1 INSTRUMENT DESCRIPTION

1.1 General

This transducer is designed to perform the bronchospasm test on the guinea pig and is particularly suitable for connecting to Ugo Basile recorders: 7050 Unirecord, 7070 Gem-ini, 17400 DataCapsule.

It enables the research worker to evaluate the spasm inducing effect of drugs having a very wide range of action, not necessarily intended to act on respiratory dynamics.

The Bronchospasm Transducer 7020 is also a useful research tool for screening sub-stances inducing the opposite effect, both those causing active bronchodilation in basal conditions and those which antagonize test drugs such as histamine, bradykinin, etc.

It is basically an air-flow meter provided with a water input valve with an adjustable pres-sure threshold, connected to the forced respiration piping via a by-pass.

The whole device is a compact unit made entirely of Perspex (see picture) mounted on a base along with its own power supply and controls.

1.2 Experimental Layout

The experimental layout follows the well known Konzett Roessler arrangement (Konzett H. Se Roessler R. - Arch. Exp. Path. Pharma-kol., 195, 171, 1940, see 6.0 BIBLIOGRA-PHY) with the anaesthetized guinea pig breathing via a reciprocating pump, according to Starling's mode of operation, see Fig. 3.

Any reciprocating or bellow-type Starling's pump is suitable, provided it is capable of de-livering up to 10 mi per stroke, at frequencies ranging from 30 to 70 strokes per minute. The Ugo Basile 7025 RODENT VENTILATOR is particularly suitable.

Neither tubing gauge nor by-pass location are critical.

1.3 Air-flow Meter

The air-flow meter is based on the hot-wire anemometer principle. An electrically-heated small diameter platinum alloy wire PW is located in front of the nozzle N (see Figure 4

Any air flow will reduce the temperature of the wire thereby decreasing its resistance in proportion to the flow (see also 3.6.2).

As the wire element is one arm of a Wheatstone bridge, variations in resistance generate a proportional voltage output which feeds the amplifier/recording unit.

The System operates as follows:-

Pressure pulses in the by-pass connection brought about by the operation of the pump cause the water column C in glass tube T to oscillate. This turn leads the water level in the valve body to rise and fall and consequently air is expelled and drawn in through the nozzle N.

Intake air follows the pattern indicated in Fig. 3 (page 4) while expelled air hits PW and is metered in the process.

The recorder therefore monitors respiration dynamics by providing a tracing appearing as a succession of "spikes".

When aerodynamic alterations take place in the bronchi, inspiration pressure pulses act-ing on the water column C change their intensity, thus air is expelled at different rates, producing a trace of narrower or wider amplitude.

In particular,

1.3.1 When bronchodilators are administered, pressure pulses are reduced to be-low normal breathing values, as the bronchi exert less aerodynamic resis-tance to forced inspiration

The tracing will decrease in amplitude to a marked degree, enabling the ac-tion of bronchodilators to be assessed, see Figure 1.

1.3.2 When bronchoconstrictors are administered and bronchospasm occurs, a quantity of air delivered by the pump and not accepted by the lungs, finds its way out via the by-pass connection: it displaces the water column from tube T and bubbles through the water, thus producing a tracing of much wider ampli-tude (see Figure 2) than that caused by oscillations of the water column.

The trace can be adjusted to an appropriate width and the instrument set to maximize contrast between normal breathing and Bronchospasm (or bronchodilation) conditions, as described in 1.4.2.

1.4 Controls

1.4.1 A BRIDGE BALANCING knob is provided, located on the front panel of the transducer case.

The bridge must be balanced to provide no signal at zero air-flow (7020 ON & clamp A tight), in particular if the amplifier/recording system is not provided with a zero suppression circuit.

1.4.2 An ATTENUATOR switch enables the output signal to be attenuated by a factor 30:1 when the switch is ON.

Owing to the high sensitivity of the air-flow meter, the amplitude of the normal breathing signal is so wide that it is convenient in most cases (see also 3.4.1 & 3.4.2) to reduce it (ATTENTUATOR ON).

The two steel rods provided supply a simple and producible means of reducing the am-plitude of the monitoring signal in two progressive steps, by inserting one of them into glass tube T.

The cross section of the water column C is in fact reduced by the rod and hence the wa-ter displaced by the pressure pulses (see also paragraph 1.3) from column C is reduced as well. See sketch below:

In this way the water level in the valve body rises and falls to a lesser degree: this in turn decreases the quantity of air expelled through N.

