fuge with a 6 x 500 ml rotor(batch processing), 20 l ofculture was harvested in 2hours and 55 minutes.
■ With a Contifuge stratos,only 40 minutes were neededto process the same amount.

And it all works very easilyThe fixed rotor head with inletand outlet rises above thecentrifuge lid.
Using a peristaltic pump, thesuspension is fed into the rotorbody via a rotating joint. Thesample flows through thechamber, and sediment par-ticles gather against the rotorwalls, while clarified medium isremoved.
When the suspension iscompletely processed or therotor’s intake capacity isreached, the centrifuge isstopped. The rotor is thenopened, and the pelletharvested.

Example applicationThe harvest of an Escherichiacoli culture with a straight,stick-like bacterium (1.1 – 1.5 x 2.6 – 6.0 µm) is describedbelow.
20 l of an E. coli culture in a LB medium were harvested at 15,000 rpm with a varying flowrate. The optical density wasdetermined from the residueand the percentage degree ofclarification was calculatedfrom it, i.e. the centrifugationseparation capacity was ascer-tained.
ResultThe degree of clarification,depending on the flow rate, is illustrated in the graph. An in-crease in the flow rate inexcess of 500 ml/min wasconnected to a considerabledecrease in the separation per-formance. Within 40 minutesthe entire 20 l culture washarvested with a 98% degreeof clarification.
Centrifugation parametersin generalA sample’s degree of separationdepends on the centrifugalforce and the flow rate. As with “traditional” centrifuges,acceleration is determined bythe rotor speed and increasesby the speed squared. Thecontinuous flow speed isadjusted via the peristalticpump’s power. A decrease inthe flow rate improves theseparation performance as aresult of the sample’s prolongedlength of stay in the rotor.
Application tipFluid culture media containinglarge volumes of yeast extractor other complex ingredientstend to produce large amountsof lather. In this case there wasa high production of lather dueto the bacteria’s formation ofgas. However, this did notimpair the separation perfor-mance in the Contifuge.

Easy – the rotor assemblyFor every particular rotorthere is an adaptor whichhouses the automatic rotorrecognition system. Thisavoids any possibility ofconfusion and guaranteesutmost safety as a result ofappropriate speed restric-tions. Work with the Conti-fuge stratos is similarlystraightforward, as withtraditional centrifuges.
Great – the high performance insertThis polypropylene insertimproves the rotor’ssedimentation capability,particularly for smallparticles and those with alow density, whilst reducingthe sedimentation capacity(50 ml). It has been deve-loped for the separation ofsuspensions with a low solidmaterial content. A typicalarea of application is theseparation of fine matter insuspensions for waterexaminations.
The heart – the rotorContinuous flow rotorsdeveloped by Kendro aredesigned to ensure gentlesedimentation of cells with-out lysis. The titanium rotorhas proven to be ideallysuited to fermenter harves-ting of bacterial cultures. It ischemically inert and isdistingnished by its highmaximum speed. Analuminium rotor is offered asan economical alternative. It is worth emphasising thatfor some applications – e.g.the harvest of human or ani-mal cells – lower speeds aresufficient, and these can beachieved with the aluminiumrotor. As with the titaniumrotor, it is fully autoclavable(20 mins., 121 ºC). It has ahigh quality special coating,the chemical resistance ofwhich is similar to that ofTeflon.
Smart – the integrated pumpTo maintain defined andreproducible flow rates, theuse of a peristaltic pumpwith a pump range of 100 to 900 ml x min-1 is recom-mended. Kendro offers aperistaltic pump which iscontrolled via the centrifuge. A minimum speed of 4000rpm is required to fill andoperate the rotor. Thecentrifuge is equipped withadditional electronics and asocket for the pump controlwhich stops the pump as
Mobile – the trolleyA movable centrifuge table – on which the pump, samplecontainers and a magneticstirrer can be placed –makes the entire systemmobile. It can be transportedto any location.
The trolley constitutes anopen, castor-mounted ca-sing in which sample bottlesand a magnetic stirringsystem can be installed. Twolocks on the front castorsimprove stability during ope-ration. The trolley’s reinforcedmetal construction is verysturdy and virtually free fromvibration.
