This manual follows certain typographical conventions in its descriptions of the software NANOCOLOR® Photometer Data Export and how to use it with your photometer. These conventions are summarized below. Keys you have to press on your computers keyboard or on the photometer keyboard are printed in bold capital letters like the ENTER key. Program or data paths, folder names and file names are printed in italic letters like in the standard program path c:\programs\macherey_nagel\dataexporte. Other software that is necessary or can be used together with this software like MS EXCEL is printed in capital letters. Buttons, switches, text fields of the program NANOCOLOR ® Photometer Data Export are printed italic and marked with a frame like the Measure button. Menu commands like File/open of the program NANOCOLOR ® Photometer Data Export are printed italic and marked with a grey background.

1. System requirements The program NANOCOLOR® Photometer Data Export V2.5 requires at least a Pentium I (>100 MHz) processor, 8 MB free hard disk memory, 64 MB RAM, Windows 98, Windows 2000 or XP. You need a free serial interface (RS 232) on your computer, USB-to-RS 232 adapters can be used alternatively. To create MS EXCEL worksheets you have to install a legal copy of Microsoft MS OFFICE 2000/2003 on your PC. To connect the photometer with the PC you need a serial cable (zero modem cable). The program is optimized for a screen resolution of 1024 x 768 pixels. For program installation your PC needs a CD ROM drive.

MS EXCEL 2000 / MS OFFICE 2000/2003, the serial cable, USB cable and a USB-RS 232 adapter are not part of this product.

2. Supported photometers The program NANOCOLOR® Photometer Data Export V2.5 is suitable for the MACHEREY-NAGEL photometers NANOCOLOR® 250 D, 300 D, 350 D, 400 D, 500 D, LINUS and for the luminometer BIOFIX® Lumi-10. All MACHEREY-NAGEL photometers can work with different COM-Port settings. SDF = semicolon delimited fields The format of some data fields like date, time and the decimal point depends on your computers country setting and / or on the setting selected in your photometer.

3. Software installation

To install software on a WINDOWS NT / WINDOWS 2000 / XP system you must have administrator rights! If you have already installed a previous program version you have to de-install the old software before you install the new version! Note: If you use MS ACCESS databases in a previous version save your database before de-installing the old version!

Please insert the delivered CD ROM into your CD drive. Usually the installation process starts automatically. If not, open your Windows Explorer and click on the drive symbol of your CD ROM. Start the program setup.exe by double click. The program will not install if you do not accept our software license contract, see figure 1. Please follow the instructions of the installation program.

If this software comes with the CD Photometer NANOCOLOR® 500 D Software – CD the installation process starts not automatically. Please click double on the file setup.exe in the folder dataexport_e of your Photometer NANOCOLOR® 500 D Software – CD.

The standard program folder used by NANOCOLOR® Data Export is

c:\programs\MACHEREY_NAGEL\ dataexporte

We recommend not to change the path or folder.

4. Start of the program Connect your photometer with your computer. Switch the photometer on. Click on Start\Programs\ MACHEREY_NAGEL\Photometer Data Export. The first window (see figure 2) of the program NANOCOLOR® Photometer Data Export V2.5 shows in the upper part four buttons for the program settings and to choose your photometer. You can start all button functions also by using menu commands. The selected settings are shown in the lower left part of the window. If you start the program the first time, these settings are usually not suitable for your system/photometer. The program can not detect the type of photometer auto-matically – accept the NANOCOLOR® 500 D. Figure 2 shows the main window of the program.

5. Device selection / Find COM-Port To communicate with your computer your photometer must be in the “Methode” mode. The photometer must display “Method:” and “cuvette”. Please click on the button Select photometer (or in the program menu on Settings/Photometer). The program opens a new window (see figure 3) where you can select your photometer and used COM-Port:

Fig. 3: Photometer selection Select the COM-Port you used to connect your photometer in the upper part of this window. Click on the button that corresponds to your photometer. All settings used for the data transfer are set automatically. The program will save the COM-Port settings. The switches 500 D with RS 232 and 500 D with USB are visible only if the NANOCOLOR® 500 D is selected. Please select here the connection you are using. The button Search Inteface / COM-Port is only displayed if the NANOCOLOR® 500 D, or 400 D is selected. If you do not know the port number you can search for the connected photometer (this function works on 400 D photometers beginning with firmware 3.0). You do not have to set up the COM-Port again until you change your COM-Port or your photometer. If your settings are correct click on Save settings.

