Overview

Vornia Fast-Gelling Thiolated Hyaluuronic Acid is anatural glycosaminoglycan and component ofextracellularmatrix(ECM)makingitparticularlysuitedto cell culture, cosmetic and medical deviceapplications. Itmay be used in conjunctionwith ECMproteins such as collagen, laminin, or fibronectin formost 3D cell culture and tissue-engineeringapplications.

The procedure for thiolating our Hyaluronic Acidproduct was developed in collaboration with leadingscientists in University College Dublin, Ireland, inresponsetocustomerrequestsforthiolatedhyaluronicacid suitable for tissue engineering applications,especially for the production of hydrogels in theinvestigation of wound healing. Vornia’s thiolatedhyaluronic acid has >98% purity and has a very shortgelation time withmulti-vinyl cross-linkers (<30min).All batches of thiol-modified hyaluronic acid areprovidedwithfullcertificateofanalysis.

OurthiolatedHyaluronicAcidproductisprovidednon-sterile, therefore, for aseptic applications we

recommend passing reconstituted Hyaluronic Acidthrougha0.22µmfilterpriortouse.

Instructions for Use ThiolatedHyaluronicAcidwillcrosslinkinthepresenceof oxygen and therefore should be re-constituted indegassed, deionized water (sterile, if using in cellculture). Our vial caps are designed to allow for theinsertionofaneedlethroughtherubberinsertforthepurposes of re-constitution without opening the vialandexposingtheproducttooxygenintheair.

I. Torelievepressure insidethevial,piercetherubberinsertwithafine-tippedneedleand,usingasyringe,extracta4mLvolumeoftheinertgaswhichblanketstheproduct.

II. Resuspend in 1 mL of degassed 1X PBS*, using aneedleandsyringe,toachieveastockconcentrationof 10mg/mL (observe aseptic technique if intendedforcellculture).

III. Todissolve,werecommendunopenedvialstobesetto shake at 600 rpm, or greater, (or vortex) for <20min (pH needs to be adjusted to neutral after fullydissolution).

IV. ForgelationofVorniaThiol-modifiedHyaluronicAcid,werecommendtheadditionofcrosslinkingmultivinylpolymer (e.g. PEGDA) in 1:4 ratio and incubation at37°C. Gelation occurs between 10 min to 30 min,dependingon%w/vofcrosslinker.

*If re-constituting indegassedwater,neutralisationofsolutionpHwillbenecessary.(1mLofH2O+8µL1MNaOHpervial)
