Flow Cytometry and HIV in Resource Scarce Countries

● CD4+ T cells play important role in adaptive immune system● HIV attacks and kills CD4+ T cells● Crucial to monitor CD4 count over HIV/AIDS disease lifetime● CD4+ Count determines what medications a patient should be on● Over 22 million of the predicted 33 million living with HIV are in Sub-Saharan

Africa● Flow Cytometry can be used to count CD4+ T cell count

Microfluidic channel fabrication

A patterned SU-8 template on glass

PMMA exposed to acetonitrile for 2 min then had an SU-8 template pressed into the surface for 10 min, which provided appropriately imprinted channels and a suitable surface for bonding.

Procedure of receiving fluorescent signal

● The microfluidic chip is inserted in the devices.

● The view area is illuminated from the side by inexpensive

LEDs.

● A fluorescent microscope movie of the flow cells can be

acquired using the cell-phone camera unit by CMOS.

● The digital frames of these fluorescent movies are rapidly

processed to determine the count of labeled WBC, which

can be used to estimate the density of the target fluorescent

objects within the sample solution.

Data Analysis of flow cytometry image ● Linear Discriminant Analysis

○ Features: Cell Morphology (Size, Shape, etc.), Protein Content, DNA content, etc. ○ Use of hyperplane to distinguish classes (whether disease is present or not, etc.)

Page 15: A Novel, Portable Device to Count CD4+ T Cells using Flow ... · Clinical Uses Brown, Michael, and Carl Wittwer. “Flow Cytometry: Principles and Clinical Applications in Hematology

Limitations● Different diseases need different set ups.● Downsizing typically reduces complexity of testing● Error categorizing each cell to get accurate count