6. Functions of the NANOCOLOR® Data Export Software The program NANOCOLOR® Photometer Data Export offers you the possibility to: - transmit data from the photometer memory into MS EXCEL (offline mode)

- transmit data from the photometer memory into MS ACCESS (offline mode)

- transmit directly every measurement into MSD EXCEL (online mode)

- create calibration curves with all necessary statistical parameters

To create MS EXCEL worksheets you need a legal copy of MICROSOFT MS EXCEL version MS EXCEL 2000 or 2003. To create ACCESS databases you do not need any version of MICROSOFT ACCESS. This function is part of the program NANOCOLOR® Photometer Data Export. But if you want to open ACCESS databases you need MICROSOFT ACCESS version 2000 or 2003 (depending on the type you have created)!

The different functions are described in the next chapters. 7. Select Export Mode Beginning with version 2.0 the program offers two different export modes. The OFFLINE mode reads the whole photometer memory after your measurements and stores the records into MS EXCEL or ACCESS. The ONLINE mode reads every single data during your measurements. The received records are stored into an MS EXCEL worksheet. This function does not work with the luminometer LUMI-10. To change the export mode please click on the menu function Settings/Run Online or Settings/Run Offline.

8. Offline Data Export Please ensure that the mode Run offline is selected (see chapter 7.)

The picture showing the moving bits symbolizes the present data transmission. Possible errors that occure during transmission are displayed in the text field. Product line NANOCOLOR® 250 D, 300 D, 350 D, 400 D, 500 D Start the export process in the program window by clicking on the button Read data offline. With this photometers the export process starts and ends automatically. The program detects the end of the transferred table and starts the MS EXCEL or ACCESS application. NANOCOLOR® LINUS The hardware of this photometer does not transfer a "end of table" signal to the computer. Because of this you have to stop the export process manually. To get data form this photometer, proceed as follows: Start the export process in the program window by clicking on the button Read data offline. Switch the photometer into the memory modus (key Mem. on the

LINUS). The LCD display shows the text "Memory:" and "print?". Press the OK key on your photometer. The data export starts now. After the end of the transfer process the LCD display flashes for a short time and you can read "Memory:" and "print?" again. The transfer process is complete and you can stop the transfer process on your computer. Just press the key q or Q on you PC keyboard. BioFix Lumi-10 Start the export process in the program window by clicking on the button Read data offline. Switch the luminometer Lumi-10 on and select the command Results / PC from the LCD display. Then select the command start transfer. The LCD display changes and "data transfer" is displayed. After this message disappears press the q key on your computers keyboard. Usually the export process takes about 1 to 5 minutes depending on the size of the transferred data. The NANOCOLOR® 250 D and 300 D are quite old and slow. If their memory is full, the export process may take up to 10 minutes. The Lumi-10 can store up to 1000 data sets. This export without preselection may take more than 5 minutes. 9. Data Export from the photometer memory into MS EXCEL

(offline mode) To communicate with your computer your photometer must be in the “Methode” mode. The photometer must display “Method:” and “cuvette”. Please click on the button Export into of the start window of the program NANOCOLOR® Data Export V2.5 (see figure 4 on page 8). Depending on the settings of your program one of three possible windows is displayed (fig. 5, fig. 6 or fig. 8).

If you enable the function Divide table after sample place the program creates for every sample place number in your photometers data a single MS EXCEL table. If the function is disabled the program stores all data into one table. The program NANOCOLOR® Data Export V2.5 handles up to 20 different sample places (see chapter 12).

Acknowledge your setting with the button Save settings, if you click on the Back button, the window closes without saving your changes. You activate the data export by clicking on the button Read data offline. The program NANOCOLOR® Data Export V2.5 creates a new MS EXCEL workbook. The name of your photometer is used as file name, e. g. 500_D.xls. If such a filename exits, the software asks you if you want to create a new, empty worksheet or if you prefer to append your data to the existing file. If you create a new worksheet, the old file is stored with the new extension old, e. g. 500_D.xls.old. 10. Data Export from the photometer memory into MS ACCESS

(offline mode) Please click on the button Export into of the start window of the program NANOCOLOR® Data Export V2.5 (see figure 4 on page 8). Depending on the settings of your program one of three possible windows is displayed (fig. 5, fig. 6 or fig. 8).

