Results: Six samples met technical criteria for successful FACSanalysis. Activation marker CD11b was significantly increasedin both CD4 + and CD8 + T cells after exposure to ‘‘severeintolerance’’ antigens. In CD4 + T cells: 3.5% of untreated cellswere CD11b + , 2.9% of cells exposed to ‘‘no reaction’’ foodswere CD11b + , while 36.2% of cells exposed to ‘‘severe intol-erance’’ foods were CD11b + . In CD8 + T cells: 6.6% of un-treated cells were CD11b + , 5.1% of cells exposed to ‘‘noreaction’’ foods were CD11b + , and 42.3% of cells exposed to‘‘severe intolerance’’ foods were CD11b + . These relation-ships were statistically significant at a = 0.05. Other activationmarker associations did not demonstrate any significant differ-ences.Conclusion: ‘‘Severe intolerance’’ on the ALCAT test is asso-ciated with an upregulation of CD11b on CD4 + and CD8 + Tcells. This study provided detailed analysis of well-characterizedcell populations, but was limited to a single time point of anal-ysis. Purpose: Current therapy for lung cancer includes chemother-apy, radiotherapy and surgery that are effective but associatedwith significant morbidity. Many compounds isolated fromplants against cancer are found to have toxic behavior as com-pare to crude extract of same plant from which compounds areisolated. It forces to find a potential plant that can be used incrude form for treatment of cancer without showing any toxicityto normal body cells.Methods: Fingerprinting of MeGj was done by HPTLC forstandardization. Cytotoxicity of MeGj was examined in lym-phocytes (5FU as positive control) and A549 cells by dye ex-clusion assay using trypan blue and MTT assay at 24 and 48 hrs.The effect of MeGj on intracellular level of oxidative stress inA549 cells was determined at 24 and 48 hrs by analyzing theactivity of SOD and Catalase enzyme. MeGj in apoptosis wasevaluated by studying apoptotic parameters such as alteration incell morphology under phase contrast microscopy, change inmorphology at nuclear level by staining cell with 4’, 6-diami-dino-2-phenylindole (DAPI), measuring DNA damage by cometassay and determining Caspase 3 activity at exposure period of48 hrs. The anti-metastatic property was studied by performingwound healing assay.Results: MeGj did not show any cytotoxicity on lymphocytecells at three different concentrations (500, 1000, 1500 lg/ml).MeGj showed dose and duration-dependent reduction in cellviability and cell growth and an increase in SOD activity ascompared to control and inactivation of Catalase activity. At48 hrs of exposure MeGj showed cell rounding, cell shrinkage,cytoplasmic membrane irregularity, chromatin fragmentationand condensation, DNA damage, high Caspase 3 activity andshowed inhibition of cell migration at highest dose (1500 lg/ml).
Conclusion: It is concluded that MeGj possesses anti-prolifer-ative and anti-metastatic activity on A549 and nontoxic tolymphocytes, making it a promising agent for further evaluation.Contact: Meghna Adhvaryu, [email protected]

Purpose: Genetic factor and diet intake are important for dia-betes development. The present pilot study was to perform theeffects of ShenQi Decoction (SQD), a Chinese herbal medicine,on blood glucose, insulin resistance and lipid profile in geneti-cally diabetic KK-Ay mice.Methods: Male mice were divided into two groups to receivednormal diet (ND) or high fat diet (HFD) for 14 weeks under freeaccess of diet. At the 9th to 14th week, the HFD group was thensubjected into two groups and administrated with present(HFD + SQD) or absent of SQD for 6 weeks. The body weight,the concentration of fasting glucose, insulin, total cholesterol,triglyceride, high-density lipoprotein (HDL), and the homeo-stasis model assessment of insulin resistance (HOMA-IR) weredetermine before and at the end of the 8th, 11th, and 14th weekof the experiment.Results: As a result, the fasting blood glucose, total cholesterol,HDL, triglyceride, body weight, and HOMA-IR of HFD werehigher than that in ND. However, insulin of HFD was less thanthat in ND. Furthermore, total cholesterol, HDL, triglyceride,and HOMA-IR of HFD + SQD were lower than that in HFD after3 weeks of SQD administration. Moreover, the concentration offasting glucose of HFD + SQD was lower than that in HFD after6 weeks of SQD treatment.Conclusion: Although HFD increased the blood glucose andlipid levels, which lead the diabetes onset earlier in geneticallydiabetic KK-Ay mice, SQD might inhibited and delayed theHFD induced-diabetic severity and development.Contact: Yu-Min Cho, [email protected]
