Principles of Operationof
Constant Cell Disruption Systems are based on the use of high pressure to force a sample through a small fixed orifice at high speed under controlled, contained and repeatable conditions.
As the high pressure piston descends, sample is introduced into the high pressure cylinder.
1.
The high pressure piston then accelerates the sample through the fixed orifice at high velocity. The controlled acceleration causes the cells to rupture.
2.
When the jet containing disrupted cells hits the target, it is spread radially across the cooled heat exchange surface. Product then flows from the disruption chamber for collection. The hydraulics are recharged and the cycle continues.
3.
Reproducible ResultsConstant Cell Disruption Systems give reproducible and superior results because control of the disruption pressure is achieved by the hydraulic control system.
The disruption pressure cycle is shown adjacent. This has been created by analysing and recording the hydraulic pressure in the operating cylinder, which is related to the disruption pressure by the ratio of the piston areas.
Contained ProcessThe disruption head, high pressure cylinder and cooling system have been designed with containment and safety in mind so that the volume of high pressure product is kept to minimum at all times. The integrity of the containment system has been tested by the Biosafety Testing Station at the Centre for Applied Microbiology Research, UK. “ ...a high level of bio-containment.” “No evidence of emission from the equipment in any test.”