When bronchospasm takes place, a definite volume of air {see also 1.3.2) displaces wa-ter. The decreased cross section of "C" has no marked effect on the quantity of expelled air but increases its speed, thus generating a larger anemometer signal (see 3.6.2) .

To sum up, the use of the rod has the effect of reducing monitoring signal while maintain-ing bronchospasm signal amplitude, thus maximizing their ratio, see Figure 2.

2 INSTALLATION

2.1 Unpacking & Preliminary Check

Check the contents of the shipment for completeness, packing list to hand, and visually inspect the instrument as soon you take it out of the packaging. Use the Check List sup-plied.

Take out the packaging material with due care: you are handling glass!

2.2 Notes on the Instruction Manual

2.3 Before Applying Power

Take a look at the Power Module, on the back wall of the control box, which encom-passes the inlet connector of the mains cord, the mains switch and the fuse holder/voltage selector.

2.3.1 Mains Switch

This two-pole toggle switch, which complies with international safety standards, provides a visual cue, meaning:-

• OFF when pressed to the “O” side • ON when pressed to the “I” side

2.3.2 Fuse Holder & Voltage Selector

The fuse holder comprises two fuses, one on the live, and the other on the neutral. For operation at 220-230 Volts, we recommend 400mA timed fuses (type T400). The fuse holder also embodies the Voltage Selector. Make sure that the flag indicates the correct voltage (i.e., the voltage of your mains).

2.3.3 Mains Cord

3 OPERATION

3.1 Aerodynamic Compatibility

There are on the market CONSTANT PRESSURE Ventilators which may be useful in some physio/pharmacological investigations but are definitely non compatible with 7020.

The Bronchospasm Test requires a Constant volume Ventilator, i.e., a Starling's Pump. The Ugo Basile 7025 Rodent Ventilator is particularly suitable.

In fact suppose to connect a Constant pressure ventilator to our system (see 1.2). When the bronchi change their aerodynamic resistance to the forced inspiration air flow, be-cause of the Constant pressure, the quantity of air entering the lungs will change accord-ingly.

Pressure however will remain Constant in the air piping {used ventilator is a Constant pressure device by definition!) and so no air will be delivered by the by-pass connection; not even pressure pulses will act on the water column (see 1.3).

3.2 Connection to Recorder

The Bronchospasm Transducer is normally supplied with a cable/connector (ECTA 1332M107MZ) designed for Ugo Basile Recorders.

If the customer wishes to make use of other recording apparatus, special connections can be provided to order, together with ad-vice on possible wiring alterations to make the Bronchospasm Transducer compatible with the customer's existing recording equipment.

At this point the customer who wishes to connect the 7020 to his amplifier/ recording Sys-tem may ask which is the output of the transducer proper, in terms of mV per mi of air, to ascertain whether his system possesses adequate sensitivity. For details of transducer output, see 3.6 Calibration.

In any case, an amplifier/recording system featuring a sensitivity of at least 0.5 mV per cm is adequate to supply an excellent recording.

When 7020 is operated in conjunction with UNIRECORD 7050 or "plug-in" preamplifiers 7080 or 7082 of GEMINI 7070, look for appropriate advice in their instruction manuals.

3.3 Preparing the Experiment

3.3.1 Fill the water valve with distilled water (see also 4.1) via the sliding lateral res-ervoir up to the level! of, say, 10 cm generally recommended in the literature.

3.3.2 Connect the output cable to the recording equipment and adjust its sensitivity knob to an intermediate value.

Check that the air-flow head leads are firmly secured in their binding posts (see also 4.2).

Switch on the recorder.

3.3.3 Let both recorder and 7020 become thermally stabilized.

Adjust the bridge balancing knob (of the 7020!) until the base line does not shift when sensitivity of the recorder is adjusted.

When this happens, it means that 7020 (see also 1.4.1) supplies no signal at zero airflow.

3.3.4 Branch the "by-pass" connection to respiration piping as indicated in Fig. 3. Keep clamp A tight.

Once the anaesthetized animal is cannulated and the pump is in operation at convenient frequency and volume settings, release the clamp A.

Adjust the water level, to cause the water column in-side the glass tube to pul-sate up and down, so as to let a minute air bubble to escape from the bottom of the tube. You will usually find 7/8 cm of water the ideal physiological condi-tion.

The recording equipment will now produce a tracing.

If the bridge balancing knob of 7020 is inadvertently moved during an experi-ment, the bridge may be rebalanced with the pump in operation and the animal linked to the system, provided that the by-pass connection A is clamped.