Select the setting create new ACCESS database . Please select the version of the MS ACCESS database the program has to create from the option field Database version. To create a new database, you have to enter the path, folder and file name. Click on the drive list and choose the correct drive. Choose your data folder from the folder list. Enter the file name you want to use for your new database into the text field. The file names extension must be mdb. We recommend not to use spaces in file or folder names. To enable the program NANOCOLOR® Photometer Data Export to start MS ACCESS automatically, you have to save the path to the ACCESS application once. Please click on the button Path to MICROSOFT ACCESS. The program shows the next window (see figure 7):

Fig. 6 : ACCESS path settings Open the drive list and select the correct drive. Select the correct program folder in the folder list. Select the ACCESS application file in the file list. The usual (standard) path for MICROSOFT ACCESS is c:\programs\Microsoft MS OFFICE\MS OFFICE11\msaccess.exe. If you have not a legal copy of MS ACCESS installed on your computer please mark the option button MS ACCESS is not installed. Acknowledge your setting with the OK button.

You activate the data export by clicking on the button Read data offline. If you want to append your data to an existing database, please select the setting append to ACCESS database.

11. Direct data transmission during measurement (online mode) Please ensure that the mode Run offline is selected (see chapter 7.)

Fig. 9: Run Export Online In the upper frame of the window you can activate the function Print Online Measurement Protocol. Please enter the path to the MS EXCEL worksheets in the frame Data path. If you do not change the value of this frame the program stores the MS EXCEL worksheet into the subfolder \work of the program installation folder as output.xls. The function Print Online Measurement Protocol prints a protocol containing date, time, method, value etc. on the standard printer of your computer. You can add further information like sample numbers and sample place descriptions to the protocol. The second option button Divide table after sample place selects if the program stores all data into one table or if the program creates one worksheet for every sample place. If the function Print Online Measurement Protocol is activated in the Run Online window, the program will ask for the operator name and sample descriptions. Figure 10 shows the corresponding window.

Fig. 10: Print Online Measurement Protocol Enter the name of the operator in the field Operator. This field must be filled out if the protocol function is activated. The second field Sample number or description may be filled with numerical or alphanumerical sample descriptions. Enter one line for each sample. If your computer is equipped with a BAR code reader and the cursor is in this field you enter sample numbers with the reader. If this field contains more descriptions than measured samples, superfluous descriptions are ignored. If it contains less descriptions, the program uses three minus (---) as sample description.

This sample description does not replace the sample number of your photometer.

The protocol contains date and time information from your computer and from your photometer. If these data differ you must adjust the computers and / or photometers clock.

Acknowledge your setting with the button OK, if you click on the Back button, the window closes without saving your changes. You activate the data export by clicking on the button Read data online. At the first time this function is used the program opens a new, empty MS EXCEL worksheet. The program waits now for measurement data. Every measurement (except the zero-values) are transmitted to your computer and stored in the MS EXCEL worksheet. Note: Please switch on the photometer first before you start the online export.

Beginning with the second call of this function the program searches for MS EXCEL worksheets existing in the folder \Work (or in the specified folder). If it finds a worksheet the program appends new data into the existing worksheet. If it finds no worksheet it will create a new one from a template.

The program does not save the created MS EXCEL worksheet. Every user takes the responsibility for the storage of his data with MS EXCEL.

12. Determining Sample Places The MACHEREY-NAGEL photometers – except the 500 D - create sample place numbers (from 00 to 99) for every measurement. During measurement you can set this number (please refer to the photometers manual). You can create clear text values for these numbers in the NANOCOLOR® Data Export Software. If you have stored sample place names the program replaces the sample place numbers in every measurement record with the names before writing the data into MS EXCEL or ACCESS. For example, the program replaces all sample place number "01" with "Inlet 1". To use sample place names instead of numbers click on the menu function Settings/Sample place. The program opens the Sample Place Window (see figure 11).

Then type a name into the text line on the right side. Select a new number and so on. If you have entered all necessary names please click on the Save button.

The settings of this software are ignored if the photometer NANOCOLOR® 500 D is used. The menu command Sample place is disabled if a NANOCOLOR® 500 D is selected.

