BD Aerosol Management OptionThe BD aerosol management option (AMO) is a device that promotes the containment of aerosols by evacuating the sort collection chamber in the BD FACSAria ow cytometer. The option uses an attached vacuum source to rapidly evacuate aerosolized particles through an ultra-low penetrating air lter during routine sorting or analysis. Option ComponentsThe BD aerosol management option (AMO) includes the following: an evacuator to generate negative pressure an ultra-low penetrating air (ULPA) lter to trap particles, with attached tubing that connects the evacuator to the instrument an air lter for the sort collection chamber door

The AMO also requires the splash shield in the BD FACSAria accessory kit.

B CAUTION

The BD aerosol management option does not eliminate the health risks of working with biohazardous material and must be used in conjunction with good laboratory practice. The BD aerosol management option is For Research Use Only. It is not for use in diagnostic or therapeutic procedures.

EvacuatorThe evacuator holds the ULPA lter and attached tubing. Air ow is controlled using pushbuttons within a membrane panel on the front of the unit. The evacuator sits on castor wheels for easy maneuverability. It can be moved using the handle attached to the unit. See Figure 1 on page 7.

ULPA FilterThe ULPA lter used in the BD aerosol management option captures and retains 99.9999% of all particles down to and including particles 0.01 microns in size. For particles of this size, only 1 out of 1,000,000 passes through the ULPA lter (on average).

Starting Up the EvacuatorNOTICE Before starting up the evacuator, make sure that the ULPA lter is completely seated against the bottom of the evacuator lter well, and that the tubing is securely attached to the lter and instrument manifold.

B WARNING

Any instrument surface that comes in contact with biological specimens can transmit potentially fatal disease. Use universal precautions when handling instrument hardware. Wear suitable protective clothing and gloves.

1 Install the splash shield or the tube holder below the aspirator drawer(Figure 3 on page 9). The splash shield is required for sorting into a multiwell plate or onto a slide. NOTICE If you are using the tube holder, make sure an O-ring is installed in the groove on top of the tube holder. The O-ring minimizes the chance of aerosols escaping. To install the splash shield, close the sort block door and open the sort collection chamber door, if needed. (The sort block door must be closed in order to open the collection chamber door.)

Remove the tube holder, if one is installed, and tilt the splash shield to insert it into the slotted ttings below the sort aspirator drawer. (See Figure 3.) Push the splash shield all the way in.

2 Ensure that an air lter is installed in the sort collection chamber door.The lter traps airborne dust that could clog the ULPA lter. Note that the lter should be changed on a monthly basis. See Replacing the Air Filter on page 18.

3 Close the sort collection chamber door.NOTICE The sort collection chamber door must be closed for the evacuator to generate negative pressure in the chamber.

4 Switch on the main power on the back of the evacuator (Figure 4).main power

5 Press the POWER button on the membrane panel of the evacuator.

6 Press the up or down arrow button to set the suction control rate to 20%.Each time either arrow button is pressed, the suction will increase or decrease by 10%. When two lights are lit on the suction control indicator, the actual air ow is the value between the two illuminated percentages. NOTICE Do not set the suction control rate above 20%. Higher rates could affect the stability of the side streams.

7 Verify that the lter ow gauge reads less than 2.4 inches of H2O (Figure 5on page 11). For a new lter, the gauge should read 1.11.4 inches of H2O. As the lter is used, the reading will increase. If the gauge reads 2.4 inches of H2O or greater, replace the lter. See Replacing the ULPA Filter on page 14.