3.4 Optimization of Tracing Amplitude

The instrument may be set up in two different ways to achieve an optimum tracing ampli-tude for evaluating drugs having a wide range of differing effects.

3.4.1 In experiments aimed at evaluating bronchospastic drugs, use the larger of the two rods referred to in 1.4 to decrease the monitoring signal amplitude.

Clamp the air piping momentarily upstream of the tracheal-al cannula (clamp B) thereby diverting all pumped air to the transducer in order to check the trac-ing amplitude in conditions of simulated maximum bronchospasm.

3.4.2 When the effect of the drug is unpredictable, i.e. when testing drugs for possi-ble respiratory side effects, it is advisable to set up the instrument to provide a normal respiratory recording of intermediate amplitude.

Use the smaller of the two rods and adjust the controls accordingly.

In this way, administration of a bronchospastic drug will be reflected as an in-crease in amplitude and a bronchodilator as a decrease, see Figure 5.

3.5 Sensitivity

The sensitivity of the instrument in comparison! with convention al Konzett-Roessler ap-paratus is illustrated in the table below:

3.6 Calibration

3.6.1 In most commercially available Starling's pumps, e.g., the Ugo Basile 7025 RODENT VENTILATOR, there is a wheel (or knob) for adjusting the pump out-put with an indicator showing the volume of air delivered in ml.

By clamping the piping in B, all the air delivered by the pump is diverted to the transducer. In this condition, the spike height gives a graphic display of the volume set on the pump, providing a simple means of calibration.

In principle, an arbitrary scale should be adequate for evaluating air flow differ-ences. However, when two or more 7020s are operated in conjunction, calibra-tion be-comes indispensable to make the results comparable.

Because of that, all delivered 7020s are accurately calibrated. In case a recali-bration is considered convenient, for instance after having serviced the instru-ment, replaced an electronic component, etc., operate as follows:-

Keep water valve level on exactly 10 cm No rods in the glass tube T - attenuator ON Set pump at 5ml tidal volume, 70 strokes per minute Check that the air is bubbling out Adjust the internal trimmer pot. R5 (see drwg 7020/22) until you monitor

0.75 mV (22 mV - attenuator OFF) peaks between pins Ib & 5b on the out-put connector.

3.6.2 The hot-wire anemometer operates in such a way that it is sensitive to both the volume and speed of air. If the frequency of the pump is altered, the instrument should be recalibrated as the absolute values will change.

Normally there is of course no need to alter the frequency, once a satisfactory value has been determined.

70/80 strokes per minute is generally recommended in the literature.

4 MAINTENANCE & SERVICING

While any service of the instrument is to be carried out by Ugo Basile personnel or by qualified personnel, authorized by UGO BASILE organization, this section of the instruc-tion manuals describes normal maintenance procedures which can be carried out at the customer’s facilities.

UNPLUG THE MAINS CORD BEFORE CARRYING OUT ANY MAINTENANCE JOB!

4.1 Few Simple Rules

The instrument has no moving part and requires practically no maintenance.

Always use distilled water to fill the valve, in order to avoid calcium deposits.

Do not use organic solvents for cleaning purposes since they are liable to impair the Per-spex surface. Cotton wool dipped in a soapy solution will clean the instrument satisfacto-rily. Ideally use ICI Perspex polish.

Protect the instrument from dust when not in use by the plastic cover provided.

4.2 Basic Checks

Check periodically that the air-flow head terminals are securely clamped in their binding posts; the power supply is protected against accidental short circuits, so there is no prob-lem when fiddling with this connection even if 7020 is ON.

If no signal or an erratic signal appears when air bubbles through the water (incidentally, you can test the transducer by gently blowing some air via the by-pass tube without the presence of the pump/animal combination), first check the above mentioned air-flow head terminals.

Then ensure that the connection to the recorder is properly made, that its sensitivity (in case recorders of other makes are used) is adequate (see 3.2), that the recorder controls are correctly set.

Check the continuity of the air-flow meter wire element by removing the leads from the binding posts and connecting them to a conventional "tester" set on OHM.

With same tester on D.C., check that there is a voltage of approx. 0.5 Volts on the bind-ing posts.

The electronic unit is highly reliable; it is assembled on a com-pact circuit board. In the remote event of a malfunction, this can be replaced en bloc.

The board is made accessible by removing the front panel which is kept in place by four self-threading screws.

The faulty one can be sent to factory for servicing or locally repaired if qualified personnel and suitable components are available.

The air-flow head can be replaced as well by simply removing the two screws which fas-ten it to the valve body.

4.3 Long Inactivity
