    NIJ Forensic Science R&D Reports for ASCLD CrimeLab Minute Vol 10These research reports have been submitted by theNational Institute of Justice (NIJ) especially for theirrelevance to crime laboratory activities. ASCLD hasnot reviewed nor does it necessarily endorse thefindings of this research. Report: NIJ’s DNA Analysis, Capacity Enhancementand other Forensic ActivitiesThis report summarizes NIJ’s approach to improvingforensic science includes long term strategicadvances and necessary immediate relief to addressthe challenges of satisfying increasing demand forDNA analysis. From 2008 to 2014, the turnaroundtime for analysis has remained nearly constant, whiletesting capacity increased by over 60 percent.Average cost per case has decreased from $1,279 in2007 to $607 in 2014. And since 2009, uploads ofDNA profiles to the Combined DNA Index System(CODIS) have increased 143 percent. The need forthis assistance has never been greater, throughcapacity building, technical assistance, research,development, evaluation and dissemination, NIJ isworking to address the challenges experienced byforensic laboratories.

The podcast covers every forensicscience discipline. Recent episodes discussed labmanagement, human factors, 3D optical topography,subjective probability, DNA mixture interpretationsand economics and crime labs. The Evidence for Very Small ParticlesNIJ recently published an article in Forensicsdiscussing the work of Stoney Forensic in developingan investigatory technique called “very small particleanalysis.” This approach involves the carefulexamination of dust on an object to trace it back to itsoriginal location, or the comparison of two or moreobjects to determine if they have, at some point, beenin the same place. The researchers have successfullyused this analysis to support U.S. governmentagencies and local and international law enforcementinvestigations, for example to trace the origins ofillegal shipments of elephant tusks. Developing Reliable Methods for MicrobialFingerprinting of SoilsSoil evidence has the potential of being a valuabletool for linking a suspect, victim, or item to a crimescene. Currently, however, only class characteristicsof soil are considered in traditional analysis. This NIJ-supported research project examined the utility of soilbacterial profiling via next-generation sequencing ofthe 16S rRNA gene to identify a soil’s origin. Theresearchers successfully differentiated soil samplesfrom diverse and similar habitats back to their locationof origin 100 percent of time in multidimensionalspace and 87.5 percent of time through supervisedclassification. Time and space within a habitat did notaffect bacterial profiles enough to hinder location-of-origin assignment. Soil collected from evidentiaryitems remained clustered nearest its location of originwith 100 percent accuracy, even after a full year orlong-term storage. The success in tracing soils backto a location of origin demonstrates the potential ofnext-generation sequencing of bacteria, in conjunctionwith a combination of robust statistical techniques, forthe individualization of forensic soil samples.     sought to minimize human contextual bias indecisions about whether torn duct tape found at acrime scene matches a duct-tape roll found in asuspect’s possession by combining digital imageanalysis and an objective, quantitative algorithm inassessing the likelihood of a match. These NIJ-supported researchers performed edge detection andmorphological smoothing operations on high-resolution images of torn duct tape edges to extractthe torn-edge coordinates. In 97 percent of examinedtears, the true match had the lowest observed sum ofsquare residuals, with false positive rates rangingfrom 0.5 percent for some types of hand-torn ducttape to 62 percent for scissors-cut duct tape. Thiswork provides a starting point for quantitativeassessment of the likelihood of physical end matchingof duct tape without human contextual bias. Non-Medical Use of Prescription Drugs: Policy, LawEnforcement, and Diversion TacticsThe goal of this University of Central Florida studywas to determine whether Florida law HB 7095 andrelated legislation reduced the number of pain clinicsabusively dispensing opioid prescriptions in the State,as well as to assess the logistics of administering thelaw. The findings presented in this NIJ-supportedreport indicate that the Florida effort against rampant“pill mills” was largely successful, which is consistentwith research that has documented reductions inopioid-related deaths in Florida. Further, the findingsshow the key role of interagency cooperation in thesuccess of the legislation, which in conjunction withFlorida’s legislation, should be considered a model forfuture efforts in reducing easy access to prescriptionopioids. Differential Sampling of Footwear to SeparateEvidentiary Particles from Background NoiseThis NIJ-supported project by Stoney Forensic testedthe ability to separate particle signals on the footwearsoles using differential analysis of loosely held,moderately held, and strongly held particle fractions.Prior research has used a generalized sampling ofparticles on footwear soles from both contact andrecessed areas shows the retention of particles fromearlier contacts. The current study determined thatalthough particles on the contact surfaces of footwearwere removed and replaced, particles on the morerecessed areas of the sole were not. Threeenvironmental exposure sites were chosen to havedifferent characteristic particle types (soil minerals)and shoes of two types (work boots and tennis shoes)were tested. Some shoes were exposed to only oneenvironment, and others were exposed to all three inone of six sequences. Sampling