Mobile Phase Filtration

Why is mobile phase filtration important? Unfiltered mobile phase may contain particulates and bacterial

contaminants that can build up and create a chain reaction of

system and column problems. Check valves, pump seals and

the injector may be damaged by particulates passing through

the system. Once these particulates reach your column, the

buildup will result in pressure increases, causing your system

to work harder, or even shut down. Trace amounts of contami-

nants can also produce split peaks or extraneous unidentifi-

able peaks, leading to unacceptable results.

How does mobile phase filtration work?There are two solutions available to transfer solvents for filtration.

vent from the source container into the filtration system

filtration

With either solution, solvents are passed through a 47 mm di-

ameter Phenex™ filter membrane which is placed between the

FilterSys™ mobile filtration system’s fritted glass support base

and the sample reservoir. The filtered, particulate-free solvents

are then stored in plasticizer-free glass reservoirs for delivery

to the chromatographic system. When the filtered solutions are

ready for use, solvent inlet filters remove any buffer that may

have precipitated or any bacterial growth that has occurred

before they enter the system.

Together, this system allows for the rapid filtration of particu-

late matter along with the removal of undissolved buffers from

your solvents.

How does sample preparation work?

Above syringe filters are non-sterile. Housing is made of medical-grade polypropylene (PP). Luer lock inlet/slip outlet connections unless otherwise indicated.1. 17 mm diameter. 2. Glass fiber filters are 28 mm diameter and made of borosilicate. They will remove 90 % of all particles > 1.2 µm. 3. Housing material is methacrylate butadiene styrene (MBS) polymerisate. Phenex™ disposable centrifugal filter units provide a convenient means of filtering multiple HPLC and GC samples without the necessity of manually forcing each sample through the filter with a syringe. All you need is a laboratory centrifuge with sufficient capacity for the number of samples to be filtered at one time. Centrifugal force drives the sample through the filter quickly without effort on the part of the chemist.

Samples contact only the Phenex filter unit itself and the polypropylene receiver tube. The upper portion of the tube containing the membrane filter can be discarded. The receiver tube serves as a container for the filtered sample and can be retained as long as desired. Separate caps are supplied for the sample chamber and the receiver tube.

Filter membrane materials available are Nylon 66, PTFE (Teflon®) and Cellulose Acetate (CA). Nylon 66 Phenex filter units are recommended for most HPLC and GC sample filtration work. They are chemically resistant, compatible with all commonly used solvents, and virtually free of extravctable materials that could contaminate samples. PTFE (Teflon®) Phenex filters are designed to handle more aggressive organic solvents, whereas Cellulose Acetate (CA) is especially well-suited for the filtration of proteinaceous samples in aqueous solvents.

Why is column protection important?Column protection systems reduce the amount of sample

and mobile phase contaminants from building up at the head

of the column, which can otherwise degrade chromatography,

increase system backpressure, and shorten column lifetimes.

Protection systems also safeguard HPLC columns and detec-

tors from damaging pump and rotor seal particulates.

How does column protection work?Column protection systems protect your HPLC columns and

detectors from microparticulates in the sample, solvent, or

wear on the polymeric seals of the pump or injector. These

protection products trap particulates before they can reach the

column or detector. Contaminants can clog frits, irreversibly

bind to column packings, degrade analytical performance, and

even damage expensive detector flow cells.

There are two types:

•TheSecurityGuard™ system protects the HPLC column

and detector by trapping both particulates and chemical

contaminants with inexpensive disposable cartridges.

Short cartridge lengths protect your HPLC columns

without altering chromatography.

• In-linefiltersremovefineparticulatesfromthesolvent

stream. They also remove microparticulates as small as

0.5 µm before they clog your column and degrade your

separation.

Stainless Steel (Analytical)

The 1.5 mm diameter filter is recommended for use with smaller diameter columns, microbore columns, and very short columns for fast HPLC procedures. The 3 mm diameter filter element is recommended for use with conventional 4.6 mm diameter columns.

KrudKatcher™ Classic PeeK or Biocompatible (Analytical)

Fit pressure rated to 5000 psi. The PEEK® filter body houses an integrated 0.5 µm HiFlo™ 316 stainless steel depth filter that efficiently removes microparticulates from the flow stream without contributing to system dead volume (2 µL). Fits columns 1.0 to 4.6 mm ID.

Stainless Steel (PreP)

The 21.2 mm diameter filter is recommended for use with 18-30 mm diameter columns and is pressure rated to 8,000 psi (551 bar).

Biocompatible (SemiPrep)

The biocompatible filter consists of a stainless steel body, two PEEK® end-fittings, and a separate replaceable PEEK® frit. This filter unit can be placed in the flow path before or after the column with little or no effect on peak shape. Pressure rated to 6,000 psi (413 bar) or columns 6 to 16 mm ID.
