ProcartaPlex Immunoassays are our most validated bead-based multiplexing assay system. Similar to our Platinum ELISAs, Platinum ProcartaPlex assays have to pass more than 30 qualification criteria to guarantee reproducible high performance of the kits.
Overcome matrix effects
Serum and plasma samples are complex matrixes with many components. This complexity can cause the “matrix effect” which
can impact the readout of many cytokines (low spike and dilution recovery). Affymetrix developed matrix-type specific diluents
to overcome these matrix and multiplex effects, assuring high-performance specifications comparable to those of traditional
ELISA. Spike and dilution recovery in a range between 70–130% are obtainable with Platinum ProcartaPlex assays.
100% combinable
Platinum ProcartaPlex Immunoassays are available in premixed panels and simplex kits. All Platinum ProcartaPlex Immunoassays are combinable with one another, eliminating any limitations on composition and size of your ProcartaPlex panel. You can select between two off the shelf panels or compile your own custom panel.

Dilution and spike recovery were evaluated in a minimum of five individual donor samples per matrix by performing 4-fold serial dilutions, from 1:4 to 1:256. Mean recoveries for each matrix were calculated across the complete portfolio of Platinum ProcartaPlex® assays, with a target specification of 100% +/–30%.

ELISA Troubleshooting
Problem Solutions
High Background Improper washing
Insufficient washing or omission of one of the wash steps can lead to high background. Check volume of ELISA wash buffer and make sure all recommended washing steps are performed. Make sure the eBioscience ELISA wash buffer is used and diluted correctly.
Leave the ELISA wash buffer in the wells for at least 15–30 seconds.
Contaminated substrate
Make sure there is no contamination of the substrate before using with metal ions or oxidizing reagents. The substrate solution should not develop color by itself.
Contaminated diluent
Most of the ELISA diluents are based on a protein matrix. When first opening, contamination of the ELISA diluent may occur. Use only sterilized pipettes to pipette ELISA diluent from the bottle.
Substrate exposed to light
Exposure to light may result in a blue color of the substrate. Keep solutions in the dark (vial) until it is ready to be dispensed on the plate.
Wrong conjugate dilution
Make sure the conjugate is diluted according to the instruction in the manual. Too high concentration of the conjugate may result in high background.
Wrong incubation times/temperatures
Adhere to the instructions in the ELISA test protocol regarding incubation times and temperature.
High Intra-CV Coating removed by scratching during washing/pipetting
Take care not to scratch the inner surface of the microwells when washing the plate or pipetting standards and samples.
Inhomogeneous samples
Make sure the samples you use for the test are homogeneous. Mix before adding to the ELISA plate.
Edge effect
Avoid incubating plates in areas where environmental conditions vary (e.g., under a vent or window sill). Seal the ELISA plate during incubation and incubate on a rotator set at 400 rpm, if possible.
Do not stack the plates during incubation.
Pipettes not properly calibrated
Make sure the pipettes used are properly calibrated.
Improper washing
Insufficient washing or omission of one of the wash steps can lead to high CV values.
Do not reduce wash volume or the number of wash steps. Leave the ELISA wash buffer in the wells for 15–30 seconds.

Poor Standard Curve Wrong reconstitution volume of lyophilized standard
Make sure the lyophilized powder is reconstituted in the correct volume of the correct reconstitution media. Reconstitution volumes are given in the manual, on the vial label, or both.
Reconstitution time too short
Make sure the reconstitution process is given enough time to completely solubilize the lyophilized powder.
Wrong dilution of concentrated stock standard
Make sure the concentrated stock standard provided is diluted properly according the manual.
Wrong storage of stock material/pre-dilutions
Storage conditions of stock standard solution as well as pre-dilutions are given in the manual. Follow these recommended guidelines.
Improper mixing at standard dilution steps
Prepare serial dilutions of the concentrated standard and mix properly at each step of dilution.
Wrong measurement wave length
Make sure the photometer used is adjusted to the correct wavelength. Please check for the reference wavelength.
Standard from a different lot used
Only use the standard(s) provided in the kit to perform assay. Do not substitute the standard from another kit.
Low OD Values Incubation time with the TMB substrate is too short
The time indicated in the test protocol may vary due to temperature or other assay conditions (temperature, kit age, humidity).
Observe the color development on the plate and use your own judgment to stop the substrate reaction. We recommend adding the ELISA stop solution when the highest standard has developed a dark blue color.
Alternatively you may monitor the color development with the ELISA reader at 620 nm after adding the TMB substrate solution. The substrate reaction should be stopped (with ELISA stop solution) as soon as an OD of 0.9–0.95 is reached.
Wrong reconstitution volume of lyophilized ELISA standard
Reconstitute with the media and volume specified in manual and/or on vial label.
Make sure the reconstitution process is given enough time to allow complete solubilizing of the lyophilized powder.
Shaking
Make sure that assay plate(s) are shaken at the indicated speed on a plate shaker during designated incubation periods. Not shaking the assay plate(s) during these noted periods result in reduced OD values.

No Color Development Incorrect storage of ELISA reagents
Storage conditions of all ELISA kit components are given in the manual—always follow these guidelines.
Omission of any incubation step
Make sure each step of the ELISA test protocol is performed as described. Addition of the colored reagents (green-/blue-/red-dye) in the Platinum ELISA Kits helps ensure that the assay protocol is followed as designed.
Microwells dried out after washing or during storage
After washing, tap ELISA microwell strips on absorbent pad or paper towel to remove excess ELISA wash buffer. Use the microwell strips immediately after washing or place upside down on a wet, absorbent paper towel for no longer than 15 minutes.
Samples not suitable for the ELISA test
A list of samples suitable for the ELISA test is provided in the manual. Other samples may not be suitable for the ELISA test.
ELISA Samples or Standard in the Overflow
Wrong dilution of sample/standard
Follow the test protocol for dilution of standard and samples.
Wrong diluent used
Only use the diluent delivered with the kit to dilute samples.
Wrong measurement wavelength
Make sure the photometer used is adjusted to the correct wavelength. Please check for the reference wavelength in the manual.
Development time of TMB substrate too long
The time indicated in the test protocol may vary due to temperature or other assay conditions (temperature, kit age, humidity).
Observe the color development on the plate and use your own judgment to stop the substrate reaction. We recommend adding the stop solution when the highest standard has developed a dark blue color.
Alternatively, you may monitor the color development with the ELISA reader at 620 nm after adding the TMB substrate solution. The substrate reaction should be stopped (with stop solution) as soon as an OD of 0.6–0.65 is reached.
Sample not suitable for the test
A list of samples suitable for the test is given in the manual. Any other sample may not be suitable for the test.
Wrong incubation time/temperature
Follow the test protocol regarding incubation time and temperature.
Plate covers were not applied during incubation
Application of plate covers during incubation prevents evaporation of liquid as well as contamination during incubation. Plate covers are supplied with each kit. Use a new plate sealer for each incubation step.