methods weredeveloped to separate particles from the contactsurface of the shoe based on how tightly they wereheld to the sole. Laser Ablation Inductively Coupled Plasma MassSpectrometry (LA-ICP-MS) and Laser InducedBreakdown Spectroscopy (LIBS) Analyses of Paper,Inks, and SoilsFlorida International University researchers sought todemonstrate the utility of an analytical chemistry toolthat is relatively new to forensic laboratories, LaserInduced Breakdown Spectroscopy (LIBS), inconducting elemental analysis of ink, paper, and soilevidence quickly and with little or no samplepreparation. The commercial LIBS instrumentalresults were compared to previously optimized in-house LIBS systems and also to the alternativeforensic tools, which offer good analyticalperformance but suffer from either very high costsand significant complexity (in the case of LS-ICP-MS)or analytical limitations in the form of samplerequirement and relatively high detection limits (in thecase of mXRF and SEM-EDS). Although less maturethan LA-ICP-MS, LIBS also shares the benefitsassociated with laser ablation methods, with theadded advantage of improved speed, versatility, easeof operation, affordability, and portability. This NIJ-supported project shows that both LA-ICP-MS andLIBS are useful analytical tools for the analysis ofpaper, inks, soil, and cotton.     Three Dimensional Imaging and Novel ComparisonAlgorithms for Firearm ForensicsTopMatch technology for structural 3D imaging andcomparison of cartridge casings shows excellent andcontinually improving match accuracy; however, inorder to fully establish the base credibility of theplatform, it was necessary to establish best-scammingpractices and to demonstrate that the method meetsthe quality-control criteria of other forensicinstruments. This NIJ-supported project by CadreResearch Labs aimed to enhance TopMatch byestablishing best practices and conducting a set ofmethodology studies. The project complements theprevious studies on accuracy and evidence stabilityby addressing the remaining issues of precision,repeatability, reproducibility, contamination risk, andperformance checks. In addition, researchers studiedthe effects of cleaning protocols, focus variation, andambient lighting variation. Measuring the Frequency Occurrence of Handwritingand Hand-Printing CharacteristicsOne of the basic axioms of handwriting comparison isthat no two writers use the exact same set ofhandwriting characteristics. The database created byNIJ-supported researchers and the resultingfrequency occurrence proportions provide the forensicand judicial communities with some empirical datawith which to assess this axiom. The specificapproach in collecting samples that match the U.S.demographics provides unique insight andsubstantive data on the quantitative relationshipbetween the presence/absence of readily identifiablefeatures and key demographic factors as noted byHuber and Headrick (age, gender, ethnicity,education, location of second/third grade schooling,and handedness) while controlling for other factors,such as temporal state and geographic locations. Theoriginal set of specimens collected from a widespectrum of participants provided an initial largecollection that was eventually pared down to achievewhat is considered a representative sample from theentire target population. Gunshot Residue in a Non-Firearm DetaineePopulationStudies have shown varying amounts of gunshotresidue (GSR) in the police environment, with mostbeing on surfaces within facilities occupied andoperated by law enforcement. This NIJ-supportedstudy by researchers at the Harris County Institute ofForensic Sciences examined whether GSRcontamination could be found on the hands ofdetainees at the Harris County Jail (Texas) who werenot charged with any offenses related to thedischarge of a weapon. Of the 175 samples obtainedand tested, not one characteristic GSR particle wasfound. Coupling the results of this study with one inwhich no GSR particles were on 100 personsanswering bench warrants in Bexar County (Texas)gives an average of less than one GSR particle in 275hand samples. Considering that a different study ofrandom surfaces within the Chicago PoliceDepartment (n=201) found a total of 56 GSR particles,the current study concludes that GSR particles arenot readily transferred to surfaces not in closeproximity to a recently fired gun. Comparison of Microspectrophotometry andFluorescence Excitation-Emission Matrix (EEM)Spectroscopy for Non-Destructive Fiber Examination.When spectral information is used in the study of fiberevidence, variations within a fiber source lead to therecommendation that multiple spectra be collectedfrom each fiber to properly characterize the sample.Although this methodology is sufficient for comparisonof profiles with obvious differences, the chemometricmethods used in this NIJ-supported study fromresearchers at the University of Central Florida showhigher accuracies than the visual method commonlyused in forensic laboratories. Current practices inforensic labs that involve fluorescence microscopy donot take advantage of the information content thatexists in the spectral signature of textile fibers. The

A physicistat the National Bureau of Standards, nowknown as the National Institute of Standardsand Technology (NIST), Souder played animportant role in the early days of forensicscience. He helped send countlessmurderers, bootleggers, gangsters andthieves to prison, and he kept such a lowprofile partly out of concern for his and hisfamily’s safety. Perhaps as a result, he wasnot long remembered for his forensic work,and his influence on the developing field offorensic science was not as great as it mighthave been.