If you have enabled the function Divide Table after Sample place (see page 9) the sample place names are used as MS EXCEL worksheet labels. 13. Data pre-selection Beside the possibility to read all photometer data you can select special data from the memory output. Please click on the button Data selection or use the menu command File/Data selection. Figure 12 shows the Select data window. This function does not work in online mode.

In the window shown above you can perform the following selections from the photometer memory: a) only sample place: The export program returns only values that correspond to the

entered sampling location (enter the place as a two digit number, as you did with the photometer keys, e.g. 03, or up to 12 characters if you use a NANOCOLOR® 500 D).

b) only method: Select one method number from the drop-down list. Only values

that correspond to this number are exported. If you work with special methods (methods that you have programmed) you have to enter the method number with a starting "P" into the text field Special method . For example, enter P03.

c) Measurements from one day: Enter the date of this day in the format dd.mm.yy, e.g. 13.01.02, into the text field only from (date)

d) Measurements over several days: Enter the start date into the text field only from (date) and the final date into the field until (date) . 14. Method calibration You will find the new calibration function in the main menu under the function Method calibration. This function does not depend on the selected export mode. After clicking on this function the program opens the Calibration Window (see figure 13).

Fig. 13: Method calibration window The text field Operator, Method name / Number, Wavelength and Unit are only for information purposes and not necessary for a calibration. If you fill out these information fields they are printed into the calibration protocol if you click on the button Print.

If the parameter F0 is lower than 3% of the highest measured value the program supposes that the curve runs through (0,0). If the regression grade is higher than 1 the curve is calculated without F0 parameter. If F0 is grater than 3 % if the highest value it is used for regression.

If you use the extinction method (method 903) the photometer requires a zero calibration. Ensure to set the correct kind of zero calibration solution: against water or against reagent blind value. If reagent blind value is set, the program supposes that the regression curve must run through (0,0) even if the parameter F0 is greater than 3% if the highest value.

How to create a calibration curve Select the number of your used standards in the list Number of points (minimum 5; 10 or more should be the rule) and mark the used kind of the zero-standard on the right side of the program window. Now click on the text field Measured value. Enter the set point of your first standard (30 mg in the example). After typing the ENTER key the value is transferred into the table and the field Measured value is cleared. Enter now the second value (60 mg/l in the example) and type enter. The program completes your table automatically with equidistant values. If these values are OK, please enter two times the ENTER key or click on the Measure button to start the measurement. If you do not need equidistant values DO NOT type the ENTER key into the empty filed Measured value but simply enter your third value. If this value is not equidistant to the last value pairs the program clears all equidistant values and waits for your input. Any typing mistake can be corrected simply by clicking on the wrong value but only after the complete table is filled. Click on the wrong value and the program opens a window that enables you to enter a new value or to measure again. After clicking on the button Measure the program waits for data from your photometer. Select the method EXTINCTION (code 903) on your photometer and select the necessary filter wavelength. Insert the zero-value cuvette into your photometer and press the Null/Zero key on your photometer. Now enter each of your standard solutions in the order of the table from top to down into the photometer and press the M key. After measuring the last value the program calculates a linear regression. The option buttons linear, quadratic, cubic and 4th grade enables you to change the kind of regression analysis. The program calculates the parameters F0 to F4 of the selected compensating curve. You can use this parameters to program methods into your photometer. The program calculates variances, correlation coefficients, standard deviations, sensitivity, method standard deviations and method variation coefficients. Use the button Print to get a complete calibration protocol. If you have used one of your cuvettes in the wrong order, simply click on the wrong value in the table. The program offers you now the possibility to measure this value again.

If the calibration curve runs not through zero, the software calculates an offset value. With this value you can correct the measured extinction values in order to get a curve with the same slope that runs through zero. The offset value is displayed only in the printed measure protocol. Figure 13 shows the relation between offset value and F0.

Applicable Law ,Jurisdiction and General Provisions This Agreement will be governed by and construed in accordance with the laws of the Federal Republic of Germany whose courts shall have exclusive jurisdiction over all disputes relating to this Agreement. If any part of this Agreement is found void and unenforceable, it will not affect the validity of the balance of the Agreement, which shall remain void and enforceable according to its terms. This Agreement shall automatically terminate upon failure by you to comply with ist terms, in which event you must destroy all copies of the software.
