Fluorescent Flow CytometryYields Optimized Productivity
The Sysmex XT Automated Hematology Systemsutilize the power of fluorescent flow cytometry andhydrodynamic focusing technologies. Using a unique,diode laser bench, Sysmex fluorescent flow cytometryprovides the sensitivity needed for measuring anddifferentiating cell types in whole blood and bodyfluid samples. Fluorescent technology andhydrodynamic focusing enable the analyzers toconsistently classify normal WBC, RBC, and PLTpopulations from abnormal populations, therebydecreasing the number of manual interventions.

Designed to be reliable and efficient,the Sysmex XT-2000i and XT-1800i (XT) offers mediumvolume labs an automated hematology system that cantruly meet and exceed their expectations. The XT streamlinesyour workflow by providing testing for up to 80 samplesper hour, enabling rapid turnaround time.

Clinically Relevant Information
High Quality Hemoglobin(HGB) Analysis
The XT utilizes the cyanide-freereagent, Sodium Laurel Sulfate(SLS). The end product is a coloredcompound that is measuredspectrophotometrically. Since hemoglobindeterminations are performed from adilution and in its own separate chamber,there is no interference from high WBCcounts, lipemia or abnormal proteins.
Direct Hematocrit (HCT)Measurement
The cumulative pulse heights of all theRBC counts yield the HCT. This is basedon the principle that the pulse height(voltage change) produced by cellspassing through the aperture isproportional to cell volume.
RBCs and PLTs
RBCs and PLTs are counted in adedicated channel using Impedance orDirect Current (DC) detection methodcombined with hydrodynamic focusingtechnology. Challenges to cell countingsuch as coincidence or recirculationare circumvented and automaticdiscriminators separate the twocell populations.
Even with samples at extremelylow or unusually high concentrations,the XT-Series analyze RBCs and PLTswith uncompromised precisionand accuracy.

WBC, A Clear Differentiation
The combination of side scatter(cell complexity), forward scatter(cell size) and fluorescence (DNA &RNA concentration) of nucleated cellsprovides a concise and precise imageof each detected peripheral blood cell.
This 3-dimensional blood cell analysisprovides unique accuracy and precision.Fluorescence labeling of peripheralblood cells is a milestone for theroutine leukocyte differential.
Fluorescent technology enablesthe XT to reliably differentiate normalWBC populations from abnormal WBCpopulations. The sensitivity of theunique application of fluorescent flowcytometry gives the lab a high level ofconfidence in reporting accurate WBCdifferentials, even on critical patientsamples when the WBC count is low.

Fluorescent Optical Platelet
Fluorescent Optical Platelet*
The XT-2000i offers both aFluorescent Optical Platelet (PLT-O)in addition to the traditional impedancePLT count. Flagging associated withatypical or abnormal platelets, due toincreased size or fragmentation isminimized by use of the opticalplatelet count. Accuracy in reportingis supported by the availability of bothtechnologies.
Fluorescent Optical Platelet is acomplementary, reportable parameterto the impedance platelet count.
The PLT-O parameter provides:
• Improved accuracy on lowplatelet counts
• Accurate counts when interferencesare present, thus reducing manualintervention
• Automated judgment for reportingPLT-O or impedance PLT throughinstrument settings, eliminatingtech-to-tech decision variability

Body Fluids
The XT analyzers include measurementfor body fluids. These analyzers providereportable WBC and RBC countsfor all common body fluid samples(CSF, synovial and serous).
The analyzers apply provenimpedance and fluorescent flowcytometry technology ensuringan accurate body fluid count froma single sample analysis.
The XT Body Fluid Count provides:

With the use of fluorescent technology, the reticulocyte count assures:
• Accurate reticulocyte % and #• Improved immature reticulocyte fraction (IRF) information for earlier diagnosis
and treatment by clinicians• Elimination of common interferences from Howell-Jolly bodies,
Pappenheimer bodies and immature reticulocytes to avoid manual counts
In addition, the Reticulocyte Hemoglobin (RET-He), is a parameter measuredin the reticulocyte channel which is used to measure the incorporation of ironinto erythrocyte hemoglobin.
The RET-He parameter supports:
• Rapid, direct analysis of an earlier stage of RBC development forprompt clinical follow-up
• Assessment of anemia. RET-He is an established parameter used inKDOQI (Kidney Disease Outcomes Quality Initiative) guidelines for assessingthe initial iron status of patients
• Accuracy and sensitivity in measurement of red cell production that aidsthe clinician in effective monitoring of costly drug protocols for cell stimulation

Reticulocyte Analysis*
