Nucleic Acid Electrophoresis and Blotting
Agarose and AcrylamideUsed in routine electrophoresis of nucleotides for DNA identification, Southerns, Northerns, or DNA recovery. Sigma offers a wide range of agarose products depending on the desired result, such as low melting point, high resolution, or pulse-field gels. Acrylamide is used to perform Polyacrylamide Gel Electrophoresis (PAGE) gels for protein or high-resolution nucleic acid electrophoresis.
Precast GelsTo speed up your electrophoresis workflow Sigma offers a variety of precast gels for nucleic acid electrophoresis. The precast gels have a variety of concentrations and wells available to run gels specific to the needs of each experiment.
Denhardt's SolutionMixture of blocking agents used in hybridization protocols such as Northerns and Southerns to prevent non-specific binding of probes.
DNA Ladders and MarkersSigma has an assortment of nucleic acid markers for use in agarose or PAGE electrophoresis. The range and number of bands in each ladder varies depending on the size of nucleotide for identification. Ladders are also optimized for use at specific gel concentrations.
Dyes and Loading BuffersDyes, such as Xylene Cyanol and Bromophenol Blue are added to nucleic acid samples to track the migration of samples as agarose or PAGE electrophoresis occurs. Sigma also provides Gel Loading Buffers that contain components such as sucrose to facilitate sample loading.
Nucleic Acid Gel StainsEthidium bromide is an intercalation agent used to identify nucleotides after conducting electrophoresis. When placed inside a UV light box ethidium bromide fluoresces enabling identification of samples.
Running BuffersBuffers such as Tris-Borate-EDTA (TBE) are most commonly used for PAGE gels while Tris-Acetate-EDTA (TAE) is used for agarose electrophoresis. Sigma has a broad portfolio of Running Buffers depending on the specific running conditions. Bionic Buffer enables gels to be run at higher voltages resulting in runtimes that are 2–3 times faster than TBE or TAE.
ssDNAUsed to prevent non-specific binding of probes to membranes when Northern or Southern blots are performed.
EquipmentSigma-Aldrich has an assortment of equipment used in electrophoresis and blotting, of which gel boxes, power supplies, and pipette tips are available. Barrier pipette tips are used to prevent sample contamination.
