Consumer Products Study

Disinfectants, Mouthwash Antibacterial Hand Soap, Antibiotics, and Spices

MATERIALS Tape

Wax pencil Petri dish

Cotton swabs Forceps

Nutrient broth Sterile disk

Inoculating wire loop Sterile water

Bunsen burner Nutrient agar plate

All consumer products

Procedure

1. Suspend 23g of nutrient agar powder in 1L purified H2O.

2. Mix and heat solution until powder is dissolved and solution becomes an translucent yellow.

3. Autoclave at 121ºC for 15 minutes.

4. Fill as many petri dishes as possible (half full) with nutrient agar and let them solidify.

5. Dip cotton swab in sterile H2O and take samples from mouth, skin, and environment.

6. Rub the swab on nutrient agar plate.

7. Cover nutrient agar plates and label them.

8. Invert nutrient agar plates and place them into the incubator at 37ºC.

Procedure (cont’d)9. For discontinuous streaking, sterilize wire

loop by flaming until the wire turns bright orange.

10.Cool loop by lightly touching the wire to the nutrient agar.

11.Using the wire loop, take bacteria from the nutrient agar plate and streak 4 times on a new agar plate and flame the loop in between.

12.Cover, label, and tape plates shut. 13.Place, inverted, in the incubator at 37°C.

14. For the continuous streaking, use the same procedure as the discontinuous streaking, except you just streak back and forth until you cover the whole nutrient agar plate.

Procedure (cont’d)Re-suspend bacteria

1. Pour 10ml of nutrient broth into the continuously streaked plates.

2. Swab bacteria until all bacteria is suspended in liquid3. Swab bacteria into new plates.4. Dip sterile disk in disinfectants, antibiotics, spices,

mouthwash, or antibacterial hand soap and place in center of plates. Tap gently to secure.

5. Label plates and place them inverted in the incubator. 6. Let plates sit in the incubator over night.7. Measure the zone of inhibition (in millimeters) and make

your conclusions.
