Enzyme-linked immunosorbent assay (ELISA) is still the most widely used method for large scale testing of plant viruses.

DSMZ offers about 250 high quality serological reagents and positive controls for routine ELISA detection of the most important viruses infecting agricultural and horticultural crops. The available diagnostic products are listed in alphabetic order in this catalogue.

ELISA sets

• contain IgG and conjugate (IgG-AP) for DAS-ELISA • contain IgG, Mab and RAM-AP for TAS-ELISA • are optimized and provided for a test volume of 200µl

and are supplied as 100 test units (smallest unit) up to >1000 test units.

ELISA controls

Plant virus materials for use as positive/negative controls in ELISA consist of freeze-dried homogenates of virus infected or healthy plants and are to be reconstituted in buffer prior to use. In some cases, desiccated leaf material may be provided as ELISA controls.

• Dissolve controls consisting of freeze dried plant homogenate in 2ml sample extraction

buffer. • Controls consisting of freeze dried leaf material have to be homogenized in sample

extraction buffer (1:50 weight/volume) prior to use.

Principle

Lateral flow tests, also known as lateral flow immunochromatographic assays, are devices intended to be used as “on-site” test for the rapid detection of viruses or other pathogens without the need for laboratory equipment. Lateral flow tests are provided for some viruses (where indicated) as complete field detection kits for 25 tests; including extraction bags, buffer and disposable pipettes.

Procedure and how to interprete the results

Samples are homogenized in provided extraction bags and a few drops of the plant homogenate are added to the sample pad (“S”).

If a sample is positive, two separate lines will appear (“T” and “C”, test and control line). The test line will be red to purple in color just as the control line although the intensity may vary.

If the sample is negative the test line will not appear. Only one single line (control “C”) will appear.

The control line assures that the test is working properly. If the control line does not appear, the test is invalid.

Positive controls in ELISA may vary in OD values according to plant virus species and strains, and plant tissue used. We strongly recommend Greiner medium binding ELISA plates, other plates could lead to increased background reactions and/or lower overall sensitivity. If observed, relevant cross-reactions with related viruses are listed.

Import License

There may be restrictions or prohibitions of import of certain antibodies, conjugates, positive controls, virus inocula and other reagents into certain countries. It is the responsibility of the customer to check such necessities and/or to receive a permit to handle our products, and where necessary to provide the DSMZ with import documentation to accompany the product.

Shipment

All items are shipped, unless otherwise advised by the customer, via FedEx or TNT at ambient temperature. Please note that we will not have any proof of delivery for standard non courier service air mail supplies, that they are not tracked and might take several weeks. Shipments by courier services (fast, tracked) are charged to the customer. Please provide clear shipping instructions when ordering.
