Pure Derivatives of Glycyrrhiza for CosmeticsMechanism of Inflammation:

Ø Topical inflammation occurs when cells are injured by factors such as the following:

• Environment (the drying wind, the scorching sun, etc.)• Hypersensitivity to certain compounds• Application of harsh chemicals to the skin such as deodorants or

permanent wave solutions.

Ø The injured cells release chemicals that attract white blood cells to phagocytize the irritants such as prostaglandin E2.

Ø Prostaglandin E2 is produced from the cyclooxygenase pathway of a complex set of chemicals reactions collectively called the arachidonic acid cascade.

Ø The arachidonic acid cascade is responsible for producing many inflammatory mediators as well as compounds that cause allergic and hypersensitivity reaction through the 5-lypoxigenase pathway.

Ø Glycyrrhizin inhibits prostaglandin E2 production..

Ø By incubating activated macrophages with glycyrrhizin, which produce prostaglandins E2 though the same chemical pathways as skin cells do, there is a decrease in prostaglandin E2 production.

Ø Prostaglandin E2 production is further inhibited the longer the glycyrrhizin is incubated with the activated macrophages.

Ø The longer the application of licorice products, the more anti-inflammatory activity it excerts.




Pure Derivatives of Glycyrrhiza for CosmeticsAnti-Allergic and Hypersensitivity Effects:

Ø Topical allergic and hypersensitivity reactions caused by the arachidonic acid cascade are also inhibited by licorice products.

Ø Glycyrrhizin decreases vascular permeability by estimating the leakage of dye from a passive anaphylactic reaction in rat conjunctiva.

Ø Allergic conjunctivitis is mediated by causing an anaphylactic (hypersensitivity) reaction in the eye, thereby, increasing vascular permeability and causing dye leakage.

Ø Comparing the inhibitory effects of glycyrrhizin to l-epinephrine bitartrate, and dexamethasone, shows that glycyrrhizin has a definite effect in inhibiting allergic reactions yet has subtle effects compared to a prescription drug such as dexamethasone.

Ø Based on similar studies various eye drop products have been developed containing glycyrrhizin to soothe irritated eyes.

Ø Glycyrrhizin can also be formulated in eye products such as eye shadows, mascara, and eye make up removers to reduce the hypersensitivity they sometime causes.




Pure Derivatives of Glycyrrhiza for CosmeticsPhospholipase A2 Inhibition:

Ø The direct inhibition of phospholipase A2, which is responsible for initiating the arachidonic acid cascade and producing inflammatory mediators and products causing allergic and hypersensitive reactions, is one of the anti-inflammatory pathways by which glycyrrhizin excerts its activity.

Ø Inhibiting arachidonic acid release prevents it from being a substrate for either the cyclooxygenase enzyme (pathway which produces inflammation mediators) or the 5-lypoxygenase enzyme (pathway produces compounds that cause allergy and hypersensitivity reactions).

Ø By measuring the effect of glycyrrhizin on the phospholipase A2activity mimicking a cell membrane with an artificial unilammellar liposome containing carboxyfluroescin (CF) (the greater the CF release, the greater the phospholipase A2 activity), glycyrrhizin was found to inhibit phospholipase A2 activity.

Ø Licorice products excert anti-inflammatory activity by indirectly inhibiting phospholipase A2 by potentiating glucocorticoids such as hydrocortisone.

Ø Glycyrrhetinic acid is a potent inhibitor of the enzyme 11 beta-hydroxysteroid dehydrogenase (OHSD) which metabolizes the active hydrocortisone to inactive cortisone.

Ø By inhibiting the OHSD found in skin, glycyrrhetinic acid increases the half life of hydrocortisone to cause an anti-inflammatory action.

Enhance Emulsion Stability:

Ø Monoammonium Glycyrrhizinate (MAG) and Dipotassium Glycyrrhizinate(DPG) decrease interfacial tension thus they are surface active.

Ø They aid to stabilize emulsion and foams.

Ø The critical micelle concentration is approximately 2-3X10-4 mol/L

Safety Records:Ø Centuries of use have proven glycyrrhizic acid compounds to be safe

and effective.

Ø Dermal toxicity studies on rabbits showed no primary irritation.

Ø Subacute toxicity studies showed no toxicity when licorice was administered.

Ø Acute toxicity was found to be 308 mg/kg glycyrrhetinic acid.

Ø Consists of Glycyrrhizic Acid to which stearyl alcohol has been ester bonded.

Ø Compared with Glycyrrhizic Acid, more rapidly soluble in oils such as olive oil, cottonseed oil, mineral oil, polyethylene glycol, stearyl alcohol and petroleum jelly.

Ø Highly effective in anti-itch/rash creams, around the eye creams, damaged skin treatment and moisturizers. Effective in hydrating and soothing visible skin irritation.

Ø More effective in its anti-inflammatory activities, used in cosmetic and medicine to prevent skin inflammation and rashes.

Glabridin 40%

Ø Glabridin based extract is a natural extract derived from licorice root of Glycyrrhiza inflata. It imports whitening effects through its anti-tyrosinessproperties.

Ø The whitening activity of this product is due to the anti-tyrosinessactivities caused by the Glabridin, which decreases the production of melanin. The extract containing Glabridin is highly non-polar and soluble in oil based systems. Thus, it is necessary to incorporate this product in the final formulation with the maximum absorption properties.

Ø The recommended dosages is between 0.02% to 0.04% for Glabridin 40% and 0.3% to 0.5% of Dipotassium Glycyrrhizinate.

Ø The Glabridin 40% is compatible with a wide range of personal care product ingredients and has a very good solubility under a wide pH range. It can be incorporated in oil based skin care products.
