What is Cryopreservation?

• Cryopreservation is to preserve and store the viable biological samples by the use of antifreeze (cryoprotectant) solutions and cooling to very low temperature for long-term storage

CONSERVATION TO SUPPORT BIODIVERSITY

• It prevents narrowing of biodiversity• It maintains food security• It prevents genetic drift of gametes, embryos• Safeguards genetic integrity of valuable strains• Offers viable alternative to maintaining active breeding

colony• Ease of transportation of genetic stock • Decrease of disease transmission • Speedy recovery of stocks following disasters

TO CONSERVE BIODIVERSITY

Cryopreservation can be used for   Conservation of plant germplasm  Vegetatively propagated species (root and tubers, ornamental, fruit

trees) Recalcitrant seed species (Howea, coconut, coffee)  Conservation of tissue with specific characteristics  Medicinal and alcohol producing cell lines Genetically transformed tissues Conservation of plant pathogens (fungi, nematodes)

Germplasm Cryopreservation

• The sperm, eggs, tissues, cultured cell lines, DNA and serum samples could be frozen and store in cryogene bank.

• Preserving the genomes of threatened and endangered species.

In Plants

• Variety of plant material can be used, including cells in tissue culture, pollen, seeds or parts of seeds, embryos, tissues from the early stages of development of mosses and ferns, buds, twigs, and meristematic (growing point) tissue.

Semen Preservation

• Semen is generally cryopreserved with “slow cooling” methods.

• Optimal cooling rates for freezing semen are mostly found between 10 and 100 °C/min.

Page 14: 10.Cryopreservation for Biodiversity Conservation

Oocyte Preservation

• Human oocyte cryopreservation(egg freezing) is a novel

technology in which a woman’s eggs (oocytes) are extracted, frozen and

stored.

Page 15: 10.Cryopreservation for Biodiversity Conservation

Embryo Cryopreservation• Embryo is frozen to

extremely cold temperature, under such extreme conditions virtually all biological process cease, include the process of cell death.

• Under these specialized conditions embryos remain dormant

DNA BANKING• The DNA bank is an efficient, simple and long-

term method used in conserving genetic resource for biodiversity.

• Compared to traditional seed or field gene banks, DNA banks lessen the risk of exposing genetic information in natural surroundings.

• It only requires small sample size for storage and keeps the stable nature of DNA in cold storage.
