Part 4 - Calibration and Measurements
Measuring Objects
Groups of pixels together make objects
From Outlines we can get measurements:
Perimeter
Area
Bounding box
Centre coordinates
Radii
Much, much more...
Calibrations
Calibrations make sense of measurement
Transforms distances from Pixels to real world units
All images must be calibrated for spatial measurements
40µm
Creating Calibrations
Use known values to estimate
•Cal = Sampling Rate
Instrument Mag
Or, take image of reference object
Creating Calibration
1.Take image of known sample
2.Go to Capture Ribbon
3.Select Quick Calibration tool
4.Drag line over known distance
5.Select units and qualify line
Calibrating Images
From Capture Ribbon
Select Calibration from Characteristics box
New automatically assume current charactristics calibrations
Use green tick mark next to Spatial drop-down for image with no or incorrect calibration
Scale Bars
Calibrations enable Scale Bars and Measurements
Enable Scale Bars from Calibration group
Use Auto Marker to apply markers to every image
Use Calibration drop-down to access Marker properties
1mm1mm1mm1mm
Manual Measurements
Use Measure ribbon for manual measurements
Use Types button to select what values to collect
Use drop-downs for additional tools (e.g. straight line Vs poly-line)
Display Option
Use Options button to change:
Outline styles
Label option
Display Option
Show statistics in Measurement table
Use groupings to arrange object data by class
Change grouping options to display class statistics
Exporting Data
Export Images using:
Quick Save in Capture ribbon
Share ribbon
Export any data table to Excel, Text files or Clipboard
Measurements
Manual Measurements are a powerful tool for getting information from an image but...
...Manual measurements are also:
slow & tedious
open to operator bias