Accounting for the variance of the EEM spectraat different regions along the length of the fiberprovides a useful training set that can be used as thebasis for principal component cluster analysis. Development of a Mobile, Automated Tool MarkCharacterization/Comparison SystemThis NIJ-supported project developed a portableprototype instrument that provides forensic examinerswith the ability to characterize a tool-marked surface,compare the data from that surface to data filesobtained from any other surface, and assess thelikelihood that the marks on the two surfaces weremade by the same tool. To date, the system has beensuccessfully tested on data sets consisting of fullystriated marks created from 50 sequentiallymanufactured screwdrivers, quasi- striated markingsproduced by 50 sequentially manufactured shear-cutpliers, and impression marks produced by 50sequentially manufactured cold chisels. The Interpretation of Patterned Injuries in MedicolegalDeath InvestigationIn this report, NIJ-supported researchers describe theresults of their survey evaluating the effect of imageprocessing and image quality on the ability of forensicpathologists to accurately interpret images ofpatterned injury of the skin. The study consisted ofthree surveys: the first, a collection of “classic” imagesthat most pathologists would diagnose with highconsensus (baseline survey); the second, degradedimages with lesser resolution poorer composition todetermine how degradation affected diagnosticconsensus; and the third, images treated with variousenhancement techniques to determine whether anybenefit was gained. Surprisingly, the first surveyproduced a median of only 74 percent consensus. Ananalysis of the second and third surveysdemonstrated the importance of context and history inforensic pathologic diagnosis. When provided withhistory, consensus rose to approximately 98 percentper question (median value) for the matching subsetof the first survey. Prediction of Drug Interactions with Methadone,Buprenorphine and OxycodoneThe impact of drug interactions spreads beyond thefield of forensic toxicology to encompass generalmedicine, pharmacy and other areas involved in druguse and dispensing. These can all directly andindirectly impact the criminal justice systems, as manyin these professions are called upon as consultantsand expert witnesses for cases. Information onpotential drug interactions with methadone,buprenorphine, and oxycodone can benefit thesefields. A NIJ-supported project by researchers at theUniversity of Utah hypothesized that the inhibition ofthe metabolism of opioids may contribute to their toxiceffects and set out to measure the in vitro inhibition ofmetabolism of three opioids: methadone,buprenorphine, and oxycodone. This report outlinesthe progress made optimizing incubation conditions,establishing positive controls and screening inhibitorypotential of the three drug classes. Evaluation of a Novel Fluorescent Dye to Detect Ano-genital Injury in Women of ColorNIJ-supported researchers at the University ofVirginia tested fluorescent dyes to detect sexualassault injuries on different skin colors andtones. Trials were conducted to identify non-toxicsuitable dyes that can be easily visualized usingequipment already in common use among forensicexaminers and will not interfere with subsequentforensic DNA testing. Findings suggest thatfluorescein is both safe and feasible to use indetecting genital injury across all skin tones.However, additional research on alternative dyesacross all skin tones is recommended. Development of an Analytical System for Comparisonand Identification of Fiber DyesIn order to overcome challenges to generalized fiberdye characterizations, this NIJ-supported research

    explored a significant range of dyestuffs that spannumerous major dye application classes, chemistries,chromophores, and fiber types, using techniques andmethodologies currently available in most forensiclaboratories. Numerous aspects of dye analysis, fromextraction through micro analytical characterization,classification, and identification were examined for aset of 300 commercially prevalent dyestuffs. Theresults show that dye identification can be conductedusing techniques available in many labs. It isanticipated that this research will provide a basis fromwhich laboratories can expand their approach from ageneralized comparison of color between questionedand known materials to a specific identification of thecolorants present in a sample. This has the potentialto increase the significance of results in theprosecution or defense of a case and improve thevalue of investigative leads in cases for which acomparison sample does not exist by constraining oridentifying specific dyes. Trace Metal Analysis by Laser Ablation-InductivelyCoupled Plasma-Mass Spectrometry and X-Ray K-Edge DensitometryThis NIJ-supported dissertation by an Iowa StateUniversity researcher describes studies ondetermining trace elements in samples with forensicimportance, including trace analysis in lipstick, copperwire, fly larvae and porcine skin tissue for gunshotresidue and bullet residue in bones, as well as, soilfrom decomposed pig carcasses. Pre-Feedback Eyewitness Statements: ProposedSafeguard against Feedback Effects on Evaluationsof Eyewitness TestimonyThis NIJ-supported study from an Iowa StateUniversity researcher tested pre-feedback statementsas a safeguard for protecting against, and correctingfor, the effects of confirming feedback on evaluationsof eyewitness testimony. Results suggest thatconfirming feedback did not appear to have anyinfluence on witnesses in the no pre-feedbackstatements condition. Additionally, confirmingfeedback did not impair evaluators’ abilities todiscriminate between accurate and mistakeneyewitnesses in the conditions in which witnessesprovided pre-feedback statements. Witnesses’ pre-feedback statements to evaluators did not improveevaluators’ abilities to discriminate between accurateand mistaken eyewitnesses who received feedback.Finally, evaluators tended to judge witnesses whogave no pre-feedback statements more favorably thanthey judged witnesses who gave pre-feedbackstatements.
